Tea (Camellia sinensis) is an important commercial crop, in which the high content of flavonoids provides health benefits. A flavonoid glycosyltransferase (CsUGT73A20), belonging to cluster IIIa, was isolated from tea plant. The recombinant CsUGT73A20 in Escherichia coli exhibited a broad substrate tolerance toward multiple flavonoids. Among them, kaempferol was the optimal substrate compared to quercetin, myricetin, naringenin, apigenin, and kaempferide. However, no product was detected when UDP-galactose was used as the sugar donor. The reaction assay indicated that rCsUGT73A20 performed multisite glycosidation toward flavonol compounds, mainly forming 3-O-glucoside and 7-O-glucoside in vitro. The biochemical characterization analysis of CsUGT73A20 showed more K7G product accumulated at pH 8.0, but K3G was the main product at pH 9.0. Kinetic analysis demonstrated that high pH repressed the glycosylation reaction at the 7-OH site in vitro. Besides, the content of five flavonol-glucosides was increased in CsUGT73A20-overexpressing tobaccos (Nicotiana tabacum).
Epidemiological reports have implicated contaminated raw or undercooked chicken as primary vehicles of Salmonella transmission to human beings. Risk assessments relating to Salmonella contamination of poultry products in China are frequently hampered by the lack of quantitative data. In this study, whole chicken carcasses (n=395) were collected from the retail markets of Beijing, and the level of Salmonella contamination was enumerated by most probable number (MPN) analysis and all Salmonella isolates were further characterized for their serotypes and antimicrobial resistance. Overall, 49.9% (197/395) of the retail whole chicken carcasses were contaminated by Salmonella and the MPN values ranged from 1.5 to >550 MPN/100 g. The 50% percentile of Salmonella MPN value was 7.5 MPN/100 g in chicken carcass. The predominant serotypes isolated were Salmonella Enteritidis (n=309, 94 samples), Salmonella Indiana (n=205, 54 samples) and Salmonella Infantis (n=89, 23 samples). Multidrug-resistant Salmonella isolates were recovered from 100 chicken carcass samples; 102 isolates (from 41 chicken carcasses) even showed resistance to both ciprofloxacin and cefotaxime. Our findings showed a high prevalence of Salmonella contamination in retail chicken carcasses, which could be a source of exposure for consumers to multidrug-resistant isolates. This study provided baseline enumeration data for the risk assessment and evaluation of new control measures of Salmonella contamination in retail chicken products.
A study on the natural occurrence of aflatoxins in Chinese peanut butter and sesame paste samples was conducted. Aflatoxin B(1) (AFB(1)) was the predominant toxin detected abundantly and frequently at a level up to 68.51 microg/kg in 41 of 50 peanut butter samples and 20.45 microg/kg in 37 of 100 sesame paste samples analyzed by liquid chromatography (LC). Of the AFB(1)-positive samples, 15 (37%) and 1 (2%) peanut butter samples with AFB(1) exceed European Union (EU) and Chinese regulations, respectively, whereas 19 and 32% of sesame paste samples contained AFB(1) higher than Chinese and EU regulations, respectively. Fourteen and 1 peanut butter samples and 10 and 7 sesame paste samples, respectively, will be legally claimed as positive, rejected, and even banned with consideration of an uncertainty of 40% for AFB(1), based on EU and Chinese regulations. Seeking to balance health benefits with the potential trade disruptions that regulations can cause is the issue of concern.
