Pesticide residues in agricultural produce pose a threat to human health worldwide. Although the detoxification mechanisms for xenobiotics have been extensively studied in mammalian cells, information about the regulation network in plants remains elusive. Here we show that brassinosteroids (BRs), a class of natural plant hormones, decreased residues of common organophosphorus, organochlorine and carbamate pesticides by 30–70% on tomato, rice, tea, broccoli, cucumber, strawberry, and other plants when treated externally. Genome-wide microarray analysis showed that fungicide chlorothalonil (CHT) and BR co-upregulated 301 genes, including a set of detoxifying genes encoding cytochrome P450, oxidoreductase, hydrolase and transferase in tomato plants. The level of BRs was closely related to the respiratory burst oxidase 1 (RBOH1)-encoded NADPH oxides-dependent H2O2 production, glutathione biosynthesis and the redox homeostasis, and the activity of glutathione S-transferase (GST). Gene silencing treatments showed that BRs decreased pesticide residues in plants likely by promoting their metabolism through a signaling pathway involving BRs-induced H2O2 production and cellular redox change. Our study provided a novel approach for minimizing pesticide residues in crops by exploiting plants' own detoxification mechanisms.
Aldehyde dehydrogenases (ALDHs) play a central role in detoxification processes of aldehydes generated in plants when exposed to the stressed conditions. In order to identify genes required for the stresses responses in the grass crop Zea mays, an ALDH (ZmALDH22A1) gene was isolated and characterized. ZmALDH22A1 belongs to the family ALDH22 that is currently known only in plants. The ZmALDH22A1 encodes a protein of 593 amino acids that shares high identity with the orthologs from Saccharum officinarum (95%), Oryza sativa (89%), Triticum aestivum (87%) and Arabidopsis thaliana (77%), respectively. Real-time PCR analysis indicates that ZmALDH22A1 is expressed differentially in different tissues. Various elevated levels of ZmALDH22A1 expression have been detected when the seedling roots exposed to abiotic stresses including dehydration, high salinity and abscisic acid (ABA). Tomato stable transformation of construct expressing the ZmALDH22A1 signal peptide fused with yellow fluorescent protein (YFP) driven by the CaMV35S-promoter reveals that the fusion protein is targeted to plastid. Transgenic tobacco plants overexpressing ZmALDH22A1 shows elevated stresses tolerance. Stresses tolerance in transgenic plants is accompanied by a reduction of malondialdehyde (MDA) derived from cellular lipid peroxidation.
Recently, it has been suggested that molecular hydrogen (H2) can selectively reduce the levels of hydroxyl radicals (.OH), and ameliorate oxidative and inflammatory injuries to organs in global cerebral ischemia reperfusion models. Global cerebral ischemia/reperfusion (I/R) can induce a sudden activation of inflammatory cytokines and later influence the systemic immunoreactivity which may contribute to a worse outcome. Regulatory T cells (Tregs) are involved in several pathological aspects of cerebral I/R. In addition, miRNA took part in the processes of cellular response to hypoxia. Since the expression of a specific set of miRNA called “hypoxamirs” is upregulated by hypoxia. Therefore, the aim of this study was to analyze the effect of HRS on I/R inducing cerebral damage, Tregs, and specific miRNA. Our results showed that rats undergone global cerebral I/R and treated with HRS have milder injury than I/R animals without HRS treatment. miR-210 expression in the hippocampus of the I/R group at 6, 24 and 96 h after reperfusion was significantly increased at each time point, while its expression in the group treated with HRS was significantly decreased. In addition, Tregs number in group I/R was decreased at each time points, while its number in the group treated with HRS was increased at 24 and 96 h after reperfusion. We focus on the relationship among Tregs, TGF-β1, TNF-α and NF-κB at 24 h, and we found that there is a high correlation among them. Therefore, our results indicated that the brain resuscitation mechanism in the HRS-treated rats may be related with the effect of upregulating the number of Treg cells.
Fragmentation and reduced continuity of habitat patches threaten the environment and biodiversity. Recently, ecological networks are increasingly attracting the attention of researchers as they provide fundamental frameworks for environmental protection. This study suggests a set of procedures to construct an ecological network. First, we proposed a method to construct a landscape resistance surface based on the assessment of habitat quality. Second, to analyze the effect of the resistance surface on corridor simulations, we used three methods to construct resistance surfaces: (1) the method proposed in this paper, (2) the entropy coefficient method, and (3) the expert scoring method. Then, we integrated habitat patches and resistance surfaces to identify potential corridors using graph theory. These procedures were tested in Changzhou, China. Comparing the outputs of using different resistance surfaces demonstrated that: (1) different landscape resistance surfaces contribute to how corridors are identified, but only slightly affect the assessment of the importance of habitat patches and potential corridors; (2) the resistance surface, which is constructed based on habitat quality, is more applicable to corridor simulations; and (3) the assessment of the importance of habitat patches is fundamental for ecological network optimization in the conservation of critical habitat patches and corridors.
Qilian juniper (Sabina przewalskii kom.) is one of the dominant tree species on Qinghai-Tibetan Plateau and has been used in dendroclimatological studies. Here we designed a test to examine whether or not the climategrowth responses in tree rings of Qilian Juniper vary with the change in tree's age. A total number of 135 increment cores were sampled from Qilian Juniper trees at five sites, in which 112 cores were selected and grouped into five 100-year age classes for analysis of age-dependent climate-growth relationships. Chronology statistics, response functions and ANOVA F-test were used to test the consistency of five age-class mean chronologies (AGCs). The results showed that mean sensitivity (MS) and standard deviation (SD) did not change significantly with age.
Symbiotic relationships with microbes may influence how plants respond to environmental change. Here, we investigated how fungal endophyte infection affected the growth of a native grass under altered water and nutrient availability. In a two-month field experiment, we compared the performance of endophyte-infected (EI) and endophyte-free (EF) Achnatherum sibiricum subjected to four treatments comprised of a factorial combination of two levels of water availability and two levels of fertilization. The greatest benefits of endophyte infection occurred in the well-watered fertilized treatment. With reduced water and/or nutrient availability, the benefits declined. EI plants subjected to drought and fertilization had higher root:shoot ratios and allocated more nitrogen to photosynthetic machinery and thus had a higher net photosynthetic rate than EF counterparts. In the wellwatered unfertilized treatment, EF plants allocated more nutrients to photosynthetic machinery, while EI plants allocated more resources to defense. Thus EI plants were superior to EF plants in terms of nutrient conservation. In the drought unfertilized treatment, no significant difference occurred between EI and EF plants. Our results support the idea that the endophytegrass interactions are dependent on available resources. However, we did not find a clear cost of endophyte infection. For A. sibiricum, fertilizer addition resulted in greater benefits of the symbiosis for plant growth, but this advantage decreased under drought.
