This paper describes an iterative algorithm for high-dimensional linear inverse problems, which is regularized by a differentiable discrete approximation of the total variation (TV) penalty. The algorithm is an interlaced iterative method based on optimization transfer with a separable quadratic surrogate for the TV penalty. The surrogate cost function is optimized using the block iterative regularized algebraic reconstruction technique (RSART). A proof of convergence is given and convergence is illustrated by numerical experiments with simulated parallel-beam computerized tomography (CT) data. The proposed method provides a block-iterative and convergent, hence efficient and reliable, algorithm to investigate the effects of TV regularization in applications such as CT.

Mutations in GUCY2D are associated with recessive Leber congenital amaurosis-1 (LCA1). GUCY2D encodes photoreceptor-specific, retinal guanylate cyclase-1 (RetGC1). Reports of retinal degeneration in LCA1 are conflicting; some describe no obvious degeneration and others report loss of both rods and cones. Proof of concept studies in models representing the spectrum of phenotypes is warranted. We have previously demonstrated adeno-associated virus (AAV)-mediated RetGC1 is therapeutic in GC1ko mice, a model exhibiting loss of cones only. The purpose of this study was to characterize AAV-mediated gene therapy in the RetGC1/RetGC2 double knockout (GCdko) mouse, a model lacking rod and cone function and exhibiting progressive loss of both photoreceptor subclasses. Use of this model also allowed for the evaluation of the functional efficiency of transgenic RetGC1 isozyme. Subretinal delivery of AAV8(Y733F) vector containing the human rhodopsin kinase (hGRK1) promoter driving murine Gucy2e was performed in GCdko mice at various postnatal time points. Treatment resulted in restoration of rod and cone function at all treatment ages and preservation of retinal structure in GCdko mice treated as late as 7 weeks of age. Functional gains and structural preservation were stable for at least 1 year. Treatment also conferred cortical-and subcortical-based visually-guided behavior. Functional efficiency of transgenic RetGC1 was indistinguishable from that of endogenous isozyme in congenic wild-type (WT) mice. This study clearly demonstrates AAV-mediated RetGC1 expression restores function to and preserves structure of rod and cone photoreceptors in a degenerative model of retinal guanylate cyclase deficiency, further supporting development of an AAV-based vector for treatment of LCA1.

The development of highly active, environmentally friendly, and long-term stable oxygen evolving catalysts at low costs is critical for efficient and scalable H production from water splitting. Here, we report a new and facile one-step electrodeposition of nanocrystalline spinel-type ZnCoO films from an alkaline Zn-Co-tartrate solution. The electrodeposited ZnCoO electrode could be directly used as the anode for the water electrolysis without any post treatment. The ZnCoO film shows a low and stable overpotential of ∼0.33 V at 10 mA cm (and ∼0.35 V at 20 mA cm) for over 10 h and a Tafel slope of ∼39 mV dec toward the oxygen evolution reaction (OER) in 1 M NaOH, comparable to the best performance of the nonprecious OER catalysts reported for alkaline media. The enhanced OER activity of ZnCoO compared to CoO could be attributed to the surface structural modification and higher density of the accessible active Co sites induced by the incorporation of Zn. The electrodeposition method in this paper could also be used to synthesize other binary and ternary metal oxide based catalytic electrodes for reactions such as the OER and oxygen reduction reaction (ORR).

BackgroundWater deficit severely reduces apple growth and production, is detrimental to fruit quality and size. This problem is exacerbated as global warming is implicated in producing more severe drought stress. Thus water-efficiency has becomes the major target for apple breeding. A desired apple tree can absorb and transport water efficiently, which not only confers improved drought tolerance, but also guarantees fruit size for higher income returns. Aquaporins, as water channels, control water transportation across membranes and can regulate water flow by changing their amount and activity. The exploration of molecular mechanism of water efficiency and the gene wealth will pave a way for molecular breeding of drought tolerant apple tree.ResultsIn the current study, we screened out a drought inducible aquaporin gene MdPIP1;3, which specifically enhanced its expression during fruit expansion in ‘Fuji’ apple (Malus domestica Borkh. cv. Red Fuji). It localized on plasma membranes and belonged to PIP1 subfamily. The tolerance to drought stress enhanced in transgenic tomato plants ectopically expressing MdPIP1;3, showing that the rate of losing water in isolated transgenic leaves was slower than wild type, and stomata of transgenic plants closed sensitively to respond to drought compared with wild type. Besides, length and diameter of transgenic tomato fruits increased faster than wild type, and in final, fruit sizes and fresh weights of transgenic tomatoes were bigger than wild type. Specially, in cell levels, fruit cell size from transgenic tomatoes was larger than wild type, showing that cell number per mm2 in transgenic fruits was less than wild type.ConclusionsAltogether, ectopically expressing MdPIP1;3 enhanced drought tolerance of transgenic tomatoes partially via reduced water loss controlled by stomata closure in leaves. In addition, the transgenic tomato fruits are larger and heavier with larger cells via more efficient water transportation across membranes.
