SUMMARY Oct4A is a core component of the regulatory network of pluripotent cells, and by itself can reprogram neural stem cells into pluripotent cells in mouse and humans. However, its role in defining totipotency and inducing pluripotency during embryonic development is still unclear. We genetically eliminated maternal Oct4A using a Cre-lox approach in mouse and found that the establishment of totipotency was not affected, as shown by the generation of live pups. After complete inactivation of both maternal and zygotic Oct4A expression, the embryos still formed Oct4-GFP– and Nanog–expressing inner cell masses, albeit non-pluripotent, indicating that Oct4A is not a determinant for the pluripotent cell lineage separation. Interestingly, Oct4A-deficient oocytes were able to reprogram fibroblasts into pluripotent cells. Our results clearly demonstrate that, in contrast to its role in the maintenance of pluripotency, maternal Oct4A is crucial for neither the establishment of totipotency in embryos, nor the induction of pluripotency in somatic cells using oocytes.
The chromophores responsible for light absorption in atmospheric brown carbon (BrC) are not well characterized, which hinders our understanding of BrC chemistry, the links with optical properties, and accurate model representations of BrC to global climate and atmospheric oxidative capacity. In this study, the light absorption properties and chromophore composition of three BrC fractions of different polarities were characterized for urban aerosol collected in Xi'an and Beijing in winter 2013−2014. These three BrC fractions show large differences in light absorption and chromophore composition, but the chromophores responsible for light absorption are similar in Xi'an and Beijing. Water-insoluble BrC (WI-BrC) fraction dominates the total BrC absorption at 365 nm in both Xi'an (51 ± 5%) and Beijing (62 ± 13%), followed by a humic-like fraction (HULIS-BrC) and high-polarity water-soluble BrC. The major chromophores identified in HULIS-BrC are nitrophenols and carbonyl oxygenated polycyclic aromatic hydrocarbons (OPAHs) with 2−3 aromatic rings (in total 18 species), accounting for 10% and 14% of the light absorption of HULIS-BrC at 365 nm in Xi'an and Beijing, respectively. In comparison, the major chromophores identified in WI-BrC are PAHs and OPAHs with 4−6 aromatic rings (in total 16 species), contributing 6% and 8% of the light absorption of WI-BrC at 365 nm in Xi'an and Beijing, respectively.
Although Cullin 4A (CUL4A) is mutated or amplified in several human cancer types, its role in gastric cancer (GC) and the mechanisms underlying its regulation remain largely uncharacterized. In the present study, we report that the expression of CUL4A significantly correlated with the clinical stage of the tumor and lymph node metastasis, and survival rates were lower in GC patients with higher levels of CUL4A than in patients with lower CUL4A levels. The upregulation of CUL4A promoted GC cell proliferation and epithelial-mesenchymal transition (EMT) by downregulating LATS1-Hippo-YAP signaling. Knocking down CUL4A had the opposite effect in vitro and in vivo. Interestingly, CUL4A expression was inhibited by the microRNAs (miRNAs), miR-9 and miR-137, which directly targeted the 3′-UTR of CUL4A. Overexpression of miR-9 and miR-137 downregulated the CUL4A-LATS1-Hippo signaling pathway and suppressed GC cell proliferation and invasion in vitro. Taken together, our findings demonstrate that perturbations to miR-9/137-CUL4A-Hippo signaling contribute to gastric tumorigenesis, and suggest potential therapeutic targets for the future treatment of GC.
Emissions from residential biomass burning are a significant source of atmospheric brown carbon (BrC). In this study, we used liquid chromatography-photodiode array-high resolution tandem mass spectrometry to investigate the chromophoric fingerprinting of BrC emitted from residential biomass burning. In total, 59 major chromophores were identified, which accounted for 49−85% of the total light absorption of BrC (averaged between 300 and 500 nm) for different samples. These chromophores include furans, lignin pyrolysis products, coumarins, flavonoids, stilbenes, N-containing aromatic compounds, and unsubstituted or oxygenated polycyclic aromatic hydrocarbons, of which some are newly reported as BrC species (e.g., stilbenes and substituted phenyl cyanates). Among the chromophores identified, seven are common to all samples while some are specific to certain biofuel or burning conditions. For instance, 3,3′dimethoxyquercetin from wheat burning, p-hydroxybenzaldehyde and apigenin from maize smoldering, 4-nitro-2-vinylphenol from maize flaming, and nodakenetin and anthanthrene from wood flaming are specific to the fuel type or burning condition. The identified optical markers will be essential for understanding atmospheric chemical and optical processes of biomass burning BrC in future studies, while the source-specific chromophore profiles developed in this study are a prerequisite for apportioning the biofuel types and burning conditions, which is a key for estimating source-specific radiative forcing.
Risk of metals in urban soils is less studied, compared to that in other types of soils, hindering accurate assessment of human exposure to metals. In this study, the concentrations of five metals (As, Cd, Cr, Pb, and Hg) were analyzed in 167 surface soil samples collected from Suzhou city and their potential ecological and human health risks were assessed. The mean concentrations of As, Cd, Pb, and Hg except Cr, were higher than the background values in Jiangsu Province. Metal concentrations varied among districts, where sites of high contamination showed a punctate distribution. Principal components and correlation analyses revealed that As, Pb, and Cd could originate from the same sources. The geo-accumulation (Igeo) and potential ecological risk indices (RI) were calculated and the relatively low values of Igeo (<0) and RI (<150) suggested generally low ecological risk. The noncarcinogenic risks of the metals were relatively low for Suzhou residents (i.e., average hazard index or HI: 0.1199 for adults and 0.5935 for children, <1), while the total carcinogenic risks (TCR) of Cr and As were acceptable (TCR in the range of 1.0 × 10−6 to 1.0 × 10−4). Children faced a higher threat than adults. Results of Monte-Carlo simulations were lower than those obtained from models using deterministic parameters. Of all the uncertain parameters, the ingestion rate and body weight were the most sensitive for adults and children, respectively, while As was an important factor for both. The results as well as the factors controlling risks of metals could help better understand the risks of metals in urban soils of industrial cities in China.
Vegetation degradation, due to climate change and human activities, changes the biomass, vegetation species composition, and soil nutrient input sources and thus affects soil nutrient cycling and enzyme activities. However, few studies have focused on the responses of soil nutrients and enzymes to vegetation degradation in high-altitude wet meadows. In this study, we examined the effects of vegetation degradation on soil nutrients (soil organic carbon, SOC; total nitrogen, TN; total phosphorus, TP) and enzyme activities (i.e., urease, catalase, amylase) in an alpine meadow in the eastern margin of the Qinghai-Tibet Plateau. Four different levels of degradation were defined in terms of vegetation density and composition: primary wet meadow (CK), lightly degraded (LD), moderately degraded (MD), and heavily degraded (HD). Soil samples were collected at depth intervals of 0–10, 10–20, 20–40, 40–60, 60–80, and 80–100 cm to determine soil nutrient levels and enzyme activities. The results showed that SOC, TN, catalase and amylase significantly decreased with degradation level, while TP and urease increased with degradation level (P < 0.05). Soil nutrient and enzyme activity significantly decreased with soil depth (P < 0.05), and the soil nutrient and enzyme activity exhibited obvious "surface aggregation". The activities of soil urease and catalase were strongest in spring and weakest in winter. The content of TN in spring, summer, and autumn was significantly higher than observed in winter (P < 0.05). The soil TP content increased in winter. Soil amylase activity was significantly higher in summerm than in spring, autumn, and winter (P < 0.05). TP was the main limiting factor for plant growth in the Gahai wet meadow. Values of SOC and TN were positively and significantly correlated with amylase and catalase (P < 0.05), but negatively correlated with urease (P < 0.05). These results suggest the significant role that vegetation degradation and seasonal freeze–thaw cycle play in regulating enzyme activities and nutrient availability in wet meadow soil.
