Quantitative biomechanical models can identify control parameters used during movements, and movement parameters encoded by premotor neurons. We fit a mathematical dynamical systems model including subsyringeal pressure, syringeal biomechanics, and upper vocal tract filtering to the songs of zebra finches. This reduced the dimensionality of singing dynamics, described as trajectories in pressure-tension space (motor “gestures”). We assessed model performance by characterizing the auditory response "replay" of song premotor HVC neurons to presentation of song variants in sleeping birds, and by examining HVC activity in singing birds. HVC projection neurons were excited and interneurons were suppressed with near-zero time lag, at times of gesture trajectory extrema. Thus, HVC precisely encodes vocal motor output via the timing of extreme points of movement trajectories. We propose that the sequential activity of HVC neurons represents the sequence of gestures in song as a “forward” model making predictions on expected behavior to evaluate feedback.
We reconstruct the physiological parameters that control an avian vocal organ during birdsong production using recorded song. The procedure involves fitting the time dependent parameters of an avian vocal organ model. Computationally, the model is implemented as a dynamical system ruling the behavior of the oscillating labia that modulate the air flow during sound production, together with the equations describing the dynamics of pressure fluctuations in the vocal tract. We tested our procedure for Zebra finch song with, simultaneously recorded physiological parameters: air sac pressure and the electromyographic activity of the left and right ventral syringeal muscles. A comparison of the reconstructed instructions with measured physiological parameters during song shows a high degree of correlation. Integrating the model with reconstructed parameters leads to the synthesis of highly realistic songs.
To further our understanding of the genetics of musicality, we explored associations between a polygenic score for self-reported beat synchronization ability (PGS rhythm ) and objectively measured rhythm discrimination, as well as other validated music skills and music-related traits. Using family data, we were able to further explore potential pathways of direct genetic, indirect genetic (through passive gene-environment correlation) and confounding effects (such as population structure and assortative mating). In 5648 Swedish twins, we found PGS rhythm to predict not only rhythm discrimination, but also melody and pitch discrimination (betas between 0.11 and 0.16, p < 0.001), as well as other music-related outcomes (p < 0.05). In contrast, PGS rhythm was not associated with control phenotypes not directly related to music. Associations did not deteriorate within families (N = 243), implying that indirect genetic or confounding effects did not inflate PGS rhythm effects. A correlation (r = 0.05, p < 0.001) between musical enrichment of the family childhood environment and individuals' PGS rhythm , suggests gene-environment correlation. We conclude that the PGS rhythm captures individuals' general genetic musical propensity, affecting musical behavior more likely direct than through indirect or confounding effects.Listening to music or engaging actively in music by singing or playing an instrument, is an important part of life for many. Twin research on music-related behavior shows that there is a substantial genetic component to variation in musical engagement, as well as to musical ear, i.e. pitch, melody and rhythm discrimination skills, with heritability estimates ranging between 13 and 86% with an average around 40-50% [1][2][3][4][5][6][7] . Twin modelling studies have also shown that there is considerable overlap between the genetic variants underlying such music-related phenotypes (so-called 'genetic pleiotropy'), not only between different specific music skills, but also between music skills and other music-related traits, like music practice or listening to music 3,8,9 . This suggests that there are generalist genes as well as specialist genes underlying variation in music-related traits.Which particular genes are relevant for music skills is not yet well understood, with the limited existing research largely being based on candidate gene and linkage studies (see Gingras et al. 10 for review), which are known to suffer from replication problems, and are hampered by small sample size 11 . Recently, a genome-wide association study (GWAS) in a well-powered sample of 606,825 individuals identified sixty-nine genetic variants that influence variation in self-reported beat synchronization ability, assessed with the question: 'Can you clap in time with a musical beat?' Yes/No 12 . The variance explained by the aggregate of these genetic effects (i.e., the SNP-based heritability) was ~ 13-16%. Psychedelic drugs, including lysergic acid diethylamide and other agonists of the serotonin 2A receptor (5HT2A-R), induce drastic changes in subjective experience, and provide a unique opportunity to study the neurobiological basis of consciousness. One of the most notable neurophysiological signatures of psychedelics, increased entropy in spontaneous neural activity, is thought to be of relevance to the psychedelic experience, mediating both acute alterations in consciousness and long-term effects. However, no clear mechanistic explanation for this entropy increase has been put forward so far. We sought to do this here by building upon a recent whole-brain model of serotonergic neuromodulation, to study the entropic effects of 5HT2A-R activation. Our results reproduce the overall entropy increase observed in previous experiments in vivo, providing the first model-based explanation for this phenomenon. We also found that entropy changes were not uniform across the brain: entropy increased in some regions and decreased in others, suggesting a topographical reconfiguration mediated by 5HT2A-R activation. Interestingly, at the whole-brain level, this reconfiguration was not well explained by 5HT2A-R density, but related closely to the topological properties of the brain’s anatomical connectivity. These results help us understand the mechanisms underlying the psychedelic state and, more generally, the pharmacological modulation of whole-brain activity.
The scope of human consciousness includes states departing from what most of us experience as ordinary wakefulness. These altered states of consciousness constitute a prime opportunity to study how global changes in brain activity relate to different varieties of subjective experience. We consider the problem of explaining how global signatures of altered consciousness arise from the interplay between large-scale connectivity and local dynamical rules that can be traced to known properties of neural tissue. For this purpose, we advocate a research program aimed at bridging the gap between bottom-up generative models of whole-brain activity and the top-down signatures proposed by theories of consciousness. Throughout this paper, we define altered states of consciousness, discuss relevant signatures of consciousness observed in brain activity, and introduce whole-brain models to explore the biophysics of altered consciousness from the bottom-up. We discuss the potential of our proposal in view of the current state of the art, give specific examples of how this research agenda might play out, and emphasize how a systematic investigation of altered states of consciousness via bottom-up modeling may help us better understand the biophysical, informational, and dynamical underpinnings of consciousness.

Background: N,N-dimethyltryptamine is a short-acting psychedelic tryptamine found naturally in many plants and animals. Few studies to date have addressed the neural and psychological effects of N,N-dimethyltryptamine alone, either administered intravenously or inhaled in freebase form, and none have been conducted in natural settings. Aims: Our primary aim was to study the acute effects of inhaled N,N-dimethyltryptamine in natural settings, focusing on questions tuned to the advantages of conducting field research, including the effects of contextual factors (i.e. “set“ and “setting“), the possibility of studying a comparatively large number of subjects, and the relaxed mental state of participants consuming N,N-dimethyltryptamine in familiar and comfortable settings. Methods: We combined state-of-the-art wireless electroencephalography with psychometric questionnaires to study the neural and subjective effects of naturalistic N,N-dimethyltryptamine use in 35 healthy and experienced participants. Results: We observed that N,N-dimethyltryptamine significantly decreased the power of alpha (8–12 Hz) oscillations throughout all scalp locations, while simultaneously increasing power of delta (1–4 Hz) and gamma (30–40 Hz) oscillations. Gamma power increases correlated with subjective reports indicative of some features of mystical-type experiences. N,N-dimethyltryptamine also increased global synchrony and metastability in the gamma band while decreasing those measures in the alpha band. Conclusions: Our results are consistent with previous studies of psychedelic action in the human brain, while at the same time the results suggest potential electroencephalography markers of mystical-type experiences in natural settings, thus highlighting the importance of investigating these compounds in the contexts where they are naturally consumed.
