Drought is one of the most important environmental constraints limiting plant growth and agricultural productivity. To understand the underlying mechanism of drought tolerance and to identify genes for improving this important trait, we conducted a gain-of-function genetic screen for improved drought tolerance in Arabidopsis thaliana. One mutant with improved drought tolerance was isolated and designated as enhanced drought tolerance1. The mutant has a more extensive root system than the wild type, with deeper roots and more lateral roots, and shows a reduced leaf stomatal density. The mutant had higher levels of abscisic acid and Pro than the wild type and demonstrated an increased resistance to oxidative stress and high levels of superoxide dismutase. Molecular genetic analysis and recapitulation experiments showed that the enhanced drought tolerance is caused by the activated expression of a T-DNA tagged gene that encodes a putative homeodomain-START transcription factor. Moreover, overexpressing the cDNA of the transcription factor in transgenic tobacco also conferred drought tolerance associated with improved root architecture and reduced leaf stomatal density. Therefore, we have revealed functions of the homeodomain-START factor that were gained upon altering its expression pattern by activation tagging and provide a key regulator that may be used to improve drought tolerance in plants.

Enhancing drought tolerance without yield decrease has been a great challenge in crop improvement. Here, we report the Arabidopsis (Arabidopsis thaliana) homodomain-leucine zipper transcription factor Enhanced Drought Tolerance/HOMEODOMAIN GLABROUS11 (EDT1/HDG11) was able to confer drought tolerance and increase grain yield in transgenic rice (Oryza sativa) plants. The improved drought tolerance was associated with a more extensive root system, reduced stomatal density, and higher water use efficiency. The transgenic rice plants also had higher levels of abscisic acid, proline, soluble sugar, and reactive oxygen species-scavenging enzyme activities during stress treatments. The increased grain yield of the transgenic rice was contributed by improved seed setting, larger panicle, and more tillers as well as increased photosynthetic capacity. Digital gene expression analysis indicated that AtEDT1/HDG11 had a significant influence on gene expression profile in rice, which was consistent with the observed phenotypes of transgenic rice plants. Our study shows that AtEDT1/HDG11 can improve both stress tolerance and grain yield in rice, demonstrating the efficacy of AtEDT1/HDG11 in crop improvement.

Many stress responsive genes have been reported with an effect on improving stress resistance in model plants under greenhouse conditions. Towards identification of genes for drought resistance breeding, seven well documented genes (CBF3, SOS2, NCED2, NPK1, LOS5, ZAT10, and NHX1) in stress resistance were selected in this study and transformed into rice cultivar Zhonghua 11 under the control of constitutive promoter Actin1 and stress-inducible promoter of a rice HVA22 homolog, and transgenic rice were tested for drought resistance under field conditions. A total of 1598 independent transgenic T0 plants were generated. The percentages of single copy and expression of the transgenes were 36.7% and 57.6%, respectively. For each gene construct, 30 T1 families with expression of transgene were selected for drought resistance testing at the reproductive stage in field, and 10 of them were tested in PVC pipes with a defined stress protocol at the same stage. Relative yield and relative spikelet fertility were used as two major criteria to evaluate drought resistance performance because significantly decreased yield was observed in the T1 generation. Transgenic families of eight constructs (HVA22P:CBF3, HVA22P:NPK1, Actin1:LOS5, HVA22P:LOS5, Actin1:ZAT10, HVA22P:ZAT10, Actin1:NHX1, and HVA22P:NHX1) showed significantly higher RY than wild-type (WT) under both drought stress field and PVC tube conditions. Transgenic families of 9 constructs (HVA22P:SOS2 and CBF3, LOS5, ZAT10, and NHX1 by both promoters) showed significantly higher relative spikelet fertility than WT in the field or PVC pipes. In the field drought resistance testing of T2 families derived from the T1 families with relatively lower yield decrease, transgenic families of seven constructs (HVA22P:CBF3, Actin1:NPK1, HVA22P:NPK1, Actin1:LOS5, HVA22P:LOS5, Actin1:ZAT10, and HVA22P:ZAT10) showed significantly higher yield per plant than WT, and families of nine constructs (Actin1:CBF3, HVA22P:CBF3, HVA22P:SOS2, HVA22P:NPK1, Actin1:LOS5, HVA22P:LOS5, Actin1:ZAT10, HVA22P:ZAT10, and Actin1:NHX1) had higher spikelet fertility than WT. In general, LOS5 and ZAT10 showed relatively better effect than the other five genes in improving drought resistance of transgenic rice under field conditions. The results and experience obtained from this study could be a useful reference for drought resistance engineering in rice.

Improvement of drought and salt tolerance of crops with transgenic approach is an effective strategy to meet the demand of the ever-growing world population. Arabidopsis ENHANCED DROUGHT TOLERANCE1/HOMEODOMAIN GLABROUS11 (AtEDT1/HDG11), a homeodomain-START transcription factor, has been demonstrated to significantly improve drought tolerance in Arabidopsis, tobacco, tall fescue and rice. Here we report that AtHDG11 also confers drought and salt tolerance in upland cotton (Gossypium hirsutum) and woody plant poplar (Populus tomentosa Carr.). Our results showed that both the transgenic cotton and poplar exhibited significantly enhanced tolerance to drought and salt stress with well-developed root system. In the leaves of the transgenic cotton plants, proline content, soluble sugar content and activities of reactive oxygen species-scavenging enzymes were significantly increased after drought and salt stress compared with wild type. Leaf stomatal density was significantly reduced, whereas stomatal and leaf epidermal cell size were significantly increased in both the transgenic cotton and poplar plants. More importantly, the transgenic cotton showed significantly improved drought tolerance and better agronomic performance with higher cotton yield in the field both under normal and drought conditions. These results demonstrate that AtHDG11 is not only a promising candidate for crops improvement but also for woody plants.

The out-of-time-ordered correlators (OTOC) have been established as a fundamental concept for quantifying quantum information scrambling and diagnosing quantum chaotic behavior. Recently, it was theoretically proposed that the OTOC can be used as an order parameter to dynamically detect both equilibrium quantum phase transitions (EQPTs) and dynamical quantum phase transitions (DQPTs) in one-dimensional many-body systems. Here we report the first experimental observation of EQPTs and DQPTs in a quantum spin chain via quench dynamics of OTOC on a nuclear magnetic resonance quantum simulator. We observe that the quench dynamics of both the order parameter and the two-body correlation function cannot detect the DQPTs, but the OTOC can unambiguously detect the DQPTs. Moreover, we demonstrate that the long-time average value of the OTOC in quantum quench signals the equilibrium quantum critical point and ordered quantum phases, thus one can measure the EQPTs from the non-equilibrium quantum quench dynamics. Our experiment paves a way for experimentally investigating DQPTs through OTOCs and for studying the EQPTs through the non-equilibrium quantum quench dynamics with quantum simulators.
