Introdução: Cinamaldeído (CND) é o principal componente ativo do óleo essencial da canela (Cinnamomum sp) e tem sido amplamente utilizado em atividades biológicas e farmacológicas, tendo sido relatado atividade antimicrobiana, antioxidante, antidiabética, dentre outras. Objetivo: Devido os diversos relatos das propriedades farmacológicas do composto, esse foi escolhido para revisão de literatura. A seleção da bibliografia foi obtida a partir de bases de dados (google scholar, NCBI - National Center for Biotechnology Information, PubMed e Scielo - Scientific Electronic Library Online). Discussão: A composição química do CND tem compostos terpenóides que têm poderosa atividade antimicrobiana contra fungos, bactérias Gram-positivas e Gram-negativas tais como Aeromonas hydrophila, Enterococcus faecalis, Clostridium botulinum, Staphylococcus aureus, Escherichia coli O157: H7 e Salmonella enterica serovar Typhimurium. A atividade anti-inflamatória induz a apoptose e inibi a proliferação celular, nas respostas imunes mediadas por monócitos/macrófagos, além de diminuir a produção de óxido nítrico induzido por lipopolissacáridos de um modo dependente da dose. CND ao ser administrado por via oral em ratos diabéticos demonstrou melhora no conteúdo de glicogênio muscular e hepático aumentando a liberação de insulina. Além disso CND estimulou a angiogênese in vivo e in vitro, regulando positivamente o fator de crescimento endotelial vascular. Conclusão: A partir do que foi relatado constatou-se que o CND possui muitas atividades com potencial farmacológico, mas percebe-se que se faz necessário estudos sobre o(s) mecanismos(s) de ação dessas atividades a fim de se proporcionar o uso seguro e eficaz do cinamaldeído.
Sugar-induced metabolic imbalances are a major health problem since an excessive consumption of saccharides has been linked to greater obesity rates at a global level. Sucrose, a disaccharide composed of 50% glucose and 50% fructose, is commonly used in the food industry and found in a range of fast, restaurant, and processed foods. Herein, we investigated the effects of a TRPC4/TRPC5 blocker, ML204, in the metabolic imbalances triggered by early exposure to sucrose-enriched diet in mice. TRPC4 and TRPC5 belong to the family of non-selective Ca+2 channels known as transient receptor potential channels. High-sucrose (HS)-fed animals with hyperglycaemia and dyslipidaemia, were accompanied by increased body mass index. mesenteric adipose tissue accumulation with larger diameter cells and hepatic steatosis in comparison to those fed normal diet. HS mice also exhibited enhanced adipose, liver, and pancreas TNFα and VEGF levels. ML204 exacerbated hyperglycaemia, dyslipidaemia, fat tissue deposition, hepatic steatosis, and adipose tissue and liver TNFα in HS-fed mice. Normal mice treated with the blocker had greater hepatic steatosis and adipose tissue cell numbers/diameter than those receiving vehicle, but showed no significant changes in tissue inflammation, glucose, and lipid levels. The results indicate that TRPC4/TRPC5 protect against the metabolic imbalances caused by HS ingestion.
Staphylococcus aureus tem sido destacado devido sua capacidade de expressar uma variedade de fatores de virulência e adquirir resistência aos agentes antimicrobianos, resultando em um largo espectro de doenças que são cada vez mais difíceis de serem tratadas. Além disso, um número cada vez maior de linhagens de S. aureus tem demonstrado resistência aos agentes antimicrobianos; por esta razão, S. aureus resistente à meticilina (MRSA) e S. aureus multirresistente (MDRSA) têm sido reconhecidos como as principais causas de infecções hospitalares. O objetivo deste trabalho foi apresentar conceitos relacionados às estratégias inovadoras desenvolvidas para combater infecções causadas por S. aureus. A obtenção dos dados bibliográficos foi realizada através da pesquisa em bancos de dados (Google scholar, NCBI - National Center for Biotechnology Information, PubMed e Scielo - Scientific Electronic Library Online). Novas estratégias para o combate de infecções causadas por S. aureus estão sendo desenvolvidas, com ênfase na (i) redução da resistência a antibióticos, (ii) inibição de fatores de virulência do patógeno (produção de coagulases, biofilme, toxinas e/ou estafiloxantina), (iii) ou no aumento da resposta imune do hospedeiro. O estudo mostrou que o desenvolvimento e implantação de novas terapias antivirulência e imunomoduladoras podem resultar no descobrimento e aplicação de novos compostos. Desta forma, o presente trabalho poderá contribuir como fonte de informação para estudos posteriores de conhecimento, prevenção e controle de infecções causadas por S. aureus.
Eugenia brejoensis Mazine (Myrtaceae) is source of an essential oil (EbEO) with anti-infective activities against Staphylococcus aureus. This study evaluated the antimicrobial and anti-inflammatory potentials of EbEO in S. aureus-infected skin wounds. The excisional lesions (64 mm2) were induced on Swiss mice back (6 to 8-week-old) that were allocated into 3 groups (n = 12): 1) non-infected wounds (CON); 2) wounds infected with S. aureus ATCC 6538 (Sa); 3) S. aureus-infected wounds and treated with EbEO (Sa + EbEO). The infected groups received approximately 104 CFU/wound. The animals were treated with EbEO (10 µg/wound/day) or vehicle from the 1-day post-infection (dpi) until the 10th dpi. The clinical parameters (wound area, presence of exudate, edema intensity, etc.) were daily analyzed. The levels of inflammatory mediators (cytokines, nitric oxide, VEGF) and bacterial load were measured at the cutaneous tissue at 4th dpi and 10th dpi. Topical application of EbEO accelerated wound contraction with an average contraction of 83.48 ± 11.27 % of the lesion area until 6th dpi. In this period, the rates of lesion contraction were 54.28 ± 5.57% and 34.5 ± 2.67% for CON and Sa groups, respectively. The positive effects of EbEO on wound contraction were associated with significantly (p < 0.05) reduction on bacterial load and the release of inflammatory mediators (IL-6, IL-17A, TNF-α, NO and VEGF). Taken together, these data confirm the antimicrobial potential of EbEO and provide insights into its anti-inflammatory effects, making this essential oil an interesting candidate for the development of new therapeutic alternatives for infected cutaneous wounds.
