Keluhan Musculockeletal Disorders (MSDs) merupakan salah satu permasalahan yang sering terjadi pada pekerja yang dapat mengganggu produktifitas pekerja. Salah satu pekerjaan sektor informal yang memiliki risiko kesehatan cukup tinggi adalah penjahit. Desain kerja dan postur tubuh dalam menjait berisiko untuk menimbulkan MSDs. Penelitian ini bertujuan ntuk menganalisis hubungan faktor pekerjaan dengan musculoskeletal disorders (MSDs) pada penjahit. Jenis penelitian adalah observasional analitik dengan desain penelitian cross-sectional dengan menggunakan data primer. Sample penelitian sebanyak 97 penjahit yang diambil dengan teknik simple random sampling. Instrumen penelitian menggunakan kuisioner The Rapid Upper Limb Assesment (RULA) dan Nordic Body Map Questionair (NBM) yang diisi secara Tanya jawab langsung atau wawancara.Uji validitas NBM berkisar antara 0,501(min.) sampai 0,823(maks.) dan indekas reliabilitas Cronbach’s alpha sebesar 0,726. Dimana NMQ memenuhi kriteria sebagai alat penilaian yang andal dengan konsistensi internal tinggi dan validitas konstruk sehingga cocok digunakan untuk menilai gangguan musculoskeletal yang terjadi di Indonesia. Analisis data dengan menghitung p-value untuk melihat ada tidaknya hubungan dan PR untuk mengetahui kekuatan hubungan. Hasil: Terdapat variable yang berhubungan dengan keluhan MSDs adalah usia (PR = 3,16), masa kerja (PR = 1,82) dengan MSDs pada penjahit.
