Mit einem histochemischen Nachweisverfahren für Akrosin Beim einzelnen Spermium (Gelatinesubstratfilm‐Methode) wurde der Einfluß uteriner, tubaler and follikulärer Genitalsekrete auf das enzymatische Akrosomensystem ejakulierter Eberspermien in vitro überprüft. Als Parameter der Enzymaktivität wurden die Lysisflächen (μm2/Spermium) der Spermien auf Gelatinesubstrat nach 10, 60 und 240 Minuten Inkubation in den Sekreten aus oestrischer bzw. dioestrischer Zyklusphase ausgewertet. Spermien aus oestrischen Sekreten entwickelten größere Lysishöfe (P ≤ 0,01) als Samenzellen aus dioestrischen Medien oder Tris‐Puffer (Kontrolle). Follikelflüssigkeit induzierte die größten Halos (P ≤ 0,01). Das Maximum der proteolytischen Aktivität wurde bei oestrischen Ansätzen nach 60 Minuten Inkubation erreicht und fiel nach 240 Minuten auf ein Minimum ab (P ≤ 0,01). Der Gelatinesubstratfilm‐Test eignet sich als Parameter zur Untersuchung akrosomaler Membranen unter Einfluß kapazitierender Medien.
Contents: The influence of female genital tract fluids on the proteo(gelatino)lytic activity of ejaculated boar spermatozoa.
Using the gelatin substrate film technique as a histochemical assay for acrosin from individual boar spermatozoa, the influence of oviductal, uterine and follicular fluids on the enzymatic state of the acrosome was investigated. Enzyme activity was estimated by determination of digested areas around the sperm head of cells which were incubated 10, 60 and 240 minutes with genital secretions from estrus or diestrus cycle. Spermatozoa incubated with estrus fluids caused more extensive halo areas (p ≤ 0.01) than those from diestrus secretions or control medium (Tris puffer). Follicular fluid induced the most extensive digestion zone (p ≤ 0.01). After a 60 minutes incubation period maximal proteolytic activity appeared and decreased to a minimum after 240 minutes (p ≤ 0.01). The gelatin substrate test yields new aspects for studying acrosomal sperm membranes on the influence of capacitating agents.
