Trabecular bone fractures constitute a major health issue for the modern societies, with the currently established prediction methods of fracture risk, such as bone mineral density (BMD), resulting in errors up to 40%. Fracture-zone prediction based on bone's microstructure has been recently proposed as an alternative prediction method of fracture risk. In this paper, a classification system (CS) for the automatic fracture-zone prediction based on an Ensemble of Imbalanced Learning methods is proposed, following the observation that the percentage of the actual fractured bone area is significantly smaller than the intact bone in the case of a fracture event. The sample is divided into Volumes of Interest (VOIs) of specific size and 29 morphometrical parameters are calculated from each VOI, which serve as input features for the CS in order for it to separate the input patterns in to two classes: fractured and nonfractured. To this end, two well-established Imbalanced Learning methods, namely Random Undersampling and Synthetic Minority Oversampling, and two popular classification algorithms, namely Multilayer Perceptrons and Support Vector Machines, are tested and combined accordingly, to provide the best possible performance on a dataset that contains 45 specimens' pre- and postfailure scans. The best combination is then compared with three well-established Ensembles of Imbalanced Learning methods, namely RUSBoost, UnderBagging and SMOTEBagging. The experimental results clearly show that the proposed CS outperforms the competition, scoring in some occasions more than 90% in G-Mean and Area under Curve metrics. Finally, an investigation on the significance of the various trabecular bone's biomechanical parameters is made using the sequential forward floating selection technique, in order to identify possible biomarkers for fracture-zone prediction.
Segmentation of biomedical images is of great importance in various studies aiming to both the identification of regions of interests within the image and the performance of quantified measurements. Nevertheless, the segmentation of the biomedical images represents a wide range of medical cases and there is not a unique technique applicable to all kinds of medical images. In this study, three popular techniques for segmenting micro-CT images of bone microstructures are evaluated. Fixed threshold, Otsu's algorithm and a modified version of the Chan-Vese segmentation technique have been applied on micro-CT images and have been compared to higher resolution golden standard, that is histological images. The modification of the Chan-Vese technique is based on the novel implementation of a new initialization process called the Branch Point Initialization. Stereological measurements were performed on all the segmented images and statistically compared to the golden standard. Fixed threshold and the modified Chan-Vese technique have shown comparable results, with a maximum significant error of about 10%. However, Chan-Vese showed an easier, faster and more reliable segmentation procedure for optimal settings identification. The Otsu's method showed a maximum error larger than 20%. Given the limits and advantages of the known segmentation techniques, the proposed modified Chan-Vese active contour technique shows high potential for use in the segmentation of micro-CT images as well as in other high-resolution X-ray images. This potential is augmented by the recent introduction of high-resolution clinical technologies for which standard techniques have already shown to be insufficient.
Effective detection of autism spectrum disorder (ASD) is a complicated procedure, due to the hundreds of parameters suggested to be implicated in its etiology. As such, machine learning methods have been consistently applied to facilitate diagnosis, although the scarcity of potent autism-related biomarkers is a bottleneck. More importantly, the variability of the imported attributes among different sites (e.g., acquisition parameters) and different individuals (e.g., demographics, movement, etc.) pose additional challenges, eluding adequate generalization and universal modeling. The present study focuses on a data-driven approach for the identification of efficacious biomarkers for the classification between typically developed (TD) and ASD individuals utilizing functional magnetic resonance imaging (fMRI) data on the default mode network (DMN) and non-physiological parameters. From the fMRI data, static and dynamic connectivity were calculated and fed to a feature selection and classification framework along with the demographic, acquisition and motion information to obtain the most prominent features in regard to autism discrimination. The acquired results provided high classification accuracy of 76.63%, while revealing static and dynamic connectivity as the most prominent indicators. Subsequent analysis illustrated the bilateral parahippocampal gyrus, right precuneus, midline frontal, and paracingulate as the most significant brain regions, in addition to an overall connectivity increment.
Abstract. Prediction of trabecular fracture zone is a very important element for assessing the fracture risk in patients. The assumption that failure always occurs in local bands, the so called 'fracture zones', with the remaining regions of the structure largely unaffected has been visually verified. Researchers agreed that the identification of the weakest link in the trabecular framework can lead to the prediction of the fracture zone and consequently of the failure event. In this paper, a decision support system (DSS) is proposed for the automatic identification of fracture zone. Initially, an automatic methodological approach based on image processing is applied for the automatic identification of trabecular bone fracture zone in micro-CT datasets, after mechanical testing. Then, a local analysis of the whole specimen is performed on order to compare the structure (Volumes of Interest -VOI) of the broken region to the unbroken one. As a result, for every VOI, 29 morphometrical parameters were computed and used as initial features to the proposed DSS. The DSS comprises of two main modules: the feature selection module and the classifier. The feature selection module is used for reducing the initial size of the input features' subset (29 features) and for keeping the most informative features in order to increase the classification's module performance. To this end, the Sequential Floating Forward Selection (SFFS) algorithm with Fuzzy C-Means evaluation criterion was implemented. For the classification, several classification algorithms including the Multilayer Perceptron (MLP), the Support Vector Machines (SVM), the Naïve Bayesian (NB), the k-Nearest Neighbor (KNN) and the k-Means (KM) have been used. Comparing the performance of these classification algorithms, the SFFS-SVM scheme provided the best performance scoring 98% in terms of overall classification accuracy.
Το αντικείμενο της παρούσας διδακτορικής διατριβής είναι η επεξεργασία δεδομένων μίκρο-τομογραφίας οστών μέσω της εφαρμογής εξειδικευμένων υπολογιστικών αλγορίθμων. Στα πλαίσια της διατριβής γίνεται η παρουσίαση καινοτόμων αυτοματοποιημένων μεθοδο-λογιών τμηματοποίησης και ευθυγράμμισης εικόνων μίκρο-τομογραφίας οστών και ταξινόμησης δεδομένων που προκύπτουν από την εξαγωγή ιστομορφολογικών χαρακτηριστικών από τις εικόνες αυτές. Αρχικά, πραγματοποιείται εισαγωγή στα κυριότερα στοιχεία και δυναμικές διαδικασίες που χαρακτηρίζουν το οστό, στις παθογένειες και στις μεθόδους ανάλυσης και απεικόνισης του οστού. Πρώτα, παρουσιάζεται το σύστημα τμηματοποίησης εικόνας, αφού έχει προηγηθεί μια ολοκληρωμένη μελέτη των πλεονεκτημάτων και μειονεκτημάτων της εφαρμογής συγκεκριμένων αλγορίθμων τμηματοποίησης στις εικόνες μίκρο-τομογραφίας σπογγώδους οστού. Το σύστημα τμηματοποίησης εικόνας υλοποιείται από ένα νέο ολοκληρωμένο πλαίσιο τμηματοποίησης εικόνων με το όνομα Τμηματοποίηση Ανεξάρτη-των Ενεργών Περιγραμμάτων που συνδυάζει ένα σύνολο από καινοτόμες τεχνικές τμηματοποίησης βασισμένες στα Ενεργά Περιγράμματα. Η προτεινόμενη μεθοδολογία καθιστά δυνατή την ευρεία χρήση των Ενεργών Περιγραμμάτων σε πλήρη σύνολα δεδομένων μίκρο-τομογραφίας οστών και ταυτόχρονα βελτιώνει αισθητά την επίδοση της τμηματοποίησης σε αντίστοιχα τεχνητά και πραγματικά δεδομένα. Επίσης, επιδεικνύεται η δυνατότητα ευρύτερης εφαρμογής της προτεινόμενης μεθόδου και σε άλλα είδη ιατρικών εικόνων, όπως εικόνες Αξονικής Τομογραφίας (CT) και Ιστολογικών δεδομένων. Στη συνέχεια, παρουσιάζεται το σύστημα ευθυγράμμισης εικόνων μίκρο-τομογραφίας οστών το οποίο εφαρμόζεται στις δυαδικές εικόνες κάθε δείγματος πριν και μετά από τεχνητό σπάσιμο, με σκοπό την ανάπτυξη ενός ποσοτικού κριτηρίου αυτόματης εύρεσης της περιοχής κατάγματος. Στη συγκεκριμένη μελέτη παρουσιάζονται δισδιάστατες και τρισδιάστατες τεχνικές ευθυγράμμισης, ενώ ακολουθούν αποτελέσματα ευθυγράμμισης με βάση τη χρήση κατάλληλων κριτηρίων αξιολόγησης προκειμένου να διερευνηθεί η ακρίβειά τους. Επιπλέον, παρουσιάζεται ένα ολοκληρωμένο σύστημα αυτόματης ταξινόμησης για την αναγνώριση των περιοχών κατάγματος οστού με βάση τις τιμές διαφόρων ιστομορφομετρικών παραμέτρων που έχουν εξαχθεί από Όγκους Ενδιαφέροντος (VOI) των τμηματοποιημένων εικόνων. Η συγκεκριμένη μελέτη συνδυάζει τις τεχνικές Μη-Ισορροπημένης μάθησης και Πολλαπλών Ταξινομητών με δημοφιλείς αλγορίθμους ταξινόμησης, στοχεύοντας στην εύρεση του βέλτιστου συνδυασμού τους. Το τελικό σύστημα ελέγχεται σε ένα εκτενές σύνολο δεδομένων όπου παρουσιάζει πολύ υψηλές επιδόσεις συγκριτικά με τις απλούστερες μεθόδους, δίχως να αυξάνει σημαντικά την πολυπλοκότητα της διαδικασίας και το υπολογιστικό κόστος. Τέλος, ακολουθεί μια αξιολόγηση των σημαντικότερων ιστομορφομετρικών παραμέτρων με χρήση τεχνικών Επιλογής Χαρακτηριστικών σε σύνολα δεδομένων που έχουν υποστεί επεξεργασία με τεχνικές Μη-Ισορροπημένης μάθησης, με στόχο την εξαγωγή πιθανών βιοδεικτών για την περιοχή κατάγματος.
