Recebido em 23/4/10; aceito em 9/9/10; publicado na web em 8/12/10 DEVELOPMENT AND VALIDATION OF ANALYTICAL METHOD FOR DETERMINATION OF SIMVASTATIN IN CAPSULES.Simvastatin is an oral anti-hyperlipidemic that has been widely used to reduce the cardiovascular disease risk. There isn't any pharmacopoeic method to assay this drug in capsules. It was proposed and validated a method for determining the content of simvastatin in capsules by UV/visible spectrophotometry (λ = 237 nm), a more affordable method for the compounding pharmacy. The method was validated for linearity, specificity, range, accuracy, precision, performance, robustness, and limits of detection and quantification.Keywords: simvastatin; spectrophotometry; validation studies. INTRODUÇÃOAs doenças cardiovasculares (DC), dentre as quais figura a doença arterial coronariana (DAC), são um grande problema de saúde pública, uma vez que é a principal causa de morte no mundo. O Brasil acompanha este fenômeno internacional apresentando valores percentuais de mortalidade em torno de 25% para DC, o que representa cerca de 250.000 óbitos ao ano. Nesse contexto, a sinvastatina insere-se como um agente hipocolesterolêmico amplamente utilizado. Trata-se de uma lactona inativa que é convertida em seu β,δ-hidroxiácido correspondente (forma ativa), via metabolismo hepático pelo citocromo P450, após administração oral. Este fármaco (Figura 1) é um potente inibidor da enzima 3-hidroxi-3-metil-glutaril-coenzima A (HMGCoA) redutase, que catalisa a síntese de colesterol.1 É comercializada com o nome de Zocor ® na forma de comprimidos revestidos nas concentrações de 10, 20, 40 e 80 mg e como Sinvastacor ® na forma de comprimido revestido de 5 mg. Além das indústrias farmacêuticas, a sinvastatina também atraiu o interesse das farmácias magistrais, que atualmente manipulam extensivamente o fármaco na forma de cápsulas. Tais farmácias passaram, nos últimos anos, por um processo de expansão no número de medicamentos dispensados, o que pode acarretar riscos para o paciente, caso os requisitos de segurança, qualidade e eficácia não sejam comprovados. Desta forma, é essencial o controle de qualidade do produto acabado nas farmácias de manipulação, pois um dos parâmetros que mais comumente apresentam desvios da qualidade é a quantidade de princípio ativo determinada no doseamento. 4,5 Dentre as monografias de produtos acabados que constam nos compêndios oficiais reconhecidos pela Agência Nacional de Vigilância Sanitária pesquisados (Farmacopeias Brasileira,
In the current study, we have coupled Fused Deposition Modelling (FDM) for the fabrication of plain polyvinyl alcohol (PVA) tablets followed by dispensing of minoxidil ethanolic solutions using inkjet printing. The use of a drop-on-solid printing approach facilitates an accurate and reproducible process while it controls the deposition of the drug amounts. For the purpose of the study, the effect of the solvent was investigated and minoxidil ink solutions of ethanol 70% v/v (P70) or absolute ethanol (P100) were applied on the plain PVA tablets. Physicochemical characterization showed that solvent miscibility with the polymer substrate plays a key role and can lead to the formation of drug crystals on the surface or drug absorption in the polymer matrix. The produced minoxidil tablets showed sustained release profiles or initial bursts strongly affected by the solvent grade used for dispensing the required dose on drug loaded 3D printed tablets. This paradigm demonstrates that the coupling of FDM and inkjet printing technologies could be used for rapid development of personalized dosage forms.
Introdução: Devido às características do organismo humano durante a infância, os medicamentos utilizados por esse público necessitam ser adequados quanto à dosagem e forma farmacêutica, garantindo tratamento correto e adesão do paciente. No entanto, a escassez comercial de alguns medicamentos leva ao uso off-label de formulações destinados ao tratamento de indivíduos adultos, o que pode ser inadequado à terapia pediátrica. Nesse sentido, a furosemida é um diurético de alça utilizado off-label em crianças e não é comercialmente disponível no Brasil sob a forma líquida, a qual é mais adequada ao público pediátrico. Objetivo: Desenvolver e avaliar a qualidade de uma preparação oral líquida off-label de uso pediátrico contendo furosemida na concentração de 10 mg/mL. Material e Métodos: Uma suspensão do ativo foi preparada e o controle de qualidade físico-quimico e microbiológico da formulação foi realizado. Do ponto de vista físico-químico, foram avaliados os parâmetros: aspecto, pH, viscosidade e teor do fármaco. Em relação às análises microbiológicas, avaliou-se o crescimento visível de microrganismos mesófilos totais (fungos e bactérias) após incubação do produto em meio de cultura específico para esses microrganismos. Resultados: Obteve-se formulação com aspecto esbranquiçado e inodora, com formação de sedimento após repouso, o qual se ressuspendia facilmente após agitação. O medicamento apresentou pH médio igual a 7,16 ± 0,138, viscosidade média de 40,79 cP ± 0,100 e teor médio de ativo igual a 108,7% ± 0,075. Além disso, não houve crescimento visível de microrganismos mesófilos no produto desenvolvido. Conclusão: A suspensão apresentou resultados satisfatórios em relação aos parâmetros avaliados, com características semelhantes às encontradas na literatura para outras suspensões de furosemida. Dessa forma, é uma opção de formulação que pode ser manipulada, sob a forma extemporânea, e administrada em pacientes pediátricos.
