Genic microsatellite markers, also known as functional markers, are preferred over anonymous markers as they reveal the variation in transcribed genes among individuals. In this study, we developed a total of 707 expressed sequence tag-derived simple sequence repeat markers (EST-SSRs) and used for development of a high-density integrated map using four individual mapping populations of B. rapa. This map contains a total of 1426 markers, consisting of 306 EST-SSRs, 153 intron polymorphic markers, 395 bacterial artificial chromosome-derived SSRs (BAC-SSRs), and 572 public SSRs and other markers covering a total distance of 1245.9 cM of the B. rapa genome. Analysis of allelic diversity in 24 B. rapa germplasm using 234 mapped EST-SSR markers showed amplification of 2 alleles by majority of EST-SSRs, although amplification of alleles ranging from 2 to 8 was found. Transferability analysis of 167 EST-SSRs in 35 species belonging to cultivated and wild brassica relatives showed 42.51% (Sysimprium leteum) to 100% (B. carinata, B. juncea, and B. napus) amplification. Our newly developed EST-SSRs and high-density linkage map based on highly transferable genic markers would facilitate the molecular mapping of quantitative trait loci and the positional cloning of specific genes, in addition to marker-assisted selection and comparative genomic studies of B. rapa with other related species.

Brassica rapa plants are highly important as vegetables, sources of oilseeds and fodder crop. Here, we developed 450 unigene derived microsatellite (UGMS) markers in B. rapa using unigenes downloaded from the National Center for Biotechnology Information database. Of the 450 UGMS primer pairs, 428 (95.1%) produced repeatable and reliable amplifications of expected size in at least one parental line of B. rapa, and 70 UGMS markers gave 72 polymorphic loci between the two contrasting parental lines. Cross-species transferability analysis of these 70 polymorphic UGMS markers in five other cultivated Brassica species showed varying transferability rates ranging from 82.9% in B. nigra to 97.1% in B. juncea and B. napus, and overall 53 UGMS markers amplified targets in all five species. The B. rapa linkage map was constructed using the 72 UGMS polymorphic loci and 154 previously developed SSRs. The newly developed UGMS markers and linkage map in this study would help in future studies to better understand the organization and evolution of Brassica genomes with respect to unigenes, in addition to mapping, tagging and cloning of economically important trait QTL/gene(s) and marker-assisted breeding in Brassica crops.

The inheritance of leaf and heading-related traits in Chinese cabbage (Brassica rapa ssp. pekinensis) are poorly understood since only a handful of studies of these traits are available till date. In this study, QTL mapping was done for seven leaf and heading related traits that includes gross weight, number of non-wrapper leaves, head weight, head length, head width, head length/head width ratio, and the number of head-forming leaves in 139 F3 families derived from a cross between two diverse Chinese cabbage lines, '501' with small head and '601' with large head. A total of 27 QTL spread over the nine linkage groups were identified for seven leaf and head-related traits, many of which showed major effects clustered in A2, A4, A6, and A8. We studied the morphological characteristics, nutritional quality, and the bioactive compounds in fruits of two avocado accessions, RN-7 and RN-8, produced in Hainan province, China. Edible and non-edible parts of the fruit (pulp and seed) were compared to evaluate their possible contribution to improve the sustainability of the food and pharmaceutical industries. The basic characteristics evaluated were moisture, ash, total lipid, fatty acid composition, soluble sugars, titratable acid, soluble protein, and minerals. We also measured the concentrations of six types of bioactive compounds; total phenolics, flavonoids, tannin, ascorbic acid, tocopheryl acetate, and carotenoids. Our analyses of the nutritional compositions demonstrated that the pulp of the RN-7 and RN-8 proved to be rich in moisture, total lipid, and soluble protein. The seed, in turn, had higher soluble sugar, titratable acidity, sodium, potassium, calcium, iron, copper, and zinc contents. Other nutritional compositions (ash, magnesium, and manganese) had little differences between the pulp and seed of avocado fruit. With regard to the contents of bioactive compounds, the seed was superior to the pulp in the contents of total phenolics, flavonoids, and tannin. Regarding the concentrations of ascorbic acid, tocopheryl acetate, and total carotenoids, the highest values were found in the pulp. The results of fatty acid compositions displayed that the palmitic, palmitoleic, stearic, oleic, and linoleic acid contents of the pulp were higher than those of the seed, while myristic and arachic acid had higher contents in the seed.

We propose and demonstrate a novel system for simultaneous ammonia recovery, carbon capture, biogas upgrading, and fertilizer production in biogas production. Biogas slurry pretreatment (adjusting the solution pH, turbidity, and chemical oxygen demand) plays an important role in the system as it significantly affects the performance of ammonia recovery. Vacuum membrane distillation is used to recover ammonia from biogas slurry at various conditions. The ammonia removal efficiency in vacuum membrane distillation is around 75% regardless of the ammonia concentration of the biogas slurry. The recovered ammonia is used for CO absorption to realize simultaneous biogas upgrading and fertilizer generation. CO absorption performance of the recovered ammonia (absorption capacity and rate) is compared with a conventional model absorbent. Theoretical results on biogas upgrading are also provided. After ammonia recovery, the treated biogas slurry has significantly reduced phytotoxicity, improving the applicability for agricultural irrigation. The novel concept demonstrated in this study shows great potential in closing the CO loop in biogas production by recycling ammonia as an absorbent for CO absorption associated with producing fertilizers.

Genomic data is a powerful tool. However, the phylogenetic relationships among different ecological races of avocado remain unclear. Here, we used the results from specific length amplified fragment sequencing (SLAF-seq) and transcriptome data to infer the population structure and genetic diversity of 21 avocado cultivars and reconstructed the phylogeny of three ecological races and two interracial hybrids. The results of the three analyses performed (unweighted pair-group methods with arithmetic means (UPGMA) cluster, Principal coordinate analysis (PCoA), and STRUCTURE) based on single nucleotide polymorphisms (SNPs) from SLAF-seq all indicated the existence of two populations based on botanical race: Mexican–Guatemalan and West Indian genotype populations. Our results based on SNPs from SLAF-seq indicated that the Mexican and Guatemalan races were more closely related to each other than either was to the West Indian race, which also was confirmed in the UPGMA cluster results based on SNPs from transcriptomic data. SNPs from SLAF-seq provided strong evidence that the Guatemalan, Mexican, and Guatemalan × Mexican hybrid accession possessed higher genetic diversity than the West Indian races and Guatemalan × West Indian hybrid accessions. Six race-specific Kompetitive allele specific PCR (KASP) markers based on SNPs from SLAF-seq were then developed and validated.
