-Addition of CaCl 2 to skim milk, from 0 to 13.5 mmol·kg -1 , leads to significant physicochemical changes in milk salts equilibrium. Indeed, acidification, insolubilizations of calcium, inorganic phosphate and citrate are observed. Calculation of ion equilibria in the resulting milk ultrafiltrates indicates that the observed ion migrations, controlled by solubilities of calciumphosphate and calcium-citrate, do not cause change in ion activity products of dicalcium-phosphate (calculated as CaHPO 4 ·2H 2 O and Ca(HPO 4 ) 0.7 (PO 4 ) 0.2 ). In the same way, the acidification of calcium-supplemented milks in the range of pH 6.7-3.5 shows a solubilization of calciumphosphate similar to non-enriched milk. So whatever the added calcium concentration, the behaviour of colloidal calcium-phosphate seems to be unchanged. However, at room temperature, addition of calcium to milk originates a decrease in soluble casein concentration and tightly bound water of ultracentrifuged pellets. The consequences are (i) an increase in milk protein hydrophobicity; (ii) a decrease in zeta potential of casein micelles, without modification of their hydrodynamic diameter; and (iii) an increase in milk lightness and turbidity. Furthermore, calcium supplementation of milk seems to lead to a proteinic reorganization of micellar casein, responsible for an increase in micellar density. L'addition de CaCl 2 à du lait écrémé, pour des concentrations allant de 0 à 13,5 mmol·kg -1 , induit d'importants changements physicochimiques du lait. En effet, une acidification, une insolubilisation du calcium, du phosphate inorganique et du citrate sont observées. Le calcul des équilibres ioniques dans les ultrafiltrats des différents laits montre que les flux ioniques observés, contrôlés principalement par les solubilités du phosphate et du citrate de calcium, ne modifient pas les produits d'activité ionique du phosphate dicalcique (calculés pour CaHPO 4 ,2H 2 O et Ca(HPO 4 ) 0,7 (PO 4 ) 0,2 ). Summary -pH-lnduced physicochemical changes and rennet coagulation times of casein micelle suspensions have been studied with native phosphocaseinate powder dissolved in water, 0.1 mol L-1 NaCI and milk ultrafiltrate. An increase in ionic strength between water and NaClled to higher diffusible calcium and phosphorus, to greater micelle voluminosity and to greater case in and minerai solubilization during acidification. Dissolution of the phosphocaseinate in ultrafiltrate resulted in physicochemical properties very close to milk. The acidification process can be divided into five successive steps. Relations between physicochemical changes are discussed, together with the reduction of rennet coagulation time with pH and ionic strength reduction.
-Le phosphocaséinate natif (PPCN) était séparé du lait cru par microfiltration tangentielle sur membrane (diamètre des pores: 0,2 l1m) suivie d'une purification par diafiltration avec de l'eau. Le suivi analytique des liquides obtenus de part et d'autre de la membrane montrait une perméation conforme aux lois de la filtration des protéines majeures du lactosérum (j3-lactoglobuline et a-lactalbumine), du lactose et des sels minéraux solubles et au contraire, une rétention élevée des composants solubles à haut poids moléculaire ainsi que des protéoses-peptones. Le rétentat final était séché par atomisation. Ses teneurs en matière azotée (N x 6,38) et en cendres étaient respectivement de 89% et de 9% par rapport à la substance sèche. L'aptitude à la coagulation par la présure, de la solution reconstituée a été comparée à celle du lait cru. Le temps de coagulation est réduit de 53%; la cinétique d'organisation du. gel (appréciée par le K2Q) et la fermeté finale (appréciée à 30 min) sont également accrues de plus de 50%. Native phosphocaseinate (PPCN) was separated from raw milk by tangential membrane microfiltration (pore diameter: 0.2 um) followed by purification through water diafiltration. Analytical survey of both tiquids issued by membrane treatment showed permeation rates matching mathematical laws for the main whey proteins (fJ-lactoglobulin and a-Iactalbumin), lactose and soluble mineraI salts. On the contrary, a high retention of high molecular weight soluble components and proteose-peptone fraction was observed. Finaldiafiltratedretentate was spray-dried. Its protein (N x 6.38) and ash contents in total solids were respectively 89% and 9%. Rennet coagulating ability of PPCN reconstituted solution was compared to that of raw milk. Coagulation time was reduced by 53%; gel development kinetics (estimated by Krol and final firmness (estimated at 30 min) were increased by more than 50%. When a severe heat treatment (100 oC-5 min) was applied, milk lost its ability to coagulate alter rennet action; PPCN solution did not, but ifs coagulation time was increased and the curd was slightly weaker.
(Reçu le 5 mai 1994; accepté le 30 juin 1994) Résumé -La microfiltration tangentielle sur membrane 0,14 um de lait épuré par le procédé Bactocatch suivie d'une diafiltration, préalablement à sa déshydratation par atomisation, permet l'obtention d'une poudre de phosphocaséinate natif à haute teneur en matière azotée totale (90,2 g pour 100 g de matière sèche) et de bonne qualité bactériologique (population bactérienne < 5 000 ufclg). La réalisation d'un concentré enrichi en caséine micellaire entraîne des modifications profondes des caractéristiques physiques des poudres de phosphocaséinate natif. L'indice de solubilité est de 73,7% à 24°C, diminuant ainsi la qualité intrinsèque de ces poudres. Dans la présente étude, nous avons essayé de comprendre au mieux la cause de cette formation d'insoluble. Des modifications biochimiques des concentrés (diminution du pH, augmentation de la force ionique, ajout de lactose) et physiques des poudres (diminution de la taille des grains) ont permis d'améliorer sensiblement J'indice de solubilité jusqu'à 92,6% à 24°C. L'insoluble observé lors de la reconstitution de concentré à partir de poudre de phosphocaséinate natif ne serait pas lié à une dénaturation thermique, mais à une diminution de la vitesse de transfert de J'eau de reconstitution. L'ajout de lactose ou "augmentation de la surface de contact entre le grain et l'eau de reconstitution a permis d'augmenter cette vitesse de transfert en améliorant ainsi l'indice de solubilité des poudres de phosphocaséinate natif.
