A flow cytometry (FCM) system was chosen to analyze and sort microbiological samples, e.g., bacteria, bacterial spores, yeasts, and fungal spores, without major changes in the commercially available state. The system was further improved by addition of a stepping motor-driven scanning table that accepts standard petri dishes or microtiter plates. The electronics of the sorting system were changed to enable the sorter to deliver only one particle at a time, working in a "handshake" mode with the scanning table. Appropriate parameters, depending on the biological material and including all fluorescent stains that do not impair growth and productivity of cells were chosen to sort distinct bioparticles under aseptic conditions and to clone colonies or cultures out of them. A mutagenized sample of spores entering the germination cycle can be followed and thus provide a means to pick only viable growing cells despite the killing effect of the mutagen. One example of a typical strain improvement is illustrated. From a spore suspension of Rhizopus arrhizus, a subpopulation of morphologically different spores comprising about 5-10% of the whole population was cloned. From approximately 8,000 clones, 10 were isolated that produced approximately five-to six-fold the amount of fungal lipase activity, compared to the original strain or to reisolated clones from the mean population of clones.

Infolge der mechanischen Trägheit der Meflwerke von Llchtstrahloezlllographen, der sogenannten Oszillographenschwinger, werden schnell veränderliche MeGvorgänge mehr ode,r weniger enteteilt wiedergegeben. Diese Verzerrung der wahren Kurvenform ist bei genügend hoher £lgenfrequenz und optimaler Dämpfung des beweglichen Organs des Schwingers minimal. An Hand des bekannten ..Wiedergabegesetzes" wird im folgenden die Verzeichnung der Niederschrift für charakteristische Fälle hergeleitet, wobei sich der günstigste Dämpfungsgrad ergibt. Die so gewonnenen Erkenntnisse ermöglichen es dem Oszillographenbenutzer, seine Oszlllogramme auf Wiedergabetreue zu prüfen und gegebenenfalls nachträglich zu entzerren. In einer späteren Arbeit wird dann die praktische Durchführung der Schwlngerdämpfung mittels viskoser Flüssigkeiten unter besonderer Berücksichtigung synthetischer Substanzen behandelt werden. A. Das Wiedergabegesetz. Von den auf verschiedenen Prinzipien beruhenden Oszillographenschwingem haben wirkliche Bedeutung nur zwei Drehspulmeßwerke erlangt, nämlich die bifilare "Meßschleife" nach Blondel, der Schleifenschwinger, und neuerdings auch ein echtes Drehspulgalvanometer kleinster Abmessungen, der Spulenschwinger. Da die beweglichen Organe dieser Systeme träge Massen darstellen, die elastisch gelagert sind, haben wir es mit schwingungsfähigen mechanischen Gebilden zu tun. Am beweglichen Organ des Schwingers greift nun das äußere, von der Meßgröße hervorgerufene elektrische Drehmoment M E an, das mit der Zeit t veränderlich ist. Dieses muß mit folgenden mechanischen Drehmomenten im Gleichgewicht stehen: a) Das dem Ausschlagswinkel aus der Euhelage proportionale elastische Drehmoment M D = D , das Eichtmoment. Hierbei ist D das Direktionsmoment, das ist das rückstellende Drehmoment bezogen auf die Winkeleinheit. b) Das der Winkelbeschlejinigung proportionale Massendrehmoment = , das notwendig ist, um die bewegliche Masse des Schwingers zu beschleunigen. Dabei ist das Trägheitsmoment des beweglichen Organs einschließlich der ihm anhaftenden Dämpfungsflüssigkeit bezüglich der Drehachse. c) Das der Winkelgeschwindigkeit proportionale Dämpfungsdrehmoment M a = , das künstlich eingeführt werden muß, um Eigenbewegungen zu unterdrücken, die das bewegliche Organ als schwingungsfähiges System sonst ausführen würde, bezeichnet man als Dämpfungsfaktor. Die bekannte Bewegungsgleichung des Schwingers lautet daher: + + Dip = ME (t) (1) Da bei getreuer Wiedergabe der Meßgröße der Ausschlag des Schwingers dem äußeren Drehmoment ME proportional sein müßte, lehrt Gl. (1), daß durch das Hinzutreten des Massen-und Dämpfungsdrehmoments naturnotwendig Abweichungen zwischen dem wahren und dem angezeigten Wert des Meßvorgangs bedingt sind. Die drei Parameter der Gl. (1) lassen sich durch zweckmäßige Umformung auf einen einzigen, den -·· r D ampfungsgrad = -^-^-2 ( reduzieren, wenn man die Eigenkreisfrequenz IJ) und den Zeitmaßstab = = 2 (2) (3) (*) einführt. ÜbersichtBeim Elektronenstrahl-Oszillographen ist die (maximale) Sdireibgescbwindigkeit erheblich geringer als die Liditfleckgeschwindigkciten, die sich auf dem Schirm der Braunsdien Röhre erreichen lassen. Die Schreibgeschwindigkeit des Elektronenstrahl-Oszillographen ist also nicht durch das Meßwerk, sondern durch lichtoptische Faktoren bedingt. Mit handelsüblichen Geräten ergeben sich -abgesehen von Stoßspannungsoszillographen -Werte von l bis 10 km/s. Bei der Beurteilung des Lichtstrahl-Oszillographen spielt die Schreibgeschwindigkeit eine untergeordnete Rolle. An Hand zweier Kurvenformen (Sinuskurve und Rechtecksprung) wird im folgenden die Abhängigkeit der Lichtfleckgeschwmdigkeit von den Eigenschaften der Oszillographenschwinger errechnet. Es zeigt sich, daß die Schreibgeschwindigkeit des Lichtstrahl-Oszillographen maximal bei etwa 1,5 km/s liegt. Dank dieses relativ hohen Wertes lassen sich bis zur Grenze des nutzbaren Frequenzbereiches Amplituden schreiben, die ein bequemes Auswerten der Originalaufzeichnung ermöglichen.
