Summary. Aim: To identify the association of serum miRNAs with neoadjuvant polychemotherapy response in patients with breast cancer of luminal A and B subtypes. Materials and Methods: We analyzed the expression levels of circulating miR-21, -155, -182, -373, -199a, -205, -375 in serum of 182 breast cancer patients using real-time polymerase chain reaction. Each case was characterized by TMN criteria using morphological and immunohistochemical analyses. Results: Serum levels of miR-205 and -375 are associated with the response of luminal A tumors and miR-205 and -21 with the response of luminal B tumors to neoadjuvant polychemotherapy in fluorouracil + doxorubicin + cyclophosphamide and doxorubicin + cyclophosphamide regimens. In addition, we found correlation of miR-155, -182, -199a, -375 with 3-year relapse-free survival of patients. Based on the obtained data, we developed innovative prognostic and predictive panels to assess the drug sensitivity of tumors and lower the risk of breast cancer recurrence, which would significantly improve the treatment outcomes and the quality of life of patients. Conclusions: Serum levels of miR-155, -182, -199a, -205, -375 can be used as predictive and prognostic panel for monitoring BC course in Ukrainian population.
ВСТУП. Подолання імунологічної толе рант-ності до пухлиноасоційованих антигенів (ПАА) є ключовим завданням імунотерапії пухлинної хвороби [1,2]. Одним із підходів до його вико-нання є розробка та застосування ксеногенних протипухлинних вакцин (ПВ), які містять пухлинні антигени організмів іншого виду [3]. Показано, що саме використання чужорідних аналогів ПАА здатне викликати імунну відповідь проти власних ендогенних протеїнів [4, 5]. Більш того, структурні відмінності ксеногенних ПАА від їх людських аналогів сприяють індукції специфічних про-типухлинних реакцій не тільки на ранніх, але й на пізніх стадіях захворювання, коли організм хво рого перебуває під імуносупресивним впли вом пухлини та власної імунної системи. То му, порівняно з гомологічними аналогами, ксеногенні ПВ можуть стати ефективнішими як для профілактики розвитку рецидивів та метастазів, так і для генерації процесів, що призводять до руйнування розвинутої пухлини [2,6, 7].Експресія ембріональних антигенів супрово-джує процес трансформації нормальної кліти-ни в злоякісну [8,9], тому їх використання як "універсальних" специфічних імуногенів від-криває нові можливості в конструюванні ксе-ногенних ПВ [10,11]. У попередній роботі ми показали, що ПВ, виготовлені на основі антигенів ембріональних тканин різного гістогенезу лю -дини та глікопротеїду в. subtilis B-7025, про-являли протипухлинну активність у мишей із саркомою 37 [12]. Одержано високомолекулярні ембріональні протеїни курки (ЕПК), які мають певну гомологію з поверхневими протеїнами клітин модельних пухлин [13] і в подальшому можуть бути використані як компоненти при конструюванні ксеногенних ПВ [14, 15].Для підвищення імуногенності зазначених ЕПК та посилення специфічної імунної відповіді необхідно підібрати відповідний ефективний ад'ювант. Особливо перспективними на сьо-год ні є ад'юванти мікробного походження, які взаємодіють з Toll-подібними рецепторами клітин природного імунітету та запускають про-цес формування адаптивного імунітету, тому їх досить широко використовують для виготовлення ПВ [16,17]. Крім того, ад'юванти, отримані з бактеріальних продуктів, здатні модифікувати ПАА, індукувати продукування широкого спектра цитокінів, що дозволяє суттєво зменшити дозу імуногена і частоту його введення [18].Метою даної роботи було вивчити можливість підсилення імунної відповіді організму тварин на ксеногенні ембріональні протеїни за допомогою ад'ювантів різного походження.
Anticancer xenogeneic vaccine -is an agent, containing antigens of embryonic origin that underwent Olena Kruts et al.: Combined Application of Anticancer Vaccines of IEPOR Series and Doxorubicin in Rats with Transplanted Walker Carcinosarcoma
The research was aimed at identifying the relationship between the oncogenic (-182, -155) and the onco-suppressive (-122, -200b) miRNAs in tumor cells with clinical and pathological characteristics of breast cancer (BC) and at establishing their clinical significance. MiRNA levels have been determined by reverse transcription polymerase chain reaction (RT-PCR) in real time. The expression of estrogen (ER) and progesterone (PR) receptors, and Her2/neu epidermal growth factor has been ascertained by immunohistochemical method. The correlation of these miRNAs expression with the disease stage, the presence of metastatic lesion of local lymphatic nodes, the differentiation degree, and the tumor molecular subtype has been established. The obtained results have confirmed the involvement of the tested miRNAs in the formation of BC malignancy degree, their association with tumor aggressiveness and have defined the prospects of further studies of the proposed miRNAs panel for their use as BC additional markers.
