Anggrek Vanda tricolor merupakan bunga asli Indonesia yang termasuk dalam kategori appendix II, yaitu salah satu spesies flora yang terancam punah apabila perdagangan terus berlanjut tanpa adanya pengaturan. Oleh sebab itu, perlu adanya langkah konservasi secara in situ maupun ex situ. Konservasi ex situ dapat dilakukan dengan perbanyakan tanaman di luar habitat asli, salah satunya melalui perbanyakan kultur in vitro. Keberhasilan pertumbuhan planlet anggrek secara kultur in vitro ditentukan oleh jenis media sebagai sumber nutrisi untuk meningkatkan pertumbuhan planlet. Media utama yang digunakan ialah Murashige and Skoog (MS) dengan dikombinasikan suplemen nutrisi dari bahan organik berupa ekstrak buah tomat, melon, dan pisang ambon. Penelitian ini bertujuan untuk menganalisis pengaruh variasi komposisi dan konsentrasi ekstrak buah terhadap pertumbuhan planlet anggrek Vanda tricolor. Penelitian ini menggunakan Rancangan Acak Lengkap yang terdiri dari 8 perlakuan dengan 3 ulangan. Pertumbuhan diamati selama 3 bulan. Teknik pengumpulan data melalui observasi. Parameter yang diamati yaitu berat segar planlet, tinggi planlet, jumlah daun, dan jumlah akar planlet. Data dianalisis dengan ANOVA dan dilanjut Uji LSD taraf 5%. Berdasarkan hasil uji ANOVA diketahui bahwa ekstrak buah berpengaruh secara signifikan pada parameter tinggi planlet (1,97 cm) dan berat segar planlet (0,602 g) namun tidak berpengaruh secara signifikan pada jumlah daun dan jumlah akar. Berdasarkan uji LSD taraf 5% media utama yang dikombinasikan dengan ekstrak buah tomat sebanyak 150 g/l merupakan media terbaik dalam meningkatkan pertumbuhan planlet anggrek Vanda tricolor secara in vitro.
