BackgroundInorganic polyphosphate (poly P) plays an important role in stress tolerance and virulence in many bacteria. PPK1 is the principal enzyme involved in poly P synthesis, while PPK2 uses poly P to generate GTP, a signaling molecule that serves as an alternative energy source and a precursor for various physiological processes. Campylobacter jejuni, an important cause of foodborne gastroenteritis in humans, possesses homologs of both ppk1 and ppk2. ppk1 has been previously shown to impact the pathobiology of C. jejuni.Methodology/Principal FindingsHere, we demonstrate for the first time that the deletion of ppk2 in C. jejuni resulted in a significant decrease in poly P-dependent GTP synthesis, while displaying an increased intracellular ATP:GTP ratio. The Δppk2 mutant exhibited a significant survival defect under osmotic, nutrient, aerobic, and antimicrobial stresses and displayed an enhanced ability to form static biofilms. However, the Δppk2 mutant was not defective in poly P and ppGpp synthesis suggesting that PPK2-mediated stress tolerance is not ppGpp-mediated. Importantly, the Δppk2 mutant was significantly attenuated in invasion and intracellular survival within human intestinal epithelial cells as well as in chicken colonization.Conclusions/SignificanceTaken together, we have highlighted the role of PPK2 as a novel pathogenicity determinant that is critical for C. jejuni survival, adaptation, and persistence in the host environments. PPK2 is absent in humans and animals; therefore, can serve as a novel target for therapeutic intervention of C. jejuni infections.

The food control and regulatory system in China is beset by several challenges. While firms have to reduce their costs in pursuit of benefits, customers are increasingly focusing on safety and quality of food products. Although the Chinese government has developed more stringent regulatory measures, food safety incidents still occur, including abuse of food additives, adulterated products as well as contamination by pathogenic microorganisms, pesticides, veterinary drug residues, and heavy metals, and use of substandard materials. A national food safety strategy has been proposed to assure food safety from “farm to table.” This paper begins with the analysis of current food regulatory systems and then discusses cogovernance of food safety management in China. We explore the practice in the city of Shenzhen where government intervention has strengthened food control, thereby creating an opportunity to form a coregulatory system. The review highlights that the current food safety regulatory system of multi‐agency structure can inevitably lead to insufficient incentives for business entities. Due to asymmetric information, lack of regulatory resources, and consumer advocacy, coregulation has been developed and is increasingly being promoted as an important instrument of food regulation.

Composition and ensilage performance of giant reed harvested in August, October, November, and December, were evaluated and compared. Generally, late-harvested giant reed had higher dry matter content, lower nitrogen content, and higher water soluble carbohydrates (WSC) content than early-harvested giant reed. During 90days of ensilage, giant reed harvested in October, November, and December showed dry matter losses of about 1%, while giant reed harvested in August showed a higher dry matter loss of about 8%. During the ensilage process, more lactic acid was produced in late-harvested giant reed than in early-harvested giant reed. Late-harvested giant reed had a higher lignin content and lower enzymatic digestibility than early-harvested giant reed. However, enzymatic digestibility of all the giant reed biomass was improved by the 90-day ensilage process, reaching levels of 43-46%. In summary, ensilage could be used for storing giant reed biomass harvested at different times and for improving its digestibility.

Artificial Intelligence (AI) technologies have been powerful solutions used to improve food yield, quality, and nutrition, increase safety and traceability while decreasing resource consumption, and eliminate food waste. Compared with several qualitative reviews on AI in food safety, we conducted an in-depth quantitative and systematic review based on the Core Collection database of WoS (Web of Science). To discover the historical trajectory and identify future trends, we analysed the literature concerning AI technologies in food safety from 2012 to 2022 by CiteSpace. In this review, we used bibliometric methods to describe the development of AI in food safety, including performance analysis, science mapping, and network analysis by CiteSpace. Among the 1855 selected articles, China and the United States contributed the most literature, and the Chinese Academy of Sciences released the largest number of relevant articles. Among all the journals in this field, PLoS ONE and Computers and Electronics in Agriculture ranked first and second in terms of annual publications and co-citation frequency. The present character, hot spots, and future research trends of AI technologies in food safety research were determined. Furthermore, based on our analyses, we provide researchers, practitioners, and policymakers with the big picture of research on AI in food safety across the whole process, from precision agriculture to precision nutrition, through 28 enlightening articles.

This study aimed to investigate the forage yield and quality and extract factors affecting them and provide scientific support for ecological protection of alpine grassland and sustainable development of animal husbandry in the Qinghai-Tibetan Plateau. Samples were collected in late August (peak of the growing season) 2014 in two mountain transects (Laji mountain, 36°21′ N, 101°27′ E, 3389-3876 m a.s.l. and Heitu mountain, 34°22′ N, 100°30′ E, 4121-4268 m a.s.l.) of Sanjiangyuan. Forage biomass and nutrients were measured by the national standard method, and influencing factors were extracted by RDA. Results showed that the forage yield decreased significantly with the increasing altitude, no matter in the mountain > 4000 m a.s.l. or < 4000 m a.s.l. Forage yield was more affected by environmental factors in low altitude areas, but less in high-altitude areas. Forage nutrients were mainly affected by altitude and soil organic carbon (SOC) (p < 0.01) on the mountain < 4000 m a.s.l., while on the mountain > 4000 m a.s.l., fiber and lignin were mainly affected by soil pH and SOC and crude protein (CP) by soil pH (p < 0.05) and total phosphorus (TP). Overall, with the increasing altitude, forage CP and fat (CF) increased, while neutral detergent fiber and acid detergent fiber decreased, implying that warming may lead to the decline of forage quality and then affect the safety of animal husbandry. Moreover, forage CP and CF were also affected by soil inorganic nitrogen and TP rather than SOC and soil total nitrogen.

BackgroundThe invasion associated marker (iam) has been detected in the majority of invasive Campylobacter jejuni retrieved from humans. Furthermore, the detection of iam in C. jejuni isolated from two important hosts, humans and chickens, suggested a role for this marker in C. jejuni's colonization of multiple hosts. However, no data exist regarding the occurrence of this marker in C. jejuni isolated from non-poultry food-animals such as cattle, an increasingly important source for human infections. Since little is known about the genetics associated with C. jejuni's capability for colonizing physiologically disparate hosts, we investigated the occurrence of the iam in C. jejuni isolated from cattle and assessed the potential of iam-containing cattle and human isolates for chicken colonization and human cell invasion.ResultsSimultaneous RAPD typing and iam-specific PCR analysis of 129 C. jejuni isolated from 1171 cattle fecal samples showed that 8 (6.2%) of the isolates were iam-positive, while 7 (54%) of human-associated isolates were iam-positive. The iam sequences were mostly heterogeneous and occurred in diverse genetic backgrounds. All iam-positive isolates were motile and possessed important genes (cadF, ciaB, cdtB) associated with adhesion and virulence. Although certain iam-containing isolates invaded and survived in INT-407 cells in high numbers and successfully colonized live chickens, there was no clear association between the occurrence, allelic sequence, and expression levels of the iam and the aforementioned phenotypes.ConclusionsWe show that the prevalence of iam in cattle C. jejuni is relatively lower as compared to isolates occurring in humans and chickens. In addition, iam was polymorphic and certain alleles occur in cattle isolates that were capable of colonizing and invading chickens and human intestinal cells, respectively. However, the iam did not appear to contribute to the cattle-associated C. jejuni's potential for invasion and intracellular survival in human intestinal cells as well as chicken colonization.
