Accumulating evidence indicates that epithelial splicing regulatory protein 1 (ESRP1) can inhibit the epithelial-to-mesenchymal transition (EMT), thus playing a central role in regulating the metastatic progression of tumors. However, it is still not clear whether ESRP1 directly influences the cell cycle, or what the possible underlying molecular mechanisms are. In this study, we showed that ESRP1 protein levels were significantly correlated with the Ki-67 proliferative index (r = −0.521; p < 0.01), and that ESRP1 overexpression can significantly inhibit cervical carcinoma cell proliferation and induced G1-phase arrest by downregulating cyclin A2 expression. Importantly, ESRP1 can bind to GGUGGU sequence in the 3′UTR of the cyclin A2 mRNA, and ESRP1 overexpression significantly decreases the stability of the cyclin A2 mRNA. In addition, our experimental results confirm that ESRP1 overexpression results in enhanced CDC20 expression, which is known to be responsible for cyclin A2 degradation. This study provides the first evidence that ESRP1 overexpression induces G1-phase cell cycle arrest via reducing the stability of the cyclin A2 mRNA, and inhibits cervical carcinoma cell proliferation. The findings suggest that the ESRP1/cyclin A2 regulatory axis may be essential as a regulator of cell proliferation, and may thus represent an attractive target for cervical cancer prevention and treatment.

Accumulating evidence indicates that hematogenous metastasis is facilitated by tumor cell-leukocyte emboli formation, and L-selectin plays a major role in the process. Several independent studies have indicated that tumor metastasis can be inhibited by chemically modified heparin with low anticoagulant activity in the different tumor models. In the present study, we demonstrated that chemically modified nonanticoagulation heparin derivate (periodate-oxidized, borohydride-reduced heparin [RO-heparin]) can inhibit the binding of L-selectin to HO-8910 cells, block the adhering of HO-8910 to Chinese hamster ovary cells expressing a transfected human L-selectin complementary DNA, and affect the interactions of neutrophils with HO-8910 cells. Flow cytometric analysis with the heparan sulfate-specific monoclonal antibody revealed that HO-8910 cells express heparan sulfate-like proteoglycans. Furthermore, heparinase treatment impaired L-selectin binding, indicating that heparan sulfate-like proteoglycans on the tumor cell surface are implicated in the binding of L-selectin to HO-8910 cells. These findings suggest that RO-heparin with low anticoagulant activities may have potential value as therapeutic agents that block L-selectin-mediated cell adhesion and prevent tumor metastasis.

The transcription factor Wilms’ tumor 1 (WT1) is involved in development, tissue homeostasis, and disease. However, the exact roles and the mechanisms of WT1 in renal carcinoma are not well understood. Therefore, in this study, we evaluated the ability of WT1 to block proliferation in renal carcinoma cells in vitro. Experimental analysis showed that WT1 overexpression inhibited the proliferation of renal carcinoma A498 cells and promoted arrest at the G2/M checkpoint. RNA-Seq identified differentially expressed genes, including IL-24, related to both the cell proliferation and the cell cycle. WT1 overexpression upregulated IL-24 expression, and IL-24 overexpression induced G2/M arrest. ChIP-Seq identified JUN as a direct target of WT1 in A498 cells, in which positive regulation was shown by RT-qPCR. It has been shown that the transcription factor JUN can regulate IL-24 expression, and therefore, we hypothesize that WT1 might regulate the IL-24 through JUN. Furthermore, analysis based on TCGA datasets showed that the expression of WT1-regulated genes, including TXNIP and GADD45A, was significantly correlated with the stage and histological grade of tumors, with high levels linked to favorable prognoses. Our results demonstrated that the overexpression of WT1 upregulates IL-24, leading to G2/M checkpoint arrest to reduce proliferation. These results indicate that regulation of IL-24 by WT1 inhibits proliferation and may represent a potential target for treating renal carcinoma.

With the continuous enhancement of exploration and development in the Ordos Basin, in-depth research has been carried out on the petrological and reservoir characteristics of Chang 2 reservoir in Area A, which provides a geological basis for the efficient development of oil reservoirs. Comprehensive use of reservoir sandstone thin section identification casting analysis, mercury intrusion analysis, logging analysis and other methods to systematically study the petrological characteristics, pore characteristics and reservoir physical properties of Chang 2 reservoir in Area A. The results show that the reservoirs in the study area are dominated by fine-grained sandstones, with low component maturity and high structural maturity. They are all medium-low porosity, low-permeability and ultra-low permeability reservoirs. Primary intergranular pores and residual intergranular pores are developed, the reservoir drainage pressure is low, which is good-medium, and the mercury removal efficiency is high, indicating that the reservoir has good storage performance and seepage properties.
