Background: Assessing the effectiveness and safety of acupuncture therapy for treating patients with COVID-19 is the main purpose of this systematic review protocol. Methods: The following electronic databases will be searched from inception to May 2020: Cochrane Central Register of Controlled Trials, PubMed, Web of Science, EMBASE, China National Knowledge Infrastructure, Traditional Chinese Medicine, Chinese Biomedical Literature Database, Wan-Fang Database, and Chinese Scientific Journal Database. All published randomized controlled trials in English or Chinese related to acupuncture for COVID-19 will be included. Primary outcomes are timing of the disappearance of the main symptoms (including fever, asthenia, cough disappearance rate, and temperature recovery time), and serum cytokine levels. Secondary outcomes are timing of the disappearance of accompanying symptoms (such as myalgia, expectoration, stuffiness, runny nose, pharyngalgia, anhelation, chest distress, dyspnea, crackles, headache, nausea, vomiting, anorexia, diarrhea), negative COVID-19 results rates on two consecutive occasions (not on the same day), CT image improvement, average hospitalization time, occurrence rate of common type to severe form, clinical cure rate, and mortality. Results: The results will provide a high-quality synthesis of current evidence for researchers in this subject area. Conclusion: The conclusion of our study will provide an evidence to judge whether acupuncture is an effective intervention for patients suffered from COVID-19. Ethics and dissemination: Formal ethical approval is not necessary as the data cannot be individualized. The results of this protocol will be disseminated in a peer-reviewed journal or presented at relevant conferences. PROSPERO registration number: CRD42020183736.
Background: There is a worldwide outbreak of covid-19, and as the number of patients increases, more and more patients are recovering. Massage is used as an alternative therapy. Currently, there are no relevant articles for systematic review. Methods: We will search the randomized controlled trials related to acupuncture therapy and postoperative anorectal diseases from inception to January 2020. The following database is our focus area: the Cochrane Central Register of Controlled Trials (CENTRAL), PubMed, EMBASE, Web of Science, China National Knowledge Infrastructure, Chinese Biomedical Literature Database, and Wan-Fang Database. All published randomized controlled trials in English or Chinese related to massage for COVID-19 will be included. Primary outcome asthe influence of massage on the quality of life of convalescent patients. Secondary outcomes were accompanying symptoms (such as myalgia, expectoration, stuffiness, runny nose, pharyngalgia, anhelation, chest distress, dyspnea, crackles, headache, nausea, vomiting, anorexia, diarrhea) disappear rate, negative COVID-19 results rate on 2 consecutive occasions (not on the same day), average hospitalization time, clinical curative effect, and improved quality of life. Results: The results will provide a high-quality synthesis of current evidence for researchers in this subject area. Conclusion: The conclusion of our study will provide evidence to judge whether massage is an effective intervention on the quality of life in patients recovering. Dihydrotestosterone (DHT) is a main androgen in the human body. Previous reports have shown that DHT can affect the proliferation, apoptosis and estrogen and progesterone secretion of ovarian granulosa cells (GCs). An imbalance in DHT secretion leads to GC dysfunction and follicular development disorder.Therefore, exploring the influence of DHT on GCs is necessary. The purpose of this study was to analyze the effect of DHT on GCs through label-free quantitative proteomics (LFQP). After primary cultured rat GCs were treated with DHT (10-8 mol/L), the effect of DHT on GCs was analyzed by LFQP, and some of the differentially expressed proteins (DEPs) were verified by western blotting.A total of 6124.0 proteins were identified, of which 4496.0 were quantifiable. Compared with the control group, 28 proteins were upregulated and 10 were downregulated after DHT intervention. The subcellular localization of DEPs indicates that DHT is involved in the proliferation, migration, molding and metabolism of GCs. Gene Ontology (GO) revealed that DHT downregulated the oxygen transport capacity and oxygen-binding protein of GCs. Orthologous Groups of proteins (COG/KOG) showed that DHT had an important effect on the survival, growth and apoptosis of GCs. Kyoto Encyclopedia of Genes and Genomes (KEGG) revealed that DHT promotes metabolism, amino acid degradation, chemical carcinogenesis, platelet activation and vasoconstriction in GCs. The western blot results were consistent with the proteomics results. Mark3 and Mre11a are DEPs that were upregulated, and Fth1 and Nqo1 were downregulated, which indicated that DHT could promote the proliferation of GCs.This study comprehensively analyzes the impact of DHT on GCs through LFQP and provides clues for further research.
Purpose: Whether FSH promotes follicle growth by inhibiting the Hippo signalling pathway.METHODS: Ovaries were cultured in vitro into a control group (no intervention), an FSH group (0.3 IU/mL FSH), and a VP group (10 µg/mL vetiporfin). HE staining and follicle counts were performed at each stage after 3 hours of in vitro culture. Immunohistochemistry was performed to study the expression levels of LATS2, YAP, PLATS2, and PYAP, and their expression levels in each group were also analysed by Western blot.The number of secondary follicles was significantly increased in the FSH group, the arrangement of granulosa cells was neater, the nuclear fixation was reduced, and the number of atretic follicles was decreased in the VP group. The number of secondary follicles was significantly increased, the number of atretic follicles was reduced, and granulosa cell nuclear consolidation was reduced in the VP+FSH group. Immunohistochemistry showed that LATS2 and YAP expression levels were significantly increased and PLATS2 and PYAP expression levels were relatively decreased in the FSH group, PYAP and PLATS2 expression levels were significantly increased and YAP expression was significantly decreased in the VP group, and YAP and LATS2 expression levels were significantly increased and PYAP and PLATS2 expression levels were significantly decreased in the VP+FSH group. By Western blot, LATS2 and YAP were elevated and PYAP and PLAT2 were decreased in the FSH group, LATS2 and YAP were decreased and PYAP and PLATS were significantly elevated in the VP group, and LATS2 and YAP were elevated and PYAP and PLATS2 were decreased in the VP+FSH group.CONCLUSION: FSH promotes follicle development by inhibiting the Hippo signalling pathway.
