ABSTRAKPantoea stewartii subsp. stewartii adalah patogen penyebab penyakit layu stewart pada tanaman jagung yang baru diketahui keberadaannya di Indonesia. Keberadaan patogen ini telah dilaporkan terdapat pada beberapa daerah, namun belum banyak laporan penelitian terkait masalah ini. Penelitian bertujuan mencirikan isolat P. stewartii subsp. stewartii dari beberapa sentra pertanaman jagung di Sumatra Barat. Isolat-isolat P. stewartii subsp. stewartii berasal dari tanaman jagung yang menunjukkan penyakit layu stewart di 6 kabupaten di Sumatera Barat. Daun tanaman jagung yang mempunyai gejala garis kuning sepanjang tulang daun digerus dan disuspensikan, selanjutnya bakterinya diisolasi pada medium nutrient glucose agar (NGA) dan diinkubasi selama 3 × 24 jam pada suhu kamar. Karakter yang diuji ialah morfologi dan fisiologi bakteri, hipersensitivitas, patogenisitas, dan uji polymorfism chain reaction. Berdasarkan karakter morfologi dan fisiologi didapatkan 7 isolat yang menunjukkan karakter Pantoea. Hasil uji PCR membuktikan bahwa isolat LA homolog dengan Pantoea stewartii galur R1-104, isolat PR1 homolog dengan Pantoea stewartii galur R1-132, dan isolat PR2 homolog dengan Pantoea stewartii galur ATCC 29923.Kata kunci: identifikasi, karakterisasi, layu stewart ABSTRACT Pantoea stewartii subsp. stewartii is the pathogen causing stewart's wilt disease in maize. The occurrence of the disease was recently reported in Indonesia with limited information. Research was conducted to characterize several isolates of P. stewartii subsp. stewartii isolated from maize plants collected from 6 districts in West Sumatra. Leaves showing yellow stripes symptoms along the vein was ground and suspeneded then the bacteria was isolated and incubated for 3 × 24 h at room temperature in nutrient glucose agar (NGA). Pure bacterial colonies gained from this medium was used for characterization. Assessment of bacterial characters was conducted based on morphological assay, physiological assay, biological assay (hypersensitivity, and pathogenicity) followed by polymerase chain reaction (PCR)-based identification. Unique characters of the family Enterobacteriaceae was observed on 7 isolates based on morphological and physiological characters, and only 4 isolates (PR1, PP, ST1, LA) gave positive reaction on hypersensitivity and pathogenicity test. Further molecular identification confirmed 3 out of 7 isolates had high homology to Pantoea stewartii, i.e. Pantoea stewartii strain R1-104, R1-132, and ATCC 29923 for isolates LA, PR1, and PR2, respectively.
ABSTRAC The research of giving POC water from washing rice and potato peels on the growth and yield of mung bean (Phaseolus radiatus L) was carried out in from February to May 2021. The aim was to obtain the best POC concentration for growth and yield of mung bean. The study used a randomized design).. The treatments were several POC concentrations, namely: A = 40 mℓ/ℓ water ; B = 50 m/ℓ water ; C = 60 mℓ/ℓ water D = 70 m/ℓ water E = 80 mℓ/ℓ l water F: 90 mℓ/ℓ water Observation data were statistically analyzed by F test and if significantly different, the test was continued with DNMRT with a significance level of 5%. The study showed that the POC of rice washing water and potato peels had no significant effect on plant height, flowering age, age of first harvest, number of pods per plant, percentage of pithy pods per plant, dry weight of harvest, and weight of 100 seeds, but significantly different to the number of primary branches. The best POC concentration as well as reduce production costs growth and yield of mung bean has not been obtained. ABSTRAK Penelitian pemberian POC air cucian beras dan kulit kentang terhadap pertumbuhan dan hasil kacang hijau (Phaseolus radiatus L dilaksanakan di lahan percobaan Universitas Ekasakti Padang, dari Februari sampai Mei 2021. Tujuan untuk mendapatkan konsentrasi POC terbaik bagi pertumbuhan dan hasil kacang hijau. Penelitian menggunakan Rancangan Acak Kelompok dengan 6 Perlakuan 5 Kelompok,. Setiap plot percobaan terdiri dari 4 tanaman, semua tanaman diamati. Perlakuan adalah beberapa konsentrasi POC yaitu: A = 40 mℓ/ℓ air. ; B = 50 mℓ/ℓ air ; C = 60 mℓ/ℓ air ; D = 70 mℓ/ℓ air; E = 80 mℓ/ℓ l air ; F : 90 mℓ/ℓ air Data-data pengamatan dianalisis secara statistik dengan uji F dan bila berbeda nyata, pengujian dilanjutkan DNMRT taraf nyata 5%. Hasil penelitian menunjukkan bahwa POC air cucian beras dan kulit kentang tidak berpengaruh nyata terhadap tinggi tanaman, umur berbunga, umur panen pertama, jumlah polong per tanaman, persentase polong bernas per tanaman, berat kering panen, dan berat berat 100 biji, tetapi berbeda nyata terhadap jumlah cabang primer. Belum diperoleh konsentrasi POC yang terbaik untuk pertumbuhan dan hasil kacang hijau Disarankan untuk melakukan penelitian lanjut, dengan meningkatkan perbedaan konsentrasi POC air cucian beras dan kulit kentang yang akan diberikan.
Labour is a huge potential in the development of work and is closely related to the skill level, number and population growth, the state of the job market, and the mental attitude or college diploma in either general or vocational. The research would like to compare the interest in entrepreneurship of students and vocational students in Padang. The research hypothesis that will be answered through this research is “there is significant lnear, relationship of parenting methods, information, and attitude, together with the information of entrepreneurial interest. Data collected parenting methods, information, attitude towards the information of interested in entrepreneurship. Data were analysed by logistic regression and for dummy variables with scores above the median = 1 and score below the median = 0. The was a significant linear relationship between parenting methods, information, and attitude together the same towards the information of the entrepreneurial interest students. The result of students of Padang State Polytechnic showed that significant linear regression of entrepreneur information to entrepreneur intention and significant linear correlation among variables that are information, model of parents guidance, and attitude of entrepreneur cooperation with entrepreneur intention.
