Numerous dynamic and complicated processes characterize development from the oocyte to the embryo. However, given the importance of functional transcriptome profiles, long non-coding RNAs, single-nucleotide polymorphisms, and alternative splicing during embryonic development, the effect that these features have on the blastomeres of 2-, 4-, 8-, 16-cell, and morula stages of development have not been studied. Here, we conducted a scRNA-seq survey of cells from sheep from the oocyte to the blastocyst developmental stages. We then carried out experiments to identify and functionally analyze the transcriptome profiles, long non-coding RNAs, single-nucleotide polymorphisms (SNPs), and alternative splicing (AS). We founded that between the oocyte and zygote groups significantly down-regulated genes and the second-largest change in gene expression occurred between the 8- and 16-cell stages. We used various methods to construct a profile to characterize cellular and molecular features and systematically analyze the related GO and KEGG profile of cells of all stages from the oocyte to the blastocyst. This large-scale, single-cell atlas provides key cellular information and will likely assist clinical studies in improving preimplantation genetic diagnosis.
Balanced mechanical properties are always the goal of high-performance general plastics for engineering purposes. In order to develop uniformly dispersed graphene oxide/poly(L-lactic acid) nanocomposites with balanced mechanical properties, a poly(ethylene glycol) (PEG) grafted graphene oxide was introduced into poly(L-lactic acid) (PLLA) matrix. The PEG grafted graphene oxide (GEG) was confirmed by the results of fourier transform infrared (FTIR) spectra and thermal gravimetric analysis (TGA). Then the mechanical properties of the nanohybrids were measured. The results showed that in contrast to the aggregation of GO, GEG could uniformly disperse in PLLA matrix. An intriguing phenomenon is that thanks to the strong interfacial interaction between GEG and PLLA, it exhibits a substantial enhancement of the elongation at break (EB) as well as a simultaneous improvement of the tensile strength (TS), while the addition of GO decreases the EB of PLLA matrix. This work provides a potential industrialized technique for high-performance PLLA nanocomposites
Air-conditioning and ventilation system which control temperature and humidity was designed in this paper. Its aim was to ensure the production process and meet the comfort requirements of workers. Considering that web press equipment load account for large proportion of the total load, we separated out the area of web press and the waste heat was discharged by ventilation system. We adopted Stratified air conditioning on the remaining area because the whole workshop is large space. By means of calculation, energy saving rate of stratified air conditioning system is 25.6% and the refrigerating capacity is reduced by this way of design. Energy-saving effect is remarkable. So it’s a good method to energy conservation.
