Liquorice (Glycyrrhiza uralensis) is an important medicinal plant for which there is a huge market demand. It has been reported that arbuscular mycorrhizal (AM) symbiosis and drought stress can stimulate the accumulation of the active ingredients, glycyrrhizin and liquiritin, in liquorice plants, but the potential interactions of AM symbiosis and drought stress remain largely unknown. In the present work, we investigated mycorrhizal effects on plant growth and accumulation of glycyrrhizin and liquiritin in liquorice plants under different water regimes. The results indicated that AM plants generally exhibited better growth and physiological status including stomatal conductance, photosynthesis rate, and water use efficiency compared with non-AM plants. AM inoculation up-regulated the expression of an aquaporin gene PIP and decreased root abscisic acid (ABA) concentrations under drought stress. In general, AM plants displayed lower root carbon (C) and nitrogen (N) concentrations, higher phosphorus (P) concentrations, and therefore, lower C:P and N:P ratios but higher C:N ratio than non-AM plants. On the other hand, AM inoculation increased root glycyrrhizin and liquiritin concentrations, and the mycorrhizal effects were more pronounced under moderate drought stress than under well-watered condition or severe drought stress for glycyrrhizin accumulation. The accumulation of glycyrrhizin and liquiritin in AM plants was consistent with the C:N ratio changes in support of the carbon-nutrient balance hypothesis. Moreover, the glycyrrhizin accumulation was positively correlated with the expression of glycyrrhizin biosynthesis genes SQS1, β-AS, CYP88D6, and CYP72A154. By contrast, no significant interaction of AM inoculation with water treatment was observed for liquiritin accumulation, while we similarly observed a positive correlation between liquiritin accumulation and the expression of a liquiritin biosynthesis gene CHS. These results suggested that AM inoculation in combination with proper water management potentially could improve glycyrrhizin and liquiritin accumulation in liquorice roots and may be practiced to promote liquorice cultivation.
Arbuscular mycorrhizal (AM) fungi, as root symbionts of most terrestrial plants, improve plant growth and fitness. In addition to the improved plant nutritional status, the physiological changes that trigger metabolic changes in the root via AM fungi can also increase the host ability to overcome biotic and abiotic stresses. Plant viruses are one of the important limiting factors for the commercial cultivation of various crops. The effect of AM fungi on viral infection is variable, and considerable attention is focused on shoot virus infection. This review provides an overview of the potential of AM fungi as bioprotection agents against viral diseases and emphasizes the complex nature of plant–fungus–virus interactions. Several mechanisms, including modulated plant tolerance, manipulation of induced systemic resistance (ISR), and altered vector pressure are involved in such interactions. We propose that using “omics” tools will provide detailed insights into the complex mechanisms underlying mycorrhizal-mediated plant immunity.
Background Water shortage can limit plant growth, which can be ameliorated by arbuscular mycorrhizal (AM) symbiosis through physiological and metabolic regulations. Deciphering which physiological and metabolic processes are central for AM-mediated regulations is essential for applications of mycorrhizal biotechnology in dryland agriculture. Methodology In this study, the influence of AM symbiosis on growth performance, photosynthesis, and organ accumulation of key C and N metabolites were assessed by growing maize (Mo17, Lancaster Sure Crop) seedlings inoculated with or without AM fungus (Rhizophagus irregularis Schenck & Smith BGC AH01) under different water regimes in greenhouse. Results Drought stress reduced shoot growth, while AM symbiosis significantly improved growth performances, with significant changes of photochemical processes and organ concentration of the key metabolites. AM symbiosis increased root levels of the metabolites in ornithine cycle and unsaturation of fatty acids regardless of water conditions. Root putrescine (Put) concentration was higher in AM than non-inoculated (NM) plants under well-watered conditions; the conversion of Put via diamine oxidase to γ-aminobutyric acid (GABA) occurred in roots of AM plants under drought stress. Leaf concentration of Put, the tricarboxylic acids, and soluble sugars significantly increased in AM plants under drought stress, showing higher values compared to that of NM plants. Moreover, photosystem II efficiency and chlorophyll concentration were higher in AM than NM plants regardless of water status. Conclusion Fatty acid- and ornithine cycle-related metabolites along with soluble sugars, Put, and GABA were the key metabolites of AM-mediated regulations in response to drought stress.
Nitrogen (N) deposition and intensified rainfall can strongly affect soil microbial community, but compared with available studies on bacteria, those on soil fungi are quite limited. Here we carried out a field experiment in a mixed deciduous forest of China to study the influences of increased N deposition and rainfall on soil fungi by using quantitative PCR and high-throughput sequencing method. The results demonstrated that (1) N addition significantly increased fungal abundance and alpha diversity (richness, Shannon index and Invsimpson index), changed fungal community composition at OTU level, and marginally increased the relative abundance of Ascomycota and Zygomycota, while water addition showed no remarkable effects on fungal abundance, biodiversity and community composition. (2) N addition significantly increased the richness of saprotrophic fungi and pathogenic fungi, and the relative abundance of saprotrophic fungi, but water addition only slightly increased the abundance of pathogenic fungi. (3) Fungal composition dissimilarity closely correlated with the disparity of soil parameters as a whole. Soil NH 4 + -N exhibited strong positive correlation with the richness of pathogenic fungi and mycorrhizal fungi, while both soil moisture and NH 4 + -Ntightly correlated with soil fungal abundance and alpha diversity indices. We concluded that in this N-limited but non-water-limited forest ecosystem, N deposition posed stronger effects on soil fungi than increased rainfall, partially mediated by changes in soil properties.
Rhizobia and arbuscular mycorrhizal (AM) fungi can potentially alleviate the abiotic stress on the legume Glycyrrhiza (licorice), while the potential benefits these symbiotic microbes offer to their host plant are strongly influenced by environmental factors. A greenhouse pot experiment was conducted to investigate the effects of single and combined inoculation with a rhizobium Mesorhizobium tianshanense Chen and an AM fungus Rhizophagus irregularis Walker & Schuessler on Glycyrrhiza uralensis Fisch. seedling performance under different water regimes. Drought stress inhibited rhizobium nodulation but increased mycorrhizal colonization. Furthermore, co-inoculation of rhizobium and AM fungus favored nodulation under both well-watered and drought stress conditions. Glycyrrhiza seedling growth showed a high mycorrhizal dependency. The seedlings showed a negative growth dependency to rhizobium under well-watered conditions but showed a positive response under drought stress. R. irregularis-inoculated plants showed a much higher stress tolerance index (STI) value than M. tianshanense-inoculated plants. STI value was more pronounced when plants were co-inoculated with R. irregularis and M. tianshanense compared with single-inoculated plants. Plant nitrogen concentration and contents were significantly influenced by inoculation treatments and water regimes. R. irregularis inoculation significantly increased plant shoot and root phosphorus contents. AM fungus inoculation could improve Glycyrrhiza plant–rhizobium symbiosis under drought stress, thereby suggesting that tripartite symbiotic relationships were more effective for promoting plant growth and enhancing drought tolerance.
Nitrogen (N) deposition and precipitation could profoundly influence the structure and function of forest ecosystems. However, conventional studies with understory additions of nitrogen and water largely ignored canopy-associated ecological processes and may have not accurately reflected the natural situations. Additionally, most studies only made sampling at one time point, overlooked temporal dynamics of ecosystem response to environmental changes. Here we carried out a field trial in a mixed deciduous forest of China with canopy addition of N and water for 4 years to investigate the effects of increased N deposition and precipitation on the diversity and community composition of arbuscular mycorrhizal (AM) fungi, the ubiquitous symbiotic fungi for the majority of terrestrial plants. We found that (1) in the 1st year, N addition, water addition and their interactions all exhibited significant influences on AM fungal community composition; (2) in the 2nd year, only water addition significantly reduced AM fungal alpha-diversity (richness and Shannon index); (3) in the next 2 years, both N addition and water addition showed no significant effect on AM fungal community composition or alpha-diversity, with an exception that water addition significantly changed AM fungal community composition in the 4th year; (4) the increment of N or water tended to decrease the abundance and richness of the dominant genus Glomus and favored other AM fungi. (5) soil pH was marginally positively related with AM fungal community composition dissimilarity, soil NH4+-N and N/P showed significant/marginal positive correlation with AM fungal alpha-diversity. We concluded that the effect of increased N deposition and precipitation on AM fungal community composition was time-dependent, mediated by soil factors, and possibly related to the sensitivity and resilience of forest ecosystem to environmental changes.
Soil microorganisms play important roles in nutrient mobilization and uptake of mineral nutrition in plants. Agricultural management, such as soil sterilization, can have adverse effects on plant growth because of the elimination of indigenous microorganisms. Arbuscular mycorrhizal (AM) fungi are one of the most important beneficial soil microorganisms for plant growth. However, whether AM fungi can compensate for the loss of indigenous microbial communities to support plant growth and metabolism is largely unknown. In this study, a pot experiment was conducted to investigate the effects of AM fungi on plant growth and secondary metabolism in sterilized and unsterilized soil. We used liquorice (Glycyrrhiza uralensis Fisch.), an important medicinal plant as the host, which was inoculated with the AM fungus Rhizophagus irregularis or not and grown in unsterilized or sterilized soil. Plant photosynthesis traits, plant growth and nutrition level, concentrations of the secondary metabolites, and expression levels of biosynthesis genes were determined. The results showed that soil sterilization decreased plant growth, photosynthesis, and glycyrrhizin and liquiritin accumulation, and moreover, downregulated the expression of related biosynthesis genes. Inoculation with R. irregularis in sterilized soil offset the loss of indigenous microbial communities, resulting in plant growth and glycyrrhizin and liquiritin concentrations similar to those of plants grown in unsterilized soil. Thus, AM fungi could compensate for the loss of indigenous microbial communities by soil sterilization to support plant growth and secondary metabolism.
