A helical aromatic foldamer was identified that undergoes tRNA acylation by a flexizyme and ribosomal peptide initiation with yields sufficiently high to perform an mRNA display selection of macrocyclic foldamer-peptide hybrids. A hybrid macrocyle binder to the C-lobe of the E6AP HECT domain was selected that showed highly converged peptide residues. A crystal structure and molecular dynamics simulations revealed that both the peptide and foldamer are helical in an intriguing reciprocal stapling fashion. The strong residue convergence could be rationalized based on their involvement in specific interactions. The foldamer stabilizes the peptide helix through stapling and through contacts with key residues. It appears to also contribute to protein binding by direct protein interactions. The results altogether highlight possible benefits in inserting an aromatic foldamer into a peptide macrocycle for the purpose of protein recognition.
Аннотация: Высокая распространенность и увеличивающаяся интенсивность стоматологических заболеваний указывает на необходимость усиления роли профилактической медицины, переориентации медицинской деятельности на поддержание здоровья человека и включает профилактику стоматологических заболеваний в число наиболее актуальных вопросов совре-менной медицины. Оптимальной лекарственной формой, обеспечивающей удобство применения и максимально возможную степень доставки действующих веществ для стоматологических профилактических препаратов, являются спреи. В данной статье приводятся результаты исследований по выбору оптимального состава препарата для профилактики заболеваний па-родонта. В качестве лекарственной формы предложен спрей, включающий в свой состав извлечения из лекарственного рас-тительного сырья -травы душицы обыкновенной и цветков ромашки, определен состав вспомогательных веществ, с учетом степени их влияния на терапевтическую активность разрабатываемого лекарственного препарата. Проведены фармакотехно-логические и биофармацевтические исследования, определена оптимальная степень высвобождения. Исходя из результатов биофармацевтических исследований установлена наибольшая степень высвобождения флавоноидов из изучаемых спрей-ком-позиций. Теоретически и экспериментально обоснован состав спрей-композиции для профилактики заболеваний пародонта: глицерина -7,5; 1 % раствора метилцеллюлозы -7,5; комплексного спирто-водного извлечения из травы душицы обыкно-венной и цветков ромашки. Эпидемиологические исследования всемирной организации здравоохранения свидетель-ствуют о широком распространении и интенсивной ди-намики инфекционно-воспалительных заболеваниях пародонта (гингивит, стоматит и др.), этими заболева-ниями страдает до 95% взрослого населения, и до 80% детей.На сегодняшний день современная номенклатура лекарственных препаратов для лечения и профилак-тики заболеваний пародонта на 75% представлена ле-карственными препаратами синтетического происхож-дения. В данной группе лекарственных препаратов присуще недостатки: высокая сенсибилизирующая ак-тивность, повышенная чувствительность к компонен-там, беременность, период грудного вскармливания, возраст до 18 лет и др. С данной точки зрения, лекар-ственные растительные препараты наряду с эффектив-ностью обладают сравнительно низкой безопасностью, мягкостью действия, широтой фармакологических эф-фектов, минимальным анальгезирующим действием, и местным раздражающим действием.
