Indonesia termasuk negara maritim yang memiliki perairan laut yang cukup luas. Dengan luasnya perairan laut tersebut menyebabkan kekayaan flora dan fauna di dalamnya cukup tinggi dan beraneka ragam. Keanekaragaman hewan laut diantaranya berasal dari filum Echinodermata. Secara umum, Echinodermata memiliki ciri morfologi yaitu bentuk tubuhnya simetri radial, tanpa kepala, tidak bersegmen, dan permukaan kulitnya berduri. Penelitian ini bertujuan untuk melakukan identifikasi dan inventarisasi beberapa jenis anggota Echinodermata yang ditemukan di perairan Pulau Lima dan Pulau Kambing yang terletak di Kecamatan Kasemen, Kota Serang, Banten. Metode pengambilan sampel yang digunakan pada penelitian ini yaitu random sampling, dimana sampel diambil secara acak dengan melakukan penjelajahan di perairan Pulau Lima dan Pulau Kambing. Berdasarkan hasil penelitian diperoleh tiga spesies anggota Echinodermata, yaitu Holothuria sp., Archaster sp., dan Macrophiotix sp.
Bacterial cellulose (BC) merupakan polisakarida polimer yang dihasilkan oleh bakteri penghasil selulosa. BC dikenal oleh masyarakat dengan sebutan ‘Nata’. Acetobacter xylinum merupakan salah satu bakteri penghasil selulosa dalam jumlah besar. Kebutuhan bakteri dalam membentuk selulosa membutuhkan unsur-unsur makro, seperti C, H, O, N, S, P, dan Se, yang berasal dari berbagai substrat, seperti air kelapa, air beras, dan tomat. Kebutuhan nitrogen dalam pembuatan BC secara komersial umumnya dipenuhi dengan penambahan ZA. Penggunaan ZA umum digunakan masyarakat karena harganya murah dan mudah diperoleh. Akan tetapi, apabila kadarnya melebihi 0,5% dari bahan total, dapat menimbulkan dampak negatif bagi kesehatan tubuh manusia. Oleh karena itu diperlukan alternatif sumber nitrogen lain seperti ekstrak kecambah. Penelitian ini bertujuan untuk mengetahui karakter fisik dengan perbedaan perlakuan sumber nitrogen dari ZA dan ekstrak kecambah pada berbagai variasi dan kombinasi substrat, diantaranya air kelapa, air beras, tomat, air kelapa-tomat, air kelapa-beras, dan air beras-tomat. Penelitian ini menggunakan metode eksperimen pada masing-masing substrat dan hasil yang diperoleh dianalisis secara deskriptif kualitatif. Formula pembuatan BC yaitu 50 mL substrat, 5 gram gula, 2 gram ZA, 2 gram ekstrak kecambah, 2 gram asam asetat glasial, dan 2 mL starter (A. xylinum). Hasil penelitian menunjukkan bahwa penggunaan ekstrak kecambah sebagai alternatif ZA cenderung efektif dalam pembentukan BC, ditunjukkan dengan ketebalan tertinggi 2,5 cm dan rendemen tertinggi 80%. Akan tetapi, kadar serat ZA lebih unggul, yaitu sebesar 3%, sedangkan kadar air pada kedua perlakuan berkisar 97-99%.
Cellulose is one of the most abundant polymers on the earth's surface and is often used as the main raw material in various industries. Bacterial Cellulose (BC) is an organic compound produced by certain types of bacteria with various variations and combinations of substrates, including coconut water, rice soaking water, tomatoes, and a mixture of tomato-rice water, coconut-tomato water, and coconut water-rice water. This study aims to determine the optimization of BC formation in various variations and combinations of substrates with organoleptic and physical tests on each sample. This study used experimental methods on each substrate and analyzed descriptively qualitatively. The formula for making BC was 50 mL of each substrate, 5 grams of sugar, 2 grams of ZA (ammonium sulfate), 2 grams of glacial acetic acid, and 2 mL of starter (Acetobacter xylinum). The results of this study obtained organoleptic BC by 90% of panelists liked it, and 10% of panelists disliked it. Meanwhile, the best optimization of BC production obtained the highest thickness and yield on the coconut-rice water substrate treatment of 2.3 cm and 80% yield, the highest water content on the coconut water, coconut-tomato water, and coconut-rice water substrate treatments, respectively, each of 97% and the highest fiber content in rice water, tomato water, and tomato-rice water substrates were 3% each.
