Chemical evaluation of the semi-purified fraction from the seeds of guaraná, Paullinia cupana H.B.K. var. sorbilis (Mart.) Ducke, yielded the following compounds: caffeine, catechin, epicatechin, ent-epicatechin, and procyanidins B 1 , B 2 , B 3 , B 4 , A 2 , and C 1 . Measurement of the antioxidant activity by reduction of the DPPH radical confirmed the anti-radical properties of the aqueous (AqE) and crude (EBPC) extracts and semi-purified (EPA and EPB) fractions. The EPA fraction showed radical-scavenging activity (RSA) and protected DPPH from discoloration at 5.23±0.08 (RSD%=1.49) µg/mL, and for the phosphomolybdenum complex showed a higher Relative Antioxidant Capacity (RAC) at 0.75±0.01 (1.75). The EPA fraction had a total polyphenolics content of 65.80%±0.62 (RSD%=0.93). The plant drug showed 5.47% ± 0.19 (RSD%=3.51) and 6.19% ± 0.08 (RSD%=1.29) for total polyphenolics and methylxanthines, respectively. In vitro assessment of the antibacterial potential of the Paullinia cupana extracts against 1951Streptococcus mutans showed that these could be used in the prevention of bacterial dental plaque.
RESUMO:Dasyphyllum tomentosum (Spreng.) Cabrera, açucará ou espinho-de-agulha, pertence à família Asteraceae, a qual compreende muitas espécies com propriedades terapêuticas. O objetivo deste trabalho foi avaliar extratos e frações de folhas e de cascas do caule de D. tomentosum, com relação as atividade antioxidante, citotóxica e hemolítica em testes in vitro. Todas as amostras apresentaram atividade antioxidante pelo método de inibição de DPPH, com destaque para a fração acetato de etila obtida das folhas cuja atividade foi comparável à dos padrões ácido ascórbico e rutina. Com relação à redução do complexo fosfomolibdênio, observou-se que esta mesma fração foi semelhante somente a rutina enquanto a fração obtida das cascas do caule apresentou resultado superior. Não foi observada atividade citotóxica e hemolítica frente aos modelos utilizados com os extratos e frações. Os resultados obtidos demonstram o potencial antioxidante da espécie sem apresentar toxicidade.Palavras-chave: antioxidante, DPPH, fosfomolibdênio, hemolítica, Asteraceae.ABSTRACT: Antioxidant activity and preliminary toxicity of the extracts and fractions obtained from the leaves and stem bark of Dasyphyllum tomentosum (Spreng.) Cabrera. Dasyphyllum tomentosum (Spreng.) Cabrera, which belongs to the Asteraceae family, is well known for having many species with therapeutic properties. The aim of this study was to evaluate the extracts and fractions from the leaves and stem bark of D. tomentosum with respect to the antioxidant, cytotoxic and hemolytic activity in in vitro tests. All samples showed antioxidant activity by the DPPH inhibition method, especially the ethyl acetate fraction obtained from the leaves, whose activity was comparable to that of standard ascorbic acid and rutin. Regarding the reduction of the phosphomolybdenum complex, we noted that this same fraction was only similar to rutin and the fraction obtained from the stem bark showed superior results. There was no cytotoxicity and hemolytic activity compared to the models used in the extracts and fractions. The results demonstrate the antioxidant potential of the species without showing toxicity.
RESUMO:Este trabalho visou avaliar a ação fitotóxica de extrato e frações obtidas das folhas de Bauhinia ungulata L sobre a divisão celular e atividade das enzimas α-amilase, catalase, peroxidase e polifenoloxidase em plântulas de alface (Lactuca sativa, var Grand rapis). Sementes de alface foram expostas às concentrações de 250, 500 e 1000 µg.mL -1 do extrato de Bauhinia ungulata e frações do mesmo, em condições ambientais controladas. Após três dias de protusão radicular, as raízes das plântulas foram cortadas e submetidas a preparo específico para visualização e contagem do número de células em cada fase mitótica. No sétimo dia, as amostras foram submetidas à avaliação da atividade enzimática utilizando técnica específica para cada enzima e leitura espectrofotométrica. Na análise da divisão celular observou-se que houve redução no número de células em mitose nas raízes das plântulas testadas, e a fração acetato de etila apresentou maior efeito inibitório sobre a divisão celular. O efeito sobre as enzimas que demonstram alterações no sistema de defesa antioxidante de L. sativa mostram que o extrato bruto e frações alteraram a produção das enzimas α-amilase, peroxidase, catalase e polifenol oxidase em, ao menos, uma das concentrações testadas, sendo um indicativo de estresse que interfere na divisão celular. Os resultados obtidos apontam para a presença de compostos com atividade inibidora ou estimulatória nas folhas de B. ungulata revelando potencial fitotóxico sobre as plântulas de alface.
