SummaryFruit ripening is characterized by dramatic changes in gene expression, enzymatic activities and metabolism. Although the process of ripening has been studied extensively, we still lack valuable information on how the numerous metabolic pathways are regulated and co-ordinated. In this paper we describe the characterization of FaMYB1, a ripening regulated strawberry gene member of the MYB family of transcription factors. Flowers of transgenic tobacco lines overexpressing FaMYB1 showed a severe reduction in pigmentation. A reduction in the level of cyanidin 3-rutinoside (an anthocyanin) and of quercetin-glycosides (¯avonols) was observed. Expression of late¯avonoid biosynthesis genes and their enzyme activities were aversely affected by FaMYB1 overexpression. Two-hybrid assays in yeast showed that FaMYB1 could interact with other known anthocyanin regulators, but it does not act as a transcriptional activator. Interestingly, the C-terminus of FaMYB1 contains the motif pdLNL D / E Lxi G / S . This motif is contained in a region recently proposed to be involved in the repression of transcription by AtMYB4, an Arabidopsis MYB protein. Our results suggest that FaMYB1 may play a key role in regulating the biosynthesis of anthocyanins and¯avonols in strawberry. It may act to repress transcription in order to balance the levels of anthocyanin pigments produced at the latter stages of strawberry fruit maturation, and/or to regulate metabolite levels in various branches of the¯avonoid biosynthetic pathway.
The blends of flavor compounds produced by fruits serve as biological perfumes used to attract living creatures, including humans. They include hundreds of metabolites and vary in their characteristic fruit flavor composition. The molecular mechanisms by which fruit flavor and aroma compounds are gained and lost during evolution and domestication are largely unknown. Here, we report on processes that may have been responsible for the evolution of diversity in strawberry (Fragaria spp) fruit flavor components. Whereas the terpenoid profile of cultivated strawberry species is dominated by the monoterpene linalool and the sesquiterpene nerolidol, fruit of wild strawberry species emit mainly olefinic monoterpenes and myrtenyl acetate, which are not found in the cultivated species. We used cDNA microarray analysis to identify the F. ananassa Nerolidol Synthase1 (FaNES1) gene in cultivated strawberry and showed that the recombinant FaNES1 enzyme produced in Escherichia coli cells is capable of generating both linalool and nerolidol when supplied with geranyl diphosphate (GPP) or farnesyl diphosphate (FPP), respectively. Characterization of additional genes that are very similar to FaNES1 from both the wild and cultivated strawberry species (FaNES2 and F. vesca NES1) showed that only FaNES1 is exclusively present and highly expressed in the fruit of cultivated (octaploid) varieties. It encodes a protein truncated at its N terminus. Green fluorescent protein localization experiments suggest that a change in subcellular localization led to the FaNES1 enzyme encountering both GPP and FPP, allowing it to produce linalool and nerolidol. Conversely, an insertional mutation affected the expression of a terpene synthase gene that differs from that in the cultivated species (termed F. ananassa Pinene Synthase). It encodes an enzyme capable of catalyzing the biosynthesis of the typical wild species monoterpenes, such as a-pinene and b-myrcene, and caused the loss of these compounds in the cultivated strawberries. The loss of a-pinene also further influenced the fruit flavor profile because it was no longer available as a substrate for the production of the downstream compounds myrtenol and myrtenyl acetate. This phenomenon was demonstrated by cloning and characterizing a cytochrome P450 gene (Pinene Hydroxylase) that encodes the enzyme catalyzing the C10 hydroxylation of a-pinene to myrtenol. The findings shed light on the molecular evolutionary mechanisms resulting in different flavor profiles that are eventually selected for in domesticated species.
Fruit flavor is a result of a complex mixture of numerous compounds. The formation of these compounds is closely correlated with the metabolic changes occurring during fruit maturation. Here, we describe the use of DNA microarrays and appropriate statistical analyses to dissect a complex developmental process. In doing so, we have identified a novel strawberry alcohol acyltransferase ( SAAT ) gene that plays a crucial role in flavor biogenesis in ripening fruit. Volatile esters are quantitatively and qualitatively the most important compounds providing fruity odors. Biochemical evidence for involvement of the SAAT gene in formation of fruity esters is provided by characterizing the recombinant protein expressed in Escherichia coli . The SAAT enzyme showed maximum activity with aliphatic medium-chain alcohols, whose corresponding esters are major components of strawberry volatiles. The enzyme was capable of utilizing short-and medium-chain, branched, and aromatic acyl-CoA molecules as cosubstrates. The results suggest that the formation of volatile esters in fruit is subject to the availability of acyl-CoA molecules and alcohol substrates and is dictated by the temporal expression pattern of the SAAT gene(s) and substrate specificity of the SAAT enzyme(s).
The Australian Community Climate and Earth System Simulator coupled model (ACCESS-CM) has been developed at the Centre for Australian Weather and Climate Research (CAWCR), a partnership between CSIRO 1 and the Bureau of Meteorology. It is built by coupling the UK Met Office atmospheric unified model (UM), and other sub-models as required, to the ACCESS ocean model, which consists of the NOAA/GFDL 2 ocean model MOM4p1 and the LANL 3 sea-ice model CICE4.1, under the CERFACS 4 OASIS3.2-5 coupling framework. The primary goal of the ACCESS-CM development is to provide the Australian climate community with a new generation fully coupled climate model for climate research, and to participate in phase five of the Coupled Model Inter-comparison Project (CMIP5). This paper describes the ACCESS-CM framework and components, and presents the control climates from two versions of the ACCESS-CM, ACCESS1.0 and AC-CESS1.3, together with some fields from the 20 th century historical experiments, as part of model evaluation. While sharing the same ocean sea-ice model (except different setups for a few parameters), ACCESS1.0 and ACCESS1.3 differ from each other in their atmospheric and land surface components: the former is configured with the UK Met Office HadGEM2 (r1.1) atmospheric physics and the Met Office Surface Exchange Scheme land surface model version 2, and the latter with atmospheric physics similar to the UK Met Office Global Atmosphere 1.0 including modifications performed at CAWCR and the CSIRO Community Atmosphere Biosphere Land Exchange land surface model version 1.8. The global average annual mean surface air temperature across the 500-year preindustrial control integrations show a warming drift of 0.35 °C in ACCESS1.0 and 0.04 °C in AC-CESS1.3. The overall skills of ACCESS-CM in simulating a set of key climatic fields both globally and over Australia significantly surpass those from the preceding CSIRO Mk3.5 model delivered to the previous coupled model inter-comparison. However, ACCESS-CM, like other CMIP5 models, has deficiencies in various aspects, and these are also discussed.
The primary purpose of this study is to assess the performance of 1D solar radiative transfer codes that are used currently both for research and in weather and climate models. Emphasis is on interpretation and handling of unresolved clouds. Answers are sought to the following questions: (i) How well do 1D solar codes interpret and handle columns of information pertaining to partly cloudy atmospheres? (ii) Regardless of the adequacy of their assumptions about unresolved clouds, do 1D solar codes perform as intended? One clear-sky and two plane-parallel, homogeneous (PPH) overcast cloud cases serve to elucidate 1D model differences due to varying treatments of gaseous transmittances, cloud optical properties, and basic radiative transfer. The remaining four cases involve 3D distributions of cloud water and water vapor as simulated by cloud-resolving models. Results for 25 1D codes, which included two line-by-line (LBL) models (clear and overcast only) and four 3D Monte Carlo (MC) photon transport algorithms, were submitted by 22 groups. Benchmark, domain-averaged irradiance profiles were computed by the MC codes. For the clear and overcast cases, all MC estimates of top-of-atmosphere albedo, atmospheric absorptance, and surface absorptance agree with one of the LBL codes to within Ϯ2%. Most 1D codes underestimate atmospheric absorptance by typically 15-25 W m Ϫ2 at overhead sun for the standard tropical atmosphere regardless of clouds. Depending on assumptions about unresolved clouds, the 1D codes were partitioned into four genres: (i) horizontal variability, (ii) exact overlap of PPH clouds, (iii) maximum/random overlap of PPH clouds, and (iv) random overlap of PPH clouds. A single MC code was used to establish conditional benchmarks applicable to each genre, and all MC codes were used to establish the full 3D benchmarks. There is a tendency for 1D codes to cluster near their respective conditional benchmarks, though intragenre variances typically exceed those for the clear and overcast cases. The majority of 1D codes fall into the extreme category of maximum/random overlap of PPH clouds and thus generally disagree with full 3D benchmark values. Given the fairly limited scope of these tests and the inability of any one code to perform extremely well for all cases begs the question that a paradigm shift is due for modeling 1D solar fluxes for cloudy atmospheres.

SUMMARKRadiative parametrizations for both ice and water clouds are developed in terms of liquid/ice water content, based on Mie scattering theory. For ice crystals the application of Mie theory is guided by the hexagonal-crystal/equivalent-spheres comparison of Takano and Liou. These parametrizations are extensively tested against measurements from aircraft and are shown to perform satisfactorily, although corrections for unobserved small crystals and the effect of crystal shape are large and not currently well defined. The parametrizations are then used to investigate the effect of mixed-phase clouds on radiative transfer. It is found that, because the radiative properties of ice crystals and liquid droplets are significantly different, the radiative properties of mixed-phase clouds cannot be simulated successfully if the ice in clouds is converted into liquid water. Both the albedo and the rate of change of albedo with ice fraction are significantly dependent on the method by which the phases are mixed; these factors may be of especial importance in climate-sensitivity experiments that incorporate mixed-phase clouds. The presence of ice in clouds below the cirrus level is often ignored in climate-model and radiation-budget studies. The calculations presented here indicate that this neglect may lead to a serious bias in cloud albedo for a given path of condensed water.
