Pet dogs have been considered to be involved in the contamination of indoor air by serving as a source of providing molds at houses. Currently, information on the molds originated from pet dogs is rarely available in Korea. The present study was carried out to obtain basic information on the fungi present on pet dogs. For this, fungal isolation was performed to the skin and hairs of 70 pet dogs at different houses and veterinary hospitals. A total of 44 fungal isolates were obtained from skin (27 isolates) and hairs (17 isolates) of the dogs investigated. Based on the observation of microstructures and colony morphology, and the ITS rDNA sequence analysis, the fungal isolates were identified at the level of genus. The identified isolates belong to the genera of Alternaria, Aspergillus, Beauveria, Chrysosporium, Cladosporium, Penicillium, Scopulariopsis, and Trichoderma. Among these genera, Aspergillus (25%), Cladosporium (23%) and Penicillium (20.5%) were 3 major genera. 63% of the 44 isolates showed color changes on dermatophyte test medium (DTM). When we tested the growth ability of 44 isolates at 37℃, 45% of the isolates were able to grow. These results show that pet dogs could carry fungi having a potentiality of affecting on human health.
Phenylalanine ammonia-lyase (PAL) from the maize pathogen Ustilago maydis was analysed immunologically to obtain insights into the structural relationships between plant PAL and fungal PAL and between PAL and histidine ammonia-lyase (HAL). Cross-reactivity was found among all the PAL proteins from different species tested, using antibodies raised against both plant and fungal PALs. Both anti-Alfalfa and anti-popular PAL antibodies strongly recognized plant PALs but only weakly recognized fungal PALs. Antibodies raised against U. maydis PAL only weakly recognized the Rhodotorula glutinis yeast PAL. The anti-U. maydis PAL antibodies showed low affinity for the plant PALs but they bound strongly to Pseudomonas bacterial HAL. Significant cross-reactivity between the two plant PAL antibodies and the bacterial HAL was also observed. Both the anti-Ustilago PAL and the anti-poplar PAL antibodies displayed similar enzyme inhibition patterns, including moderate inhibition of bacterial HAL activity. However, the bacterial HAL antibody inhibited only Ustilago PAL. The PAL and HAL antibodies tested showed no inhibition against yeast PAL. This is first report on the immunological relationships between PAL and HAL.
Mold contamination is one of the detrimental factors affecting sawdust media-based shiitake cultivation in greenhouses. During mold monitoring of indoor air of greenhouses, several fungi were isolated. Among them, Aspergillus pulverulentus and Cosmospora butyri were found to be new in Korea and Lecanicillium psalliotae and L. antillanum were known in Korea without taxonomic validation. In this study the morphological characteristics and phylogenetic analysis based on the internal transcribed spacer (ITS) rDNA region or β-tubulin gene of the four identified species were described.
