Tumorous vasculature plays key roles in sustaining tumor growth. Vascular disruption is accompanied by internal coagulation along with platelet recruitment and the resulting suppression of oxygen supply. We intend to artificially create this physiological process to establish the mutual feedback between vascular disruption and platelet-mimicking biotaxis for the cascade amplification of hypoxia-dependent therapy. To prove this concept, mesoporous silica nanoparticles are co-loaded with a hypoxia-activated prodrug (HAP) and a vessel-disruptive agent and then coated with platelet membranes. Upon entering into tumors, our nanotherapeutic can disrupt local vasculature for tumor inhibition. This platelet membrane-coated nanoplatform shares the hemorrhage-tropic function with parental platelets and can be persistently recruited by the vasculature-disrupted tumors. In this way, the intratumoral vascular disruption and tumor targeting are biologically interdependent and mutually reinforced. Relying on this mutual feedback, tumorous hypoxia was largely promoted by more than 20-fold, accounting for the effective recovery of the HAP’s cytotoxicity. Consequently, our bioinspired nanodesign has demonstrated highly specific and effective antitumor potency via the biologically driven cooperation among intratumoral vascular disruption, platelet-mimicking biotaxis, cascade hypoxia amplification, and hypoxia-sensitive chemotherapy. This study offers a paradigm of correlating the therapeutic design with the physiologically occurring events to achieve better therapy performance.

MOD17A2 provides near real-time estimates of gross primary production (GPP) globally. In this study, MOD17A2 GPP was evaluated using eddy covariance (EC) flux measurements at eight sites in five various biome types across China. The sensitivity of MOD17A2 to meteorological data and leaf area index/fractional photosynthetically active radiation (LAI/FPAR) products were examined by introducing site meteorological measurements and improved Global Land Surface Satellite (GLASS) LAI products. We also assessed the potential error contributions from land cover and maximum light use efficiency (ε max ). The results showed that MOD17A2 agreed well with flux measurements of annual GPP (R 2 = 0.76) when all biome types were considered as a whole. However, MOD17A2 was ineffective for estimating annual GPP at mixed forests, evergreen needleleaf forests and croplands, respectively. Moreover, MOD17A2 underestimated flux derived GPP during the summer (R 2 = 0.46). It was found that the meteorological data used in MOD17A2 failed to properly estimate the site measured vapor pressure deficits (VPD) (R 2 = 0.31). Replacing the existing LAI/FPAR data with GLASS LAI products reduced MOD17A2 GPP uncertainties. Though land cover presented the fewest errors, ε max prescribed in MOD17A2 were much lower than inferred ε max calculated from flux data. Thus, the qualities of meteorological data and LAI/FPAR products need to be improved, and ε max should be adjusted to provide better GPP estimates using MOD17A2 for Chinese ecosystems.

Current organoid models are limited by the incapability of rapidly fabricating organoids that can mimic the immune microenvironment for a short term.Here, an acoustic droplet-based platform is presented to facilitate the rapid formation of tumor organoids, which retains the original tumor immune microenvironment and establishes a personalized bladder cancer tumor immunotherapy model. In combination with a hydrophobic substrate, the acoustic droplet printer can yield a large number of homogeneous and highly viable bladder tumor organoids in vitro within a week. The generated organoids consist of all components of bladder tumor, including diverse immune elements and tumor cells. By coculturing tumor organoids with autologous immune cells for 2 days, tumor reactive T cells are induced in vitro. Furthermore, it is also demonstrated that these tumor-reactive T cells can also enhance the killing efficiency of matched organoids. Because of the easy operation, repeatability, and stability, the proposed acoustic droplet platform will provide a reliable approach for personalized tumor immunotherapy.

A homogenate-assisted vacuum-cavitation extraction (HVE) method with a “green” solvent (a deep eutectic solvent, DES) was developed to extract phenolic compounds from rattan (Calamoideae faberii). In this study, the optimum molar ratio of choline chloride (ChCl) and ethylene glycol (EG) was 1:3, the optimum volume ratio of ChCl-EG:H2O was 6:4, the solid-liquid ratio of HVE was 1:15, and the extraction time of homogenate and vacuum-cavitation were 2.0 min and 25 min, respectively. Under the optimum parameters of HVE, the extraction yield of total phenolic content with ChCl-EG solution was 6.82 mg/g. The higher total phenolic content was detected in fruit tissues (seeds 81.24 ± 1.55 mg/g, episperm 43.21 ± 0.87 mg/g, and arillus 38.47 ± 0.74 mg/g), followed by in leaves (sheath 19.5 ± 0.38 mg/g and blade 17.81 ± 0.33 mg/g). In addition, the content of specific phenolic compounds in aqueous and DES extracts was determined. Chlorogenic acid was the most abundant phenol in most organs of the rattan plant. Gallic acid was mainly distributed in the arillus; protocatechuic acid was mainly distributed in the arillus, sheath, and blade; protocatechuic aldehyde was mainly distributed in the blade, seed, and sheath; (+)-catechins were mainly distributed in the episperm, seed, and sheath; and epigallocatechin gallate was mainly distributed in the blade. The recovery rates of gallic acid, protocatechuic acid, protocatechuic aldehyde, (+)-catechins, chlorogenic acid, and epigallocatechin gallate were 93.77%, 94.09%, 97.32%, 97.83%, 94.41%, and 92.47%, respectively, by AB-8 resin.

Cell-based high-throughput screening is a key step in the current disease-based research, drug development, and precision medicine. However, it is challenging to establish a rapid culture and screening platform for rare cells (patient-derived) due to the obvious differences between the traditional 2D cell model and the tumor microenvironment, as well as the lack of a lowconsumption screening platform for low numbers of cells. Here, we developed an acoustic drop-assisted superhydrophilic−superhydrophobic microarray platform for the rapid culture and screening of a few cells. By employing hydrophilic and hydrophobic microarrays, we can automatically distribute the cell suspension into uniform droplets, and these cells can spontaneously form compact 3D cell spheroids within 36 h (similar to the microenvironment of tumors in vivo). By using the acoustic droplet ejection device, we can accurately inject a drug solution with a volume of ∼pL to ∼nL into the droplet, and the whole process can be completed within 20 ms (one print). By using three different cell lines (Caco-2, MCF-7, and HeLa) to optimize the platform, the culture and screening of five patients' colon cancer were subsequently realized. Using three conventional chemotherapeutics (5-fluorouracil, cetuximab, and panitumumab) of various concentrations, the best treatment was screened out and compared with the actual treatment effect of the patients, and the results were extremely similar. As a proof-of-concept application, we have proved that our platform can quickly cultivate patient samples and effectively screen the best treatment methods, highlighting its wide application in precision medicine, basic tumor research, and drug development.
