Este produto tem alto poder edulcorante e cristalização lenta, causando um melhoramento na textura e no sabor dos alimentos. Para tal finalidade, a invertase foi extraída da levedura do purê e do resíduo de pêssego c.v. Jubileu, que é amplamente cultivada no sul do Rio Grande do Sul, Brasil. A invertase de levedura comercial de pão foi utilizada como testemunha. Os extratos enzimáticos foram obtidos usando o método de extração com bicarbonato de sódio. As condições de reação ótimas foram estudadas utilizando sacarose como substrato, a 40 mM para a invertase de pão e a 220 mM para a invertase de purê de pêssego. A glicose formada foi determinada por colorimetria utilizando DNS e as proteínas pelo método de Biureto. O pH ideal foi na faixa de 5 -6 para ambas as enzimas. As temperaturas ótimas foram na faixa de 40 -50 °C para invertase de levedura de pão e na faixa de 20 -30 °C para invertase de levedura de purê de pêssego. Ambas as enzimas foram afetadas pela desnaturação de calor, sendo a invertase de pão mais termoestável. Definidas as condições ótimas de hidrólise da sacarose, estudou-se o efeito da concentração do substrato na atividade das invertases, bem como para determinar seus parâmetros cinéticos. Ambas as enzimas apresentaram comportamento Michaeliano. Os parâmetros cinéticos foram calculados por regressão não linear, sendo que os valores de Km foram 20 mM e 180 mM para a invertase de levedura de pão e para invertase de levedura de purê de pêssego, respectivamente. Para ambas, o gráfico da velocidade de hidrólise em função da concentração de sacarose foi confirmado pela linearidade da Lineweaver-Burk. Finalmente, observou-se uma equivalência entre as atividades das invertases isoladas de purê de pêssego e de seu resíduo a 25 °C e no pH de 5.Palavras-chave: purê de pêssego; levedura; invertase; hidrólise da sacarose; açúcar invertido. ResumoO questionamento sobre a tendência percebida no mercado alimentício de valorizar a alimentação da forma como era consumida no passado e sua congruência com o ritmo de vida e as tecnologias atuais foi a tônica das apresentações e discussões ocorridas durante o 4.º Simpósio de Segurança Alimentar, em Gramado. Os debates aconteceram ao redor dos cinco eixos temáticos: Tendências da alimentação, Comunicação e alimentos, Direito à alimentação, Sustentabilidade e produção de alimentos, e Garantia da qualidade. Em todos esses eixos, ficou muito claro que esta busca pelas origens − seja em seu conteúdo, seja na sua forma − já é mais do que uma tendência, vindo a impor-se como uma nova demanda social e de mercado. Por outro lado, também foi quase uma unanimidade a constatação de que esta acomodação entre antigos valores e novos modos de vida e tecnologia traz muitos desafios, que somente serão vencidos por meio de uma visão colaborativa de segurança alimentar e nutricional sustentável, que envolva os diversos atores por ela responsáveis. SummaryThe trend seen in the food market to value food as it was consumed in the past and its congruence with the pace of today's life and current technologies was the focus of the presentations and discussions that occurred during the 4th Symposium on Food Safety and Security in Gramado . The discussions were carried out around five themes: Food trends , Communication and food, Access to food, Sustainability and food production and Quality assurance and it was very clear in all of them that this search for "original" food either in content or in form is already more than a trend, emerging as a new social and market demand. On the other hand, it was almost unanimous that this accommodation between old values and new ways of life and technology brings many challenges, which will only be overcome through a collaborative vision of food safety and security, involving the various actors responsible for it.
The best conditions for in vitro sucrose hydrolysis based on invertase from Candida guilliermondii (ICg) were studied and the kinetic parameters KM,Vmax, and thermal stability of ICg were determined. Candida guilliermondii (Cg) yeast isolated and lyophilized from peach solid wastes was identified using the API 20C AUX method. Subsequently, the Cg was submitted to an autolysis process using NaHCO3 at 200 mM under 200 rpm stirring and 40 °C for 24 h. The enzyme extracts obtained were recovered through precipitation with acetone followed by dialysis and ion-exchange chromatography. The extract purified through precipitation with acetone had activity of 27.7 U.mg-1 and 56% recovery whereas the chromatography process yielded 46.5 U.mg-1 and 44.8%. The optimal sucrose hydrolysis conditions were pH 5.0 and 50 °C, resulting in KM of 30.5 mM and 28.7 mM sucrose, respectively, at 25 °C and 50 °C, both with Michaelian behavior. Thermal inactivation of ICg exhibited first-order apparent kinetics and its residual activity was typically linear between 40 °C and 70 °C. Three isoenzymes were detected through electrophoresis.
