TFs involved in drought tolerance in plants may be utilized in future for developing drought tolerant cultivars of wheat and some other crops. Plants have developed a fairly complex stress response system to deal with drought and other abiotic stresses. These response systems often make use of transcription factors (TFs); a gene encoding a specific TF together with -its target genes constitute a regulon, and take part in signal transduction to activate/silence genes involved in response to drought. Since, five specific families of TFs (out of >80 known families of TFs) have gained widespread attention on account of their significant role in drought tolerance in plants, TFs and regulons belonging to these five multi-gene families (AP2/EREBP, bZIP, MYB/MYC, NAC and WRKY) have been described and their role in improving drought tolerance discussed in this brief review. These TFs often undergo reversible phosphorylation to perform their function, and are also involved in complex networks. Therefore, some details about reversible phosphorylation of TFs by different protein kinases/phosphatases and the co-regulatory networks, which involve either only TFs or TFs with miRNAs, have also been discussed. Literature on transgenics involving genes encoding TFs and that on QTLs and markers associated with TF genes involved in drought tolerance has also been reviewed. Throughout the review, there is a major emphasis on wheat as an important crop, although examples from the model cereal rice (sometimes maize also), and the model plant Arabidopsis have also been used. This knowledge base may eventually allow the use of TF genes for development of drought tolerant cultivars, particularly in wheat.

Genetic dissection of grain weight in bread wheat was undertaken through both genome-wide quantitative trait locus (QTL) interval mapping and association mapping. QTL interval mapping involved preparation of a framework linkage map consisting of 294 loci {194 simple sequence repeats (SSRs), 86 amplified fragment length polymorphisms (AFLPs) and 14 selective amplifications of microsatellite polymorphic loci (SAMPL)} using a bi-parental recombinant inbred line (RIL) mapping population derived from Rye Selection111 9 Chinese Spring. Using the genotypic data and phenotypic data on grain weight (GW) of RILs collected over six environments, genome-wide single locus QTL analysis was conducted to identify main effect QTL. This led to identification of as many as ten QTL including four major QTL (three QTL were stable), each contributing [20% phenotypic variation (PV) for GW. The above study was supplemented with association mapping, which allowed identification of 11 new markers in the genomic regions that were not reported earlier to harbour any QTL for GW. It also allowed identification of closely linked markers for six known QTL, and validation of eight QTL reported earlier. The QTL identified through QTL interval mapping and association mapping may prove useful in marker-assisted selection (MAS) for the development of cultivars with high GW in bread wheat.

Foxtail millet (Setaria italica), the second largest cultivated millet crop after pearl millet, is utilized for food and forage globally. Further, it is also considered as a model crop for studying agronomic, nutritional and biofuel traits. In the present study, a genome-wide association study (GWAS) was performed for ten important agronomic traits in 142 foxtail millet core eco-geographically diverse genotypes using 10 K SNPs developed through GBS-ddRAD approach. Number of SNPs on individual chromosome ranged from 844 (chromosome 5) to 2153 (chromosome 8) with an average SNP frequency of 25.9 per Mb. The pairwise linkage disequilibrium (LD) estimated using the squared-allele frequency correlations was found to decay rapidly with the genetic distance of 177 Kb. However, for individual chromosome, LD decay distance ranged from 76 Kb (chromosome 6) to 357 Kb (chromosome 4). GWAS identified 81 MTAs (marker-trait associations) for ten traits across the genome. High confidence MTAs for three important agronomic traits including FLW (flag leaf width), GY (grain yield) and TGW (thousand-grain weight) were identified. Significant pyramiding effect of identified MTAs further supplemented its importance in breeding programs. Desirable alleles and superior genotypes identified in the present study may prove valuable for foxtail millet improvement through marker-assisted selection.

In bread wheat, high grain protein content (GPC) determines nutritional value, processing properties and quality of the end-product. In view of this, marker-assisted selection (MAS) was performed for introgression of a major gene for high GPC (Gpc-B1) into 10 wheat genotypes. As a result, 124 BC 3 F 5 /F 6 progenies with Gpc-B1 were developed and evaluated in multi-location field trials. Significant interaction of Gpc-B1 with the recipient parent genotypes and the environment was noticed. However, a total of seven MASderived progenies with significantly higher GPC (14.83-17.85%) than their recipient parental genotypes and having no yield penalty were obtained. In these selected progenies, no significant negative correlation of grain yield with GPC (%) or protein yield was observed suggesting that GPC could be improved without yield penalty. This study thus suggested that MAS in combination with phenotypic selection is a useful strategy for development of wheat genotypes with high GPC associated with no loss in yield.

Multi-locus genome wide association study was undertaken using a set of 320 diverse spring wheat accessions, which were each genotyped for 9,626 SNPs. The association panel was grown in replicated trials in four environments [two each in irrigated (IR) and rainfed (RF) environments], and phenotypic data were recorded for five traits including days to heading, days to maturity, plant height, thousand grain weight and grain yield. Forty-six significant marker-trait associations (MTAs) were identified for five traits. These included 20 MTAs in IR and 19 MTAs in RF environments; seven additional MTAs were common to both the environments. Five of these MTAs were co-localized with previously known QTL/MTAs and the remaining MTAs were novel and add to the existing knowledge. Three desirable haplotypes for agronomic traits, one for improvement in RF environment and two for improvement in IR environment were identified. Eighteen (18) promising candidate genes (CGs) involved in seven different biological activities were also identified. The expression profiles of four (Trehalose-6-Phosphate, APETALA2/Ethylene-responsive factor, DNA-binding One Zinc Finger and Gibberellin-dioxygenases) of the 18 genes showed that they were induced by drought stress in the wheat seedlings. The MTAs, haplotypes and CG-based markers may be used in marker-assisted breeding for drought tolerance in wheat.
