Das Studium des Stoffwechsels der Steroide beschränkte sich bisher auf den Nachweis, die Isolierung und Identifizierung von Metaboliten. Für die Beurteilung des Steroidhaushaltes ist jedoch die Erfassung der meist unwirksamen Umwandlungsprodukte von geringerer Bedeutung als die Kenntnis der Umwandlungsrate. Diese ist aber weitgehend abhängig von der Stoff Wechselaktivität der Leber, der damit, wenn nicht die Funktion eines allgemeinen Regulators im Hormonhaushalt, so doch wenigstens die eines Gegenspielers zu den endokrinen Drüsen zugeschrieben werden muß. Denn die Hormonkonzentration, die zu einem gegebenen Zeitpunkt im Blut vorliegt, wird durch die biogenetische und sekretorische Aktivität der endokrinen Drüsen ebenso bestimmt wie durch die Stoffwechselaktivität der Leber.Die Steroidstoffwechselaktivität der Leber ist abhängig von endogenen (Schilddrüsenfunktion 1 , Sexualstatus 2 ) und exogenen (Stressbedingungen 3 , Antirheumatica 4 ) Faktoren, die somit Einfluß auf das endokrine Gleichgewicht nehmen können.Zur intensiveren Bearbeitung solcher und ähnlicher Beziehungen haben wir deshalb in Perfusionsversuchen mit Cortison, Prednison und 9a-Fluor-hydrocortison an Lebern normaler Ratten einige Stoffwechselgrößen zu erfassen gesucht: Ausmaß des Gesamtumsatzes, die Geschwindigkeit, mit der einzelne Reaktionen am Steroidmolekül ablaufen und die zeitliche Folge des Auftretens von Stoff Wechselprodukten. 1
