The translated PQA tests may be appropriate for use with Japanese applicants, though further research considering cultural differences is required.
The uropygial gland is one of the epidermal derivatives in birds. In the beginning of the morphogenesis of the uropygial gland of the quail embryo, placode formation occurs: Epidermal basal cells remain cuboidal until day 7 of incubation. At day 8 they begin to elongate, become columnar, and develop placode-like structure at day 9. To examine the proliferative activity of the epidermal basal cells, we recorded positions of [3H]-thymidine-labelled nuclei in serial sections by autoradiography (ARG) using a digitizer and a microcomputer. The distribution pattern of labelled nuclei was reconstituted and redisplayed on the cathode ray tube (CRT) as two-dimensional computer graphics. Three-dimensional reconstitution of the same serial sections revealed that cuboidal basal cells in the presumptive uropygial placode became columnar. Unlike the feather and scale placodes, the uropygial placode possessed no resting period of DNA synthesis. The labelling index of the uropygial placode increased considerably compared with that of nonplacode epidermis. No dermal condensation was observed beneath the uropygial placode.
Formation of the uropygial papilla and glandular lumena was inhibited when the uropygial rudiment of a day 8 1/3 quail embryo was cultured for 2 days in a chemically defined medium in the presence of 50 ng/mL of epidermal growth factor (EGF). The epithelium of EGF-treated explants remained at the placode stage, or underwent minor invagination into the mesenchyme and became stratified like that of a 12- or 13-day-old embryo. EGF promoted cellular proliferation in the uropygial epithelium and the epidermis adjacent to the gland and it shortened the lag phase of proliferation and markedly stimulated epithelial DNA synthesis, detected immunocytochemically by labeling explants with 5-bromodeoxyuridine (BrdU). The maximal labeling index in EGF-treated uropygial epithelium was 55% higher than in the control. Electron microscopic observation revealed that the basal lamina had become irregular in the EGF-treated explants and that epithelial cytoplasmic processes penetrated through the basal lamina toward the mesenchyme. These same phenomena are observed in vivo when the glandular buds are formed during day 12-13. Some precocious changes occurred in the uropygial epithelium when the rudiment was cultured in the presence of EGF.
Introduction: In this study, we compared the choice of medical specialty and subspecialty interest among problem-based-learning (PBL) graduates and non-PBL graduates. Materials and Methods: Questionnaires were mailed to a total of 1398 female doctors who graduated from Tokyo Women’s Medical University (TWMU) between 1989 and 2003. The response rate was over 30%, giving 248 respondents who had undergone a PBL curriculum (PBL+) and 220 subjects who had not (PBL-). Current specialty of the graduates were compared between the PBL+ and PBL-, and also compared with the general Japanese female doctors (Control 1 and 2) of similar age groups. Respondents were analysed in terms of their interests in subspecialty medical care or general medical practise, which includes comprehensive medical care, primary care and basic medicine. Internal medicine doctors working in the university hospitals were compared with those working outside the university hospitals. Internal medicine doctors were also compared with specialists in ophthalmology, otolaryngology, dermatology and psychiatry. Subjects were compared by odds ratio (OR) to examine group difference in the field of interest. OR >2.0 was considered statistically significant. Results: Most doctors in all groups chose internal medicine. More PBL+ internal medicine doctors showed interests in comprehensive medical care and primary care; more PBL+ internal medicine doctors working outside university hospitals showed interest in comprehensive medical care and primary care when compared with those who were working in the university hospitals. The PBL- graduates did not show such a characteristic. Conclusions: More PBL+ graduates who chose internal medicine showed interest in holistic medical practices such as primary care and community medicine and more PBL+ specialists showed sustained interest in their respective fields. A localized increase of labeling index (L.I.) was observed in bud‐forming epithelium of the uropygial gland of the Japanese quail embryo through computer‐aided analysis. At day 10 glandular buds are not yet formed anywhere in the epithelium and the distribution of proliferative activity in the epithelium is not differential. On the following day the peripheral buds are being formed at the externo‐lateral end of the basal luminal epithelium, L.I. increases locally in these buds. At day 12 a localized increase of L.I. takes place also at the median end; extremely high labeling indices are seen at the tip of the peripheral buds and developing central buds.
