The Land Transport Authority of Singapore has a continuing program of highway bridge upgrading for refurbishing and strengthening bridges to allow for increasing vehicle traffic and increasing axle loads. One subject of this program has been a short-span bridge taking a busy main road across a coastal inlet near a major port facility. Experiment-based structural assessments of the bridge were conducted before and after upgrading works including strengthening. Each assessment exercise comprised three separate components: ͑1͒ a strain and acceleration monitoring exercise lasting approximately one month; ͑2͒ a full-scale dynamic test carried out in a single day without closing the bridge; and ͑3͒ a finite-element model updating exercise to identify structural parameters and mechanisms. This paper presents the dynamic testing and the modal analysis used to identify the vibration properties and the quantification of the effectiveness of the upgrading through the subsequent model updating. Before and after upgrade, similar sets of vibration modes were identified, resembling those of an orthotropic plate with relatively weak transverse bending stiffness. Conversion of bearings from nominal simple supports to nominal full fixity was shown via model updating to be the principal cause of natural frequency increases of up to 50%. The utility of the combined experimental and analytical process in direct identification of structural properties has been proven, and the procedure can be applied to other structures and their capacity assessments.
Three ATP binding cassette (ABC) transporter-like activities directed toward large amphipathic organic anions have recently been identified on the vacuolar membrane of plant cells. These are the Mg-ATP-energized, vanadate-inhibitable vacuolar accumulation of glutathione S-conjugates (GS conjugates), chlorophyll catabolites, and bile acids, respectively. Although each of these activities previously had been assigned to distinct pumps in native plant membranes, we describe here the molecular cloning, physical mapping, and heterologous expression of a gene, AtMRP2, from Arabidopsis thaliana that encodes a multispecific ABC transporter competent in the transport of both GS conjugates and chlorophyll catabolites. Unlike its isoform, AtMRP1, which transports the model Brassica napus chlorophyll catabolite transporter substrate Bn-NCC-1 at low efficiency, heterologously expressed AtMRP2 has the facility for simultaneous high-efficiency parallel transport of GS conjugates and Bn-NCC-1. The properties of AtMRP2 therefore establish a basis for the manipulation of two previously identified plant ABC transporter activities and provide an explanation for how the comparable transporter in native plant membranes would be systematically mistaken for two distinct transporters. These findings are discussed with respect to the functional organization of AtMRP2, the inability of AtMRP2 and AtMRP1 to transport the model bile acid transporter substrate taurocholate (despite the pronounced sensitivity of both to direct inhibition by this agent), the differential patterns of expression of their genes in the intact plant, and the high capacity of AtMRP2 for the transport of glutathionated herbicides and anthocyanins.
Piezoelectric ceramic Lead Zirconate Titanate (PZT) based electro-mechanical impedance (EMI) technique for structural health monitoring (SHM) has been successfully applied to various engineering systems. However, fundamental research work on the sensitivity of the PZT impedance sensors for damage detection is still in need. In the traditional EMI method, the PZT electro-mechanical (EM) admittance (inverse of the impedance) is used as damage indicator, which is difficult to specify the effect of damage on structural properties. This paper uses the structural mechanical impedance (SMI) extracted from the PZT EM admittance signature as the damage indicator. A comparison study on the sensitivity of the EM admittance and the structural mechanical impedance to the damages in a concrete structure is conducted. Results show that the SMI is more sensitive to the damage than the EM admittance thus a better indicator for damage detection. Furthermore, this paper proposes a dynamic system consisting of a number of single-degree-of-freedom elements with mass, spring and damper components to model the SMI. A genetic algorithm is employed to search for the optimal value of the unknown parameters in the dynamic system. An experiment is carried out on a two-storey concrete frame subjected to base vibrations that simulate earthquake. A number of PZT sensors are regularly arrayed and bonded to the frame structure to acquire PZT EM admittance signatures. The relationship between the damage index and the distance of the PZT sensor from the damage is studied. Consequently, the sensitivity of the PZT sensors is discussed and their sensing region in concrete is derived.
