ABSTRACT. We investigated the role of 7 single nucleotide polymorphisms in the carbonic anhydrase (CA) VI gene (rs2274328, rs17032907, rs11576766, rs2274333, rs10864376, rs3765964, and rs6680186) and the possible association between these polymorphisms and dental caries susceptibility in a Northwestern Chinese population. We collected samples from 164 high caries experience and 191 very low caries experience and conducted a case-control study according to the number of decayed, missing, and filled teeth index and genotyped the 7 polymorphisms using a 384-well plate format with the Sequenom MassARRAY platform. Individuals carrying the rs17032907 TT genotype were more likely to have an increased risk of dental caries compared with carriers of the C/C genotype in the co-dominant model, with an odds ratio (95% confidence interval) of 2.144 (1. 096-4.195). We also found that the haplotype (ACA) (rs2274328, rs17032907 and rs11576766) was associated with a low number of decayed, missing, and filled teeth index with an odds ratio (95% confidence interval) of 5987 Genetic polymorphisms and dental caries ©FUNPEC-RP www.funpecrp.com.br Genetics and Molecular Research 14 (2): 5986-5993 (2015) 0.635 (0.440-0.918). However, we found no association between dental caries susceptibility and the rs2274328, rs11576766, rs2274333, rs10864376, rs3765964, and rs6680186 polymorphisms and other haplotypes. The rs17032907 genetic variant and the haplotype (ACA) of CA VI may be associated with dental caries susceptibility.
Tumor necrosis factor-α (TNF-α), a pro-inflammatory cytokine, produces pain and hyperalgesia by activating and/or sensitizing nociceptive sensory neurons. In the present study, using whole-cell patch clamp techniques, the regulation of potassium currents by TNF-α was examined in acutely dissociated small dorsal root ganglion neurons. We found that acute application of TNF-α inhibited, in a dose-dependent manner, the non-inactivating sustained potassium current without changing the rapidly inactivating transient current or the kinetics of steady-state inactivation. The effects of TNF-α on potassium currents were similar to that of prostaglandin E2 as reported previously and also demonstrated in the current study. Furthermore, indomethacin, a potent inhibitor for both cyclooxygenase (COX) -1 and COX-2, completely blocked the effect of TNF-α on potassium currents. These results suggest that TNF-α may sensitize or activate sensory neurons by suppressing the sustained potassium current in nociceptive DRG neurons, possibly via stimulating the synthesis/ release of endogenous prostaglandins.
The main contribution of this article is the proposal of a path-following method for fixed-wing unmanned aerial vehicles. This path-following method employs the multi-loop framework that consists of an outer guidance loop and an inner control loop. The guidance loop relies on the idea of tracking a virtual target. The virtual target is assumed to travel along the defined path and its speed is explicitly specified. This guidance law guarantees the asymptotic convergence to the desired path and can anticipate the transition of the flight path in advance, which reduces the command for the inner control loop. In the inner control loop, the flight control law based on dynamic surface control is derived to overcome the 'explosion of complexity' problem in the backstepping design. The numerical simulation result illustrates the effectiveness of the proposed method.
The (99m)Tc-peptide-ZHER2:342 molecular probe is a promising tracer agent, and the results in this study provide a foundation for future development of protocols for earlier visual detection of cancer in the clinical setting.
The aim of this study was to detect the effects of different combinations of cryoprotectants with different equilibrium time on the mouse ovarian tissue during vitrification. Ovarian tissue of mice was vitrified-thawed. Mice (n = 80) were randomly assigned to treatment groups according to different vitrification solutions [I: 20% (v/v) ethylene glycol (EG) + 20% (v/v) Dimethylsulfoxde (DMSO), II: 20% (v/v) EG + 20% (v/v) PROH, III: 20% (v/v) PROH + 20% (v/v) DMSO] and different lengths of equilibrium time (a: 15 min, b: 30 min, c: 45 min). The serum levels of estradiol, the follicular density and the percentage of cells expressing Proliferating-cell nuclear antigen (PCNA) of grafts in Group IIb were the highest in all these treatment groups. In addition, the serum levels of estradiol, the follicular density and the percentage of cells expressing PCNA of grafts in Group Ib were significantly higher than those in Group Ia and Group Ic, while the serum levels of estradiol, the follicular density and the percentage of cells expressing PCNA of grafts in Group IIIb were significantly higher than those in Group IIIa and Group IIIc. In conclusion, vitrification solution [20% (v/v) EG + 20% (v/v) PROH] with equilibrium time of 30 min is optimal selection for vitrifying mouse ovarian tissue.
