The purpose of this study was to determine the biochemical, histopathological and genotoxic effects of tetramethrine, which is widely used in domestic and agricultural activities as well as identify the protective effect of caffeic acid phenethyl ester (CAPE). 30 Swiss albino laboratory mice (Mus musculus) were used in the study. 10 μM/kg tetramethrine, dissolving in 10 mM DMSO (Dimethyl sulfoxide), was intraperitoneally injected to the Tetramethrine group (n = 10). To the CAPE-Tetramethrine group, 10 µM / kg -1 CAPE, dissolving in 1% ethanol, was applied in three days before the experiment and then, CAPE and 10 μM / kg tetramethrine, dissolving in 10 mM DMSO, was intraperitoneally injected. No injection was made to the control group. At the end of the experiment, the rats anaesthetized with diethyl ether were killed by cervical dislocation and their livers and femur were removed for analysis. There was no statistical difference between three groups in terms of mitotic index (MI) and micronucleus frequency (P > 0.05). Chromosomal aberration frequency showed an increase in the other two groups compared to the control group (P < 0.05). Although severe degeneration and necrosis areas were identified in the liver, CAPE decreased the severity of degeneration in the Tetramethrine group. While Tetramethrine increased the malondialdehyde (MDA) level in the liver, CAPE decreased MDA and increased the GSH level. We think that CAPE may be used for therapeutic purposes in order to provide a limited protection against the tetramethrine-related detrimental effects in humans and other living organisms.
ÖzetÇalışmada, Kars Çayı'ndan yakalanan Siraz balığı (Capoeta capoeta) ve Tatlısu kefali (Squalius cephalus) üzerine hekzavalent kromun (CrVI) (potasyum dikromat olarak) etkileri histopatolojik ve elektroforetik yöntemlerle incelendi. Balıklar çeşme suyunda 10 gün süreyle bekletilerek ortama adaptasyonları sağlandı. Daha sonra, her bir balık türü için her grupta 10'ar adet olmak üzere 3 grup oluşturuldu. I. gruptaki (kontrol) balıklar çeşme suyu içeren tankta, II. gruptaki balıklar 10 gün süreyle, III. gruptaki balıklar ise 20 gün süreyle 10 mg/L dozunda CrVI içeren tanklarda bekletildi. Bu süre sonunda, balıklardan kan ve doku örnekleri alındı, daha sonra analizler yapıldı. Histopatolojik inceleme sonucunda CrVI'a maruz kalan Capoeta capoeta ve Squalius cephalus'ların karaciğer dokularında fokal nekroz alanları, hidropik ve vakuolar dejenerasyonlar tespit edildi. Bu dejenerasyonların şiddetinin hekzavalent kroma maruz kalma süresiyle artış gösterdiği gözlemlendi. Serum proteinlerinin SDS-PAGE'inde ise 10 gün süreyle CrVI uygulamasına bağlı olarak Capoeta capoeta'nın bazı protein bantlarında hafif kalınlaşma olduğu, Squalius cephalus'un protein bantlarında ise belirgin bir değişikliğin şekillenmediği saptandı. Hekzavalent kromun 20 gün süreyle uygulandığı Capoeta capoeta'nın birçok protein bantlarında incelmede, bazı protein bantlarında ise belirgin derecede kalınlaşmalar gözlendi. Squalius cephalus'da ise bazı protein bantlarında hafif derecede kalınlaşma meydana geldiği tespit edildi. Sonuç olarak; potasyum dikromat uygulamasının zamana bağlı olarak artan derecelerde Capoeta capoeta ve Squalius cephalus karaciğer dokularında ve protein ekspresyonlarında bozulmalara neden olduğu sonucuna varıldı. Bu çalışma, Deltamethrin uygulanan Carassius gibelio türü balıkların solungaç, karaciğer ve bağırsak dokuları üzerine histopatolojik etkilerini incelenmek amacı ile yapıldı. Araştırmada 60 adet balık, her grupta 12 balık bulunan, 5 gruba ayrıldı. Bu gruplardan biri kontrol, diğer dördü deney grubu olarak oluşturuldu. Kontrol grubu dışında, deney gruplarındaki balıklar deltametrinin; 0.48, 0.64, 0.80 ve 0.96 mg/ L'lik konsantrasyonlarının etkisinde 96 saat bırakıldı. Deney sonunda sakrifiye edilen balıklardan alınan doku örneklerinin tespit ve takip işlemlerinden sonra kesitleri alındı ve bu kesitler Hematoksilen+Eozin (H+E) ile boyanarak histopatolojik değişiklikler incelendi. Deney gruplarındaki balıklarda; solungaç epitel hücrelerinde hipertrofi, epitelin lameller ayrılması ve mukus hücre hipertrofisi gözlendi. Karaciğerde kistik yapılar, vena centralislerde düzensizlikler, venlerin, hücrelerin ve diğer elemanların birbirinden ayrılamadığı görüldü. Bağırsaklarda ise lezyonlar, mukus hücre proliferasyonu, ölü villus epiteli ve epitel dejenerasyonu saptandı. Deltamethrin'in Carassius gibelio'nun solungaç, karaciğer ve bağırsak dokuları üzerinde dejeneratif etkilerinin olduğu bulundu. Sonuç olarak Deltamethrin, insan sağlığı ve çevre için çok toksik insektisitlerden biridir. Bu zehirli maddenin kontaminasyonundan sucul ekosistemleri korumak için gerekli önlemler alınmalıdır.
