We have previously described an alternatively spliced isoform of IL-4 mRNA that omits exon 2 and is termed IL-4δ2. However, the natural production of IL-4δ2 protein and its association with disease have not been previously assessed due to unavailability of an antibody that interacts with IL-4δ2 without cross-reactivity with full length IL-4. We used a unique monoclonal antibody (mAb) that reacts with IL-4δ2, but not with IL-4, and observed that IL-4δ2 is naturally produced by T cells from patients with asthma, but not from healthy controls. The kinetics of IL-4δ2 and IL-4 production by phorbol myristate acetate (PMA)/ionomycin-activated cells differed, with IL-4δ2 increasing at 48 – 72 h and IL-4 peaking at 24 h. The steady-state levels of IL-4δ2 mRNA varied significantly among the donors and were discordant with the corresponding protein levels, suggesting post-transcriptional regulation of protein production. Polarized Th1 or Th2 lymphocytes were not a major source of IL-4δ2. Stimulation of cultured T lymphocytes with IL- 4δ2 caused elevated production of IFN-γ, IL-10, IL-6, MCP-1, and TNF-α, with notable differences between patients and controls in the production of IFN-γ, IL-10, and IL-6. Thus, IL- 4δ2 is natively produced not only as mRNA but also as a protein by cells other than Th1 or Th2. It is regulated post-transcriptionally, is associated with allergic asthma, and regulates production of other cytokines by primary T lymphocytes. Alternatively spliced interleukin-4 may be a new biomarker, a pathophysiological player, and possibly a molecular target for future therapies in asthma.
A natural splice variant of IL-4 mRNA omits exon 2. It is unknown whether a corresponding protein, IL-4δ2, is also naturally produced. We have developed an mAb against IL-4δ2 with no cross-reactivity against IL-4. Freshly purified T cells from 8 patients with mild or moderate asthma and from 4 healthy controls were tested after up to 96 h activation with PMA/ionomycin. Levels of IL-4 and IL-4δ2 mRNAs were measured by RT-Q-PCR, and secretion of IL-4 and IL-4δ2 proteins was measured by ELISA. All cells expressed IL-4 and IL-4δ2 mRNAs; stimulation caused a 5-12 fold increase in IL-4 mRNA, and a 20-70 fold increase in IL-4δ2 mRNA at 12-24 h of activation. T cells from controls secreted IL-4 but not IL-4δ2 protein, whereas T cells from asthmatics produced both IL-4 and IL-4δ2 proteins in comparable amounts. Levels of IL-4 protein peaked at 24-48 h of activation, whereas IL-4δ2 protein peaked at 72 h. Adenoviral gene delivery of either mouse IL-4 or mouse IL-4δ2 to mouse lungs in vivo induced immune inflammation with accumulation of T and B cells in both cases. Delivery of IL-4 but not IL-4δ2 induced pulmonary eosinophilia, suggesting that IL-4 does and IL-4δ2 does not engage STAT6 signaling. Experiments in primary human T cell cultures revealed that, indeed, IL-4 but not IL-4δ2 induces phosphorylation of STAT6 or STAT3 in a time- and dose-dependent fashion. Thus, IL-4δ2 is naturally secreted as a protein; it is biologically active in vivo without engaging STAT6 or STAT3 signaling.

Ранее нами было показано увеличение эффективности препарата карбамазепина (КБЗ), включенного в наночастицы PLGA (КБЗ-НЧ) в 30 раз, по сравнению со свободной формой (КБЗ). Хроническое применение КБЗ может оказать влияние на поведение. Для КБЗ показаны нормотимические эффекты, а также снижение двигательной активности и тревожности. Изменение поведения при хроническом приеме КБЗ-НЧ в данном исследовании изучили впервые. Эксперимент проводили на крысах Wistar. Животным из 5 групп (n = 60) в течение 28 дней в хвостовую вену вводили следующие препараты: 1) КБЗ: карбамазепин (раствор в 10% Poloxamer 188) 30 мг/кг; 2) КБЗ-НЧ: КБЗ, включенный наночастицы PLGA (суспензия в 1% Poloxamer 188) 1 мг/кг; 3) ненагруженные наночастицы PLGA (плацебо) в 1% Poloxamer 188. Животные в контрольных группах получали физраствор или 10% Poloxamer 188. Поведение исследовали в тестах «открытое поле» (ОП), «приподнятый крестообразный лабиринт» (ПКЛ) и «Лабиринт Барнс».Животные в группах, получавших плацебо, раствор Poloxamer 188 и физраствор, не отличались от контроля ни по одному из исследованных параметров. В тестах ОП и ПКЛ в группе КБЗ отличий от контроля выявлено не было. Крысы из группы КБЗ-НЧ в тесте ОП показали увеличение двигательной и исследовательской активности, а также снижение тревожности животных. В тесте ПКЛ эта же группа показала увеличение времени нахождения в светлых рукавах лабиринта, что также свидетельствует о снижении тревожности. В тесте «Лабиринт Барнс» животные из группы КБЗ-НЧ в процессе обучения дольше оставались на поверхности лабиринта, что связано с их пониженной тревожностью. Однако, они совершали меньше ошибок в финальном тестировании. Животные из группы КБЗ в процессе обучения не отличались от контроля, однако, в финальном тестировании больше времени тратили на поиск убежища.Таким образом, мы показали, что КБЗ снижает обучаемость животных, в то время как КБЗ-НЧ не только повышает обучаемость, но и снижает тревожность, а также увеличивает двигательную и исследовательскую активность.
