This paper reports on the comparison analysis of four main types of silicon-based microfilter for isolation of white blood cells (WBCs) from red blood cells (RBCs) in a given whole blood. The microfilter designs, namely, weir, pillar, crossflow, and membrane, all impose the same cut-off size of 3.5 mum to selectively trap WBCs. Using human whole blood, the microfilters have been characterized and compared for their blood handling capacity, WBCs trapping efficiency and RBCs passing efficiency. Based on the experimental results, the crossflow microfilter is superior and can be integrated with downstream components for on-chip genomic analysis.

Cyanobacteriochromes are phytochrome homologues in cyanobacteria that act as sensory photoreceptors. We compare two cyanobacteriochromes, RGS (coded by slr1393) from Synechocystis sp. PCC 6803 and AphC (coded by all2699) from Nostoc sp. PCC 7120. Both contain three GAF (cGMP phosphodiesterase, adenylyl cyclase and FhlA protein) domains (GAF1, GAF2 and GAF3). The respective full-length, truncated and cysteine point-mutated genes were expressed in Escherichia coli together with genes for chromophore biosynthesis. The resulting chromoproteins were analyzed by UV-visible absorption, fluorescence and circular dichroism spectroscopy as well as by mass spectrometry. RGS shows a red-green photochromism (k max = 650 and 535 nm) that is assigned to the reversible 15Z ⁄ E isomerization of a single phycocyanobilin-chromophore (PCB) binding to Cys528 of GAF3. Of the three GAF domains, only GAF3 binds a chromophore and the binding is autocatalytic. RGS autophosphorylates in vitro; this reaction is photoregulated: the 535 nm state containing E-PCB was more active than the 650 nm state containing Z-PCB. AphC from Nostoc could be chromophorylated at two GAF domains, namely GAF1 and GAF3. PCB-GAF1 is photochromic, with the proposed 15E state (k max = 685 nm) reverting slowly thermally to the thermostable 15Z state (k max = 635 nm). PCB-GAF3 showed a novel red-orange photochromism; the unstable state (putative 15E, k max = 595 nm) reverts very rapidly (s 20 s) back to the thermostable Z state (k max = 645 nm). The photochemistry of doubly chromophorylated AphC is accordingly complex, as is the autophosphorylation: E-GAF1 ⁄ E-GAF3 shows the highest rate of autophosphorylation activity, while E-GAF1 ⁄ Z-GAF3 has intermediate activity, and Z-GAF1 ⁄ Z-GAF3 is the least active state. The thermal and photochemical processes associated with the acid-induced conversions of 6-nitroBIPS, SP-1, to form the protonated merocyanine (MC-OH+) were investigated via UV/vis spectrophotometric studies in acetone. It was found that the mechanism of trifluoroacetic acid (TFA)-induced ring-opening of the SP and the rate of MC-OH+ formation follows a general acid catalysis mechanism. In accord with this mechanism, the thermal growth of the acid-induced ring-opened form (MC-OH+) was retarded as the concentration of TFA in the medium was increased. The N-protonated SP, i.e., SP-NH+, is formed in a competing side-equilibrium process as an unreactive "sink", with the nitrogen lone-pair no longer available to drive the ring-opening process and resulting in the inverse rate dependence as a linear 1/kobs vs [HA] plot. Addition of a tertiary amine to MC-OH+ regenerated MC which underwent thermal ring closure to the SP, thus restoring its function as a molecular switch. NMR titration of SP samples showed a downfield shift of the N-substituent peak upon increasing the TFA concentration. However, a saturation behavior could not be observed with SP-1 up to 1 M acid, unlike the model compound, N,N-dimethylaniline (N,N-DMA), which indicates a base strength order of N,N-DMA > SP-1. Further, we have demonstrated that in solvent acetone, on acidification, the normal photo- and thermochromic behavior is reversed; now MC-OH+ is photochemically transformed into SP-H+, which undergoes thermal ring-opening to MC-OH+.

Chemical conditioning process included rapid aggregation and floc densification. SRF correlated well with the change in content of soluble EPS, LB-EPS and TB-EPS. Compression of EPS structure was a major mechanism of dewaterability improvement. This study investigated the effects of chemical conditioning with three representative inorganic coagulants (FeCl 3 , polyaluminum chloride (PACl) and high performance PACl (HPAC) on sludge dewaterability. We monitored the particle size, kinetic viscosity (KV), fractal dimension (D F ) and extracellular polymeric substances (EPS) located in different layers of sludge floc (soluble, loosely-bound (LB), tightly-bound (TB)) to understand the correlation of dewaterability and physicochemical properties of sludge under chemical conditioning. The conditioning process included the rapid aggregation of sludge particles induced by charge neutralization and bridging followed by floc densification caused by double electric layer compression. The floc size and D F were increased after chemical conditioning, indicating that larger and more compact floc formed. The floc conditioned with FeCl 3 was smaller but denser than that of PACl and HPAC. Furthermore, sludge dewaterability correlated well with the change in concentration of soluble EPS, LB-EPS and TB-EPS, but not with KV, D F , floc size.

One of the main bottlenecks that limit the performance of reversible protonic ceramic electrochemical cells (R‐PCECs) is the sluggish kinetics of the oxygen reduction and evolution reactions (ORR and OER). Here, the significantly enhanced ORR and OER kinetics and stability of a conventional La0.6Sr0.4Co0.2Fe0.8O3–δ (LSCF) air electrode by an efficient catalyst coating of barium cobaltite (BCO) is reported. The polarization resistance of a BCO‐coated LSCF air electrode at 600 °C is 0.16 Ω cm2, about 30% of that of the bare LSCF air electrode under the same conditions. Further, an R‐PCEC with the BCO‐coated LSCF air electrode shows exceptional performance in both fuel cell (peak power density of 1.16 W cm−2 at 600 °C) and electrolysis (current density of 1.80 A cm−2 at 600 °C at 1.3 V) modes. The performance enhancement is attributed mainly to the facilitated rate of oxygen surface exchange.
