The complexity of the pathogenetic mechanisms of the development of chronic inflammation in asthma determines its heterogeneity and insufficient treatment effectiveness. Nuclear transcription factors, which include peroxisome proliferatoractivated receptors, that is, PPARs, play an important role in the regulation of initiation and resolution of the inflammatory process. The ability of PPARs to modulate not only lipid homeostasis but also the activity of the inflammatory response makes them an important pathogenetic target in asthma therapy. At present, special attention is focused on natural (polyunsaturated fatty acids (PUFAs), endocannabinoids, and eicosanoids) and synthetic (fibrates, thiazolidinediones) PPAR ligands and the study of signaling mechanisms involved in the implementation of their anti-inflammatory effects in asthma. This review summarizes current views on the structure and function of PPARs, as well as their participation in the pathogenesis of chronic inflammation in asthma. The potential use of PPAR ligands as therapeutic agents for treating asthma is under discussion.
The aim of the study was to determine the role of rs11562975 polymorphism of TRPM8 gene and tobacco smoking in the formation of severe bronchial obstruction in asthma patients. According to this aim, 416 patients with varying severity of asthma were examined. The study design included standard spirometry, genotyping of rs11562975 TRPM8 gene polymorphism and evaluation of the status and duration of smoking. The study revealed the detrimental effect of smoking on the respiratory function in patients with a pack/year index ≥10. rs11562975 polymorphism had a modulating effect on the smoking in patients with asthma. The influence of smoking ≥10 packs-years on the formation of severe bronchial obstruction was more pronounced among carriers of GC genotype, compared to patients with GG genotype. In case of pack-year history ≥20, the GC genotype significantly increased the risk of severe bronchial obstruction in the examined patients.Keywords: asthma, TRPM8, cigarette smoke, genetic polymorphism, bronchial obstruction.Тяжелая бронхиальная астма (БА) представляет собой одну из сложнейших проблем современной пульмонологии, так как является заболеванием, тре-бующим тщательной диагностики и лечения. Состав-ляя не более 10% от всех случаев БА, тяжелая астма обусловливает более половины расходов здравоохра-нения на лечение больных БА в целом [4]. Причины тя-желого течения астмы гетерогенны и до конца не установлены. Тем не менее, вне зависимости от формы заболевания, отчетливо прослеживается эффект куре-ния на многие клинико-функциональные показатели. Курение у больных БА тесно связано с активацией вос-паления и развитием необратимых морфологических изменений в дыхательных путях, с формированием воздушных ловушек и нарушением функции легких. Курение сопровождается более выраженной персисти-рующей симптоматикой БА [1, 18], ускоренными тем-пами снижения функции легких [2, 10], повышенной частотой госпитализаций [17] и сниженным ответом на терапию ингаляционными [5,19] и пероральными глю-кокортикостероидами [6]. Наряду с активным, пассив-ное курение также сопряжено со снижением функции легких, увеличением частоты обострений, госпитали-заций и бронхиальной гиперреактивности у больных БА [7].Помимо курения, дополнительный эффект на тя-жесть заболевания могут оказывать неблагоприятные факторы окружающей среды, а в частности климати-ческие факторы и аэрополлютанты. В этом аспекте внимание привлекают каналы с транзиторным рецеп-торным потенциалом TRPM8, эспрессированные в ды-хательных путях. Доказано, что TRPM8 могут выступать в качестве рецепторов низких температур [3] и продуктов сгорания угля [8]. Более того, была об-наружена прямая взаимосвязь между курением и сте-пенью экспрессии TRPM8 [11]. The results of the first study on the effects of multi-walled carbon nanotubes (MWNTs) on the exploratory activity and the emotional state in laboratory rats assessed by the open field test are reported. During three or ten days, rats received 8-10 nm MWNTs added to their food at a dose of 500 mg/kg. It was demonstrated that, in the group of rats which were fed with MWNTs, the integrated anxiety level index began to increase as early as the third day of the experiment; on the tenth day, it appeared to be twice increased. It was also demonstrated that MWNTs decreased the integrated exploratory activity index nearly twofold on the third day and nearly fourfold on the tenth day.
