Clustered regularly interspaced short palindromic repeats (CRISPR)-associated Cas9protein is an effector that plays a major role in a prokaryotic adaptive immune system, by which invading DNA can be targeted and cut for inactivation. The Cas9 endonuclease is directed to target sites by a guide RNA (gRNA) where Cas9 can recognize specific sequences (PAMs) in foreign DNA, which then serve as an anchoring point for cleavage of the adjacent RNA-matching DNA region. Although the CRISPR-Cas9 system has been widely studied and repurposed for diverse applications (notably, genome editing), its origin and evolution remain to be elucidated. Here, we investigate the evolution of Cas9 from resurrected ancient nucleases (anCas) in extinct firmicutes species as old as 2600 myr to the current day. Surprisingly, we demonstrate that these ancient forms were much more flexible in their PAM and gRNA scaffold requirements compared to modern day Cas9 enzymes. In addition, anCas portrays a gradual paleoenzymatic adaptation from nickase to double-strand break activity, suggesting a mechanism by which ancient CRISPR systems could propagate when harboring Cas enzymes with minimal PAMs. The oldest anCas also exhibit high levels of activity with ssDNA and ssRNA targets, resembling Cas nucleases in related system types. Finally, we illustrate editing activity of the anCas enzymes in human cells. The prediction and characterization of anCas proteins uncovers an unexpected evolutionary trajectory leading to ancient enzymes with extraordinary properties.
The Inventory of Emotions Experienced by Adolescents when Solving Mathematical Tasks (INETAM, for its acronym in Spanish), measures four influential emotions related to Math performance: Enthusiasm, Frustration, Enjoyment, and Boredom. Content validity, construct validation and item calibration analyses were performed to obtain evidence of its validity, using a sample of 448 adolescents enrolled in ninth grade. Factor analysis showed adequate reliability coefficients and fit indices. Calibration analysis showed that the items are highly informative and discriminate between response levels. Regression analysis indicated that emotions are predictors of math achievement. INETAM is solid psychometrically, suitable for measuring academic emotions of adolescents, and can contribute to improve our understanding of their influence on academic achievement in mathematics.
Abstract.Under the theoretical basis of the interbehavioral psychology, were developed and validated two tests to assess reading comprehension of texts related with two topics: one literary and one of biology, in three functional levels. The aim was to analyze the performance of junior high school students, in reading comprehension. Participants were 90 students enrolled in the first grade of three school groups from a public school. The results show that students achieved low percentages of correct answers in both tests, in particular, their worst performance was related with the topic of biology. The results of the students were classified in three ranges of performance. The advantages of evaluating reading comprehension considering the functional levels of behavior and topic, are discussed.
Shared story time reading promotes psychological abilities which are important in child development, including linguistic, conceptual and socio-emotional skills. The propose of the study was to analyse the relation between the actions of preschool children´s mothers with the types of interaction and the behaviour showed by their children during dyadic story time activities. This was an observational-analytic study. We studied 30 mothers with children (13 girls and 17 boys; average age five years) registered in third-grade preschool from a public school of low sociocultural level. Each pair was filmed in two sessions and analysed through taxonomy of behavioural categories. Initial data showed that most mothers presented limited skills as storytellers, readers, promoters of linguistic interactions and as informal teachers. The differences between groups and the correlations among maternal-infant behavioural categories indicated synchrony between the behaviour in the mothers and the performance on the child, particularly on the linguistic interactions.
