Στην παρούσα διατριβή παρουσιάζεται η σύνθεση δυο τύπων πολυπεπτιδίων του PLys-b-(PHis-co-PBLG)-b-PLys-b-(PHis-co-PBLG)-b-PLys και του PLys-b-(PHis-co-PBLG)-b-PEO-b-(PHis-co-PBLG)-b-PLys, τα οποία σχηματίζουν υδρογέλες με την προσθήκη νερού με μοναδικές ιδιότητες. Οι υδρογέλες αυτές αποτελούν ένα κατάλληλο υλικό για στοχευμένη και διαρκή χορήγηση του αντικαρκινικού φαρμάκου gemcitabine σε στερεούς παγκρεατικούς καρκινικούς ιστούς. Το υλικό προς χορήγηση αποτελείται από το πολυπεπτίδιο, νερό και γεμσιταβίνη, τα οποία σχηματίζουν ισχυρή υδρογέλη και απελευθερώνουν τη γεμσιταβίνη επιλεκτικά σε εντοπισμένους παγκρεατικούς καρκινικούς ιστούς, εκμεταλλευόμενοι τις τοπικές συνθήκες της καρκινικής φλεγμονής. Οι υδρογέλες είναι ενέσιμες, σχηματίζονται in situ, έχουν ικανότητα αυτοίασης και δεν εμφανίζουν κυτταροτοξικότητα έως 400mg/Kg μετά από ίn νίνο δοκιμές σε πειραματόζωα. Η σύνθεση ανταποκρίνεται στην υψηλότερη θερμοκρασία και στο χαμηλότερο pΗ των καρκινικών σε σχέση με τα υγιή κύτταρα διαμέσου της αλλαγής της δευτεροταγούς δομής. Αυτές οι ιδιότητες επιτρέπουν την τοποθέτηση της υδρογέλης κοντά στον ιστό του καρκίνου με την απλούστερη δυνατή μέθοδο χορήγησης, δηλ. με ένεση με λεπτή βελόνα. Επιπλέον, παραμένει μια ισχυρή υδρογέλη που παρεμποδίζει την απελευθέρωση της γεμσιταβίνης στον υγιή ιστό, κατευθύνοντας έτσι την παροχή του φαρμάκου στον καρκινικό ιστό. Οι υδρογέλες αυτές αποκρίνονται σε ένζυμα παρόντα στους καρκινικούς ιστούς από τα οποία βιοαποικοδομούνται. Εξετάστηκαν οι ιξωδοελαστικές και δομικές τους ιδιότητες που σχετίζονται με τις λειτουργίες τους. Βρέθηκε ότι η ανάπτυξη των όγκων στα ποντίκια μπορεί να καθυστερήσει κατά τη χορήγηση 40% λιγότερης ποσότητας gemcitabine μέσω της υδρογέλης, συγκριτικά με μια κανονική και προτεινόμενη δόση ελεύθερης gemcitabine σε πειραματόζωα με καρκίνο στο πάγκρεας.
