We incidentally found that osteoclast precursors and mature osteoclasts express Fas ligand (FasL) as well as Fas, which was confirmed by flow cytometry, immunofluorescent staining, and RT-PCR. The aim of this study was to determine the role of FasL in differentiation and cell death of osteoclasts. To study the role of FasL in osteoclastogenesis, neutralizing anti-FasL mAb or rFasL was added during receptor activator of NF-κB ligand (RANKL)-induced osteoclastogenesis using bone marrow-derived macrophages. Neutralization of endogenous FasL by anti-FasL mAb decreased osteoclastogenesis, whereas rFasL enhanced osteoclast differentiation in a dose-dependent manner. In addition, rFasL up-regulated the secretion of osteoclastogenic cytokines, such as IL-1β and TNF-α, and the activation of NF-κB. Functional blocking of IL-1β and TNF-α using IL-1 receptor antagonist and soluble TNFR confirmed that those cytokines mediated the effect of FasL on osteoclastogenesis. The osteoclast precursors were relatively resistant to rFasL-induced apoptosis especially before RANKL treatment, resulting in minimal cell loss by rFasL treatment during osteoclastogenesis. Although rFasL increased the cell death of mature osteoclasts, growth factor withdrawal induced much more cell death. However, anti-FasL mAb did not affect the survival of mature osteoclasts, suggesting that the endogenous FasL does not have a role in the apoptosis of osteoclasts. Finally, in contrast to the effect on apoptosis, rFasL-assisted osteoclastogenesis was not mediated by caspases. In conclusion, FasL has a novel function in bone homeostasis by enhancing the differentiation of osteoclasts, which was not considered previously.
BackgroundPlasma cholesterol acts as a negative acute phase reactant. Total cholesterol decreases after surgery and in various pathological conditions, including trauma, sepsis, burns, and liver dysfunction. This study aimed to determine whether hypocholesterolemia after emergency gastrointestinal (GI) surgery is associated with in-hospital mortality in patients with diffuse peritonitis.MethodsThe medical records of 926 critically ill patients who had undergone emergency GI surgery for diffuse peritonitis, between January 2007 and December 2015, were retrospectively analyzed. The integrated areas under the curve (iAUCs) were calculated to compare the predictive accuracy of total cholesterol values from postoperative days (PODs) 0, 1, 3, and 7. Cox proportional hazard regression modeling was performed for all possible predictors identified in the univariate and multivariable analyses.ResultsThe total cholesterol level measured on POD 7 had the highest iAUC (0.7292; 95% confidence interval, 0.6696–0.7891) and was significantly better at predicting in-hospital mortality than measurements on other days. The optimal total cholesterol cut-off value for predicting in-hospital mortality was 61 mg/dL and was determined on POD 7. A Cox proportional hazard regression analysis revealed that a POD 7 total cholesterol level < 61 mg/dL was an independent predictor of in-hospital mortality after emergency GI surgery (hazard ratio, 3.961; 95% confidence interval, 1.786–8.784).ConclusionSevere persistent hypocholesterolemia (<61 mg/dL) on POD 7 independently predicted in-hospital mortality, after emergency GI surgery, in critically ill patients with diffuse peritonitis.
An important capability for a subdivision scheme is the reproducing property of circular shapes or parts of conics that are important analytical shapes in geometrical modeling. In this regards, this study first provides necessary and sufficient conditions for a non-stationary subdivision to have the reproducing property of exponential polynomials. Then, the approximation order of such non-stationary schemes is discussed to quantify their approximation power. Based on these results, we see that the exponential B-spline generates exponential polynomials in the associated spaces, but it may not reproduce any exponential polynomials. Thus, we present normalized exponential B-splines that reproduce certain sets of exponential polynomials. One interesting feature is that the set of exponential polynomials to be reproduced is varied depending on the normalization factor. This provides us with the necessary accuracy and flexibility in designing target curves and surfaces. Some numerical results are presented to support the advantages of the normalized scheme by comparing them to the results without normalization.
A single-scan dual-energy low-dose cone-beam CT (CBCT) imaging technique that exploits a multi-slit filter is proposed in this paper. The multi-slit filter installed between the x-ray source and the scanned object is reciprocated during a scan. The x-ray beams through the slits would generate relatively low-energy x-ray projection data, while the filtered beams would make high-energy projection data. An iterative image reconstruction algorithm that uses an adaptive-steepest-descent method to minimize image total-variation under the constraint of data fidelity was applied to reconstructing the image from the low-energy projection data. Since the high-energy projection data suffer from a substantially high noise level due to the beam filtration, we have developed a new algorithm that exploits the joint sparsity between the low- and high-energy CT images for image reconstruction of the high-energy CT image. The proposed image reconstruction algorithm uses a gradient magnitude image (GMI) of the low-energy CT image by regularizing the difference of GMIs of the low- and high-energy CT images to be minimized. The feasibility of the proposed technique has been demonstrated by the use of various phantoms in the experimental CBCT setup. Furthermore, based on the proposed dual-energy imaging, a material differentiation was performed and its potential utility has been shown. The proposed imaging technique produced promising results for its potential application to a low-dose single-scan dual-energy CBCT.
A dramatic increase has been registered in the number of social media posts in photo form as well as in hashtag activism. Hashtags, which manifest thoughts and feelings clearly and concisely, originated on Twitter, where the length of a post is limited; their use, however, has expanded into other social media services, including Instagram. Hashtags, which make it easy to find and express support for posts of interest, have been widely used for online activism, although they have been criticized for fostering confirmation bias. Moreover, hashtag activism in photo form has been relatively understudied. This research analyzed Instagram photos with antivaccination hashtags as an example of hashtag activism through photos. In addition, we examined how the photo features were related to public response, which was manifested via engagement and comment sentiment. The results suggest that the photos which were categorized into “text” took the largest share. It was also found that the major way of claiming was to imprint key messages that persuade people not to vaccinate with remarks from professionals on photos and provide a source of supporting information in the post text with hashtags of antivaccine intention. Various photo features showed associations with engagement and comment sentiment, but the directions of correlation were usually the opposite: these results suggest that engagement and comment sentiment are separate domains that reveal different public responses.
