Mikroalga merupakan alga kecil (ukuran 2-20 ?m) berupa tanaman talus yang memiliki klorofil sehingga mampu melakukan fotosintesis. Salah satu jenis mikroalga yang berpotensi untuk diambil minyaknya adalah Nannochloropsis sp. Nannochloropsis sp. merupakan sel berwarna kehijauan, tidak motil, dan tidak berflagela. Sebagian besar mikroalga menggunakan cahaya dan karbondioksida (CO2 ) sebagai sumber energi dan sumber karbon. Pada perairan CO2 terlarut biasanya dalam bentuk bikarbonat (HCO3 - ). Tujuan penelitian ini untuk mengetahui pengaruh peningkatan konsentrasi bikarbonat (HCO3 - ) terhadap peningkatan pertumbuhan mikroalga Nannochloropsis sp. Perlakuan yang diberikan kepada Nannochloropsis sp. adalah variasi bikarbonat, yaitu 25 ppm, 50 ppm, 100 ppm, dan 200 ppm dan dikultivasi selama 14 hari. Media pertumbuhan yang digunakan adalah Walne. Paramater yang diamati adalah pH, temperatur, Optical Density (OD), jumlah sel, biomassa dan lipid. Metode ekstraksi yang digunakana adalah maserasi dengan pelarut heksan:etanol (1:1). Hasil penelitian menunjukkan bahwa pertumbuhan mikroalga Nannochloropsis sp. berada pada kondisi optimum ketika menggunakan penambahan bikarbonat 200 ppm. Pada kondisi tersebut diperoleh jumlah sel tertinggi sebesar 22,72 x 106 sel/mL dengan laju pertumbuhan 0,0176 sel/jam dan biomassa sebesar 3,85 g/L. Kondisi pH kultur selama penelitian rata-rata berada pada pH 8 dan temperatur kultur berada pada rentang 25-28o C. Sedangkan untuk kadar lipid tertinggi berada pada konsentrasi bikarbonat 25 ppm dengan kadar lipid 20,236 % berat kering.
Abstract. Chlorella vulgaris and Nannochloropsis oculata are a highly potential microalgae to be used in pilot-scale of biodiesel synthesis. The essential content from these microalgae is the fatty acid of lipid which is the main target for the feed and biodiesel industries. One of the key factor in improving lipid microalgae are the arrangemment of nutrients in the growth medium. Research on the regulation of nutrients using bicarbonate (HCO3 -) as an additional inorganic carbon source has been done by many studies, but the yield of lipids obtained has not been much. The aim of the study was to improve the lipid yield of Chlorella vulgaris and Nannochloropsis oculata.
