Background: RNA isolation from bacteria within dentine caries lesions could be difficult due to reduced amount of collectable biomass and high mRNA instability. Attempting to overcome this challenge we describe one protocol developed to extract and purify total RNA from dentine lesions. Objective: customize a bacterial RNA extraction and purification method from human carious dentine. Methods: quantity and purity of extracted RNA were measured with a microvolume UV-VIS spectrophotometer, RNA integrity was assessed by standard denaturing agarose gel electrophoresis and images were captured under ultraviolet light with camera and analyzed. DNase treatment removed genomic DNA and an additional step of purification was carried out in silica spin column. Results: final yield (ng/μl) was 67.01 ± 22.33, absorbance ratio A260/A280 = 2.0 ± 0.07 and RNA integrity were obtained. The purified samples were reversely transcribed and the expression of atpD and fabM gene from Streptococcus mutans analyzed by quantitative real-time PCR. Conclusion: the extraction methodology developed produced high-quality RNA from dentine microbiota for transcriptional analysis.
Aim: This study aimed to assess an ex situ model of biofilm-associated wounds on porcine skin for the study of Staphylococcus aureus and Pseudomonas aeruginosa biofilms in a host-like environment, after 48 to 120 h of incubation. Material and results: Ex situ and in vitro biofilms were comparatively analysed. Overall, CFU-counts and matrix quantification yielded significantly (P < 0Á05) higher results for ex situ than in vitro biofilms. Confocal microscopy revealed greater (P < 0Á05) biomass and thickness at 48-72 h and greater (P < 0Á05) robustness at 72 h of growth. S. aureus ex situ biofilms produced less (P < 0Á05) siderophore and proteases than in vitro biofilms, while P. aeruginosa ex situ biofilms produced more (P < 0Á05) siderophores and less proteases than in vitro biofilms. Conclusions: Biofilms grown ex situ present a greater amount of bacterial cells and polymeric matrix than their in vitro counterparts, reaching maturity at 72 h of growth. Moreover the production of virulence factors differs between ex situ and in vitro biofilms. Significance and Impact of the Study: These findings emphasize the importance of using ex situ biofilm models, once they mimic in vivo conditions. The use of these models brings perspectives for the pursuit of therapeutic alternatives, as tests may be performed in a host-like environment.
Introdução: A cárie dentária é a doença crônica mais prevalente na infância, sendo um problema significativo de saúde pública. A saliva é considerada um dos fatores hospedeiros mais importantes e um modulador biológico essencial que controla a velocidade e progressão da cárie. Objetivo: Realizar uma revisão da literatura sobre cárie na primeira infância (CPI) e sua possível relação com proteínas e peptídeos salivares. Métodos: Uma busca eletrônica foi realizada nas bases de dados Scopus, SciELO e PubMed Medline, aplicando a combinação dos termos "cárie na primeira infância", "saliva", "peptídeos" e "proteínas". Foram incluídos nesta pesquisa estudos que visam relacionar proteínas e peptídeos salivares e cárie dentária, indivíduos com idade entre 0-71 meses, estudos publicados na língua inglesa, ensaios randomizados e ensaios clínicos que demonstram a relação entre cárie na primeira infância e proteínas salivares. Estudos realizados com pacientes com síndromes congênitas e/ou cromossômicas, distúrbios sistêmicos ou deficiências motoras, bem como estudos em animais, estudos de revisão, cartas ao editor ou relatos de caso não foram incluídos nesta revisão. Resultados: Dos 48 estudos identificados, 13 foram selecionados para esta revisão. A maioria dos estudos mostrou associação entre proteínas e peptídeos salivares e a presença ou ausência de cárie. Seis estudos mostraram um alto nível de qualidade, enquanto o restante dos estudos foi classificado como de qualidade moderada. Conclusão: Embora a associação entre proteínas salivares e CPI seja estabelecida, mais evidências são necessárias para que os biomarcadores salivares sejam considerados preditores de risco de CPI.
Incontinentia Pigmenti is a rare multi-system dominant genetic disorder caused by a mutation of the IKBKG/NEMO gene, localized on the X-chromosome, locus Xq28, characterized by dermatological, ocular, neurological, and dental alterations. This case report shows Incontinentia Pigmenti dental findings and emphasizes the importance of the dentist's knowledge about these anomalies so that the patient can be regularly monitored throughout the development of the dentition.
