Severe acute respiratory syndrome coronavirus 2 was isolated from feces of a patient in China with coronavirus disease who died. Confirmation of infectious virus in feces affirms the potential for fecal–oral or fecal–respiratory transmission and warrants further study.
SARS-CoV-2 caused a major outbreak of severe pneumonia (COVID-19) in humans. Viral RNA was detected in multiple organs in COVID-19 patients. However, infectious SARS-CoV-2 was only isolated from respiratory specimens. Here, infectious SARS-CoV-2 was successfully isolated from urine of a COVID-19 patient. The virus isolated could infect new susceptible cells and was recognized by its' own patient sera. Appropriate precautions should be taken to avoid transmission from urine.
Despite development in the understanding of the pathogenesis of acute lung injury (ALI)/acute respiratory distress syndrome (ARDS), the underlying mechanism still needs to be elucidated. Apart from leukocytes and endothelial cells, macrophages are also essential for the process of the inflammatory response in ALI/ARDS. Notably, macrophages play a dual role of proinflammation and anti-inflammation based on the microenvironment in different pathological stages. In the acute phase of ALI/ARDS, resident alveolar macrophages, typically expressing the alternatively activated phenotype (M2), shift into the classically activated phenotype (M1) and release various potent proinflammatory mediators. In the later phase, the M1 phenotype of activated resident and recruited macrophages shifts back to the M2 phenotype for eliminating apoptotic cells and participating in fibrosis. In this review, we summarize the main subsets of macrophages and the associated signaling pathways in three different pathological phases of ALI/ARDS. According to the current literature, regulating the function of macrophages and monocytes might be a promising therapeutic strategy against ALI/ARDS.
Mitofusin 1 (MFN1) mediates mitochondrial fusion, but the mechanisms involved are unclear. Qi et al. present the crystal structures of a minimal GTPase domain of human MFN1, which suggest that MFN1 tethers apposing membranes through nucleotide-dependent dimerization.
Fusion of the outer mitochondrial membrane is mediated by the dynamin-like GTPase mitofusin (MFN). Here, we determined the structure of the minimal GTPase domain (MGD) of human MFN1 in complex with GDP-BeF. The MGD folds into a canonical GTPase fold with an associating four-helix bundle, HB1, and forms a dimer. A potassium ion in the catalytic core engages GDP and BeF (GDP-BeF). Enzymatic analysis has confirmed that efficient GTP hydrolysis by MFN1 requires potassium. Compared to previously reported MGD structures, the HB1 structure undergoes a major conformational change relative to the GTPase domains, as they move from pointing in opposite directions to point in the same direction, suggesting that a swing of the four-helix bundle can pull tethered membranes closer to achieve fusion. The proposed model is supported by results from in vitro biochemical assays and mitochondria morphology rescue assays in MFN1-deleted cells. These findings offer an explanation for how Charcot-Marie-Tooth neuropathy type 2 A (CMT2A)-causing mutations compromise MFN-mediated fusion.
Glacial-interglacial changes in the distribution of C 3 /C 4 vegetation on the Chinese Loess Plateau have been related to East Asian summer monsoon intensity and position, and could provide insights into future changes caused by global warming. Here, we present δ 13 C records of bulk organic matter since the Last Glacial Maximum (LGM) from 21 loess sections across the Loess Plateau. The δ 13 C values (range: -25‰ to -16‰) increased gradually both from the LGM to the mid-Holocene in each section and from northwest to southeast in each time interval. During the LGM, C 4 biomass increased from <5% in the northwest to 10-20% in the southeast, while during the mid-Holocene C 4 vegetation increased throughout the Plateau, with estimated biomass increasing from 10% to 20% in the northwest to >40% in the southeast. The spatial pattern of C 4 biomass in both the LGM and the mid-Holocene closely resembles that of modern warm-season precipitation, and thus can serve as a robust analog for the contemporary East Asian summer monsoon rain belt. Using the 10-20% isolines for C 4 biomass in the cold LGM as a reference, we derived a minimum 300-km northwestward migration of the monsoon rain belt for the warm Holocene. Our results strongly support the prediction that Earth's thermal equator will move northward in a warmer world. The southward displacement of the monsoon rain belt and the drying trend observed during the last few decades in northern China will soon reverse as global warming continues.
