It has been a great challenge to make the thickness of engineered cartilage adjustable to cover the range of both partial-thickness and full-thickness articular cartilage defects. We developed a novel kind of composite web scaffold that could be used for tissue enginnering of articular cartilage with the thickness adjustable between 200 microm and 8 mm. The composite web showed a unique structure having web-like collagen microsponges formed in the openings of a mechanically strong knitted mesh of poly(lactic-co-glycolic acid). The knitted mesh served as a skeleton reinforcing the composite web, while the web-like collagen microsponges facilitated cell seeding, cell distribution, and tissue formation. Bovine chondrocytes cultured in the composite web showed a spatially even distribution, maintained their natural morphology, and produced cartilaginous extracellular matrices such as type II collagen and aggrecan. The thickness of the implant can be simply adjusted by laminating or rolling the web sheets. Not only did the histological structure of the engineered cartilage patches match the bovine native articular cartilage, but also their dynamic complex modulus, structural stiffness, and phase lag reached 37.8, 57.0, and 86.3% of those of native bovine articular cartilage, respectively. The composite web could be an important scaffold for tissue engineering.

Effects of long-term exercise on volumetric bone mineral density (vBMD), bone mineral content, bone geometric properties, and the strength indexes of bone were examined in a cross-sectional study of athletes and controls. Tibias of 25 jumpers (13 women), 30 swimmers (15 women), and 25 controls (15 women), aged 18-23 yr, were scanned at midsite by using peripheral quantitative computed tomography. The cortical vBMD of female athletes was lower than that of the controls (2.00 +/- 0.05, 1.90 +/- 0.08, and 1.92 +/- 0.12 g/cm3, respectively, for controls, swimmers, and jumpers). On the other hand, periosteal areas of male jumpers and female athletes were greater than that of controls (460 +/- 50, 483 +/- 46, and 512 +/- 55 mm2, respectively, for male controls, swimmers, and jumpers, and 283 +/- 52, 341 +/- 73, and 378 +/- 75 mm2, respectively, for female controls, swimmers, and jumpers). The endocortical area of female swimmers was greater than that of controls (103 +/- 29, 148 +/- 52, and 135 +/- 54 mm2, respectively, for controls, swimmers, and jumpers). The polar moment of inertia and strength strain index of male jumpers and female athletes were significantly greater than those of controls, except for the difference in strength strain index between male jumpers and controls. We conclude that the improvement of mechanical properties of young adult bone in response to long-term exercise is related to geometric adaptation and not to vBMD.

Tension is necessary to maintain and restore the mechanical properties of soft connective tissues. Conversely, reduced tension states such as produced by immobilization weaken mechanical properties and facilitate joint contracture. We assessed the effect of low torque-long duration stretching to increase the range of motion (ROM) and to restore the mechanical properties of contracted joints in 66 rat knees immobilized for 40 days. After remobilization, we randomly divided the contracted knees into four treatment groups treated with repeated stretches of diverse torques and duration: stretching with low-torque and long-duration, high-torque and short-duration, high-torque and long-duration, low-torque and short duration. We included control and natural recovery groups. Phase lag in all treatment groups recovered to the same range as in the normal controls. Dynamic stiffness, which was not altered by joint immobilization, increased in all treatment groups. Deformation and load to failure improved substantially only in the low-torque and long-duration stretching group. Low-torque and long-duration repeated stretching leads to a greater restoration of ROM with more normal mechanical properties compared to high-torque and short duration stretching.

The experiment shows that using bFGF, better mechanical strength was obtained but with poor cartilage growth.
