Tóm tắt: Trong nghiên cứu này chúng tôi tiến hành đánh giá ảnh hưởng của thời gian trồng lên sinh trưởng và khả năng tích lũy một số thành phần saponin-ginsenoside chính trong các phần khác nhau (dưới mặt đất và khí sinh) của cây Sâm việt nam (SVN) trồng tại Nam Trà My (Quảng Nam). Kết quả cho thấy: cây SVN sinh trưởng và phát triển theo thời gian trồng (2-6 năm) nhưng tốc độ tăng rõ rệt nhất ở năm thứ 4. Hàm lượng saponin tổng số ở phần dưới mặt đất tăng dần theo thời gian trồng với hàm lượng tích lũy được khá cao (từ 5,23% đến 13,88% sau 2-6 năm), hơn hẳn so với trong phần khí sinh (ở mức 2,51%-3,61% và không biến động nhiều qua thời gian). Các thành phần Rg1, Rb1, Rd đều phân tích được trong toàn cây SVN nhưng chiếm lượng nhiều hơn trong phần dưới mặt đất (sau 6 năm tương ứng đạt 2,04%, 2,56% và 1,72%), còn ở phần khí sinh hàm lượng chủ yếu là Rb1 (1,15-1,4%), Rg1 và Rd có rất ít (dưới 0,15%). Thành phần Re không phát hiện thấy trong phần khí sinh và có rất ít ở phần dưới mặt đất (dưới 0,05%). Thành phần MR2 đặc trưng cho SVN không phát hiện trong phần khí sinh nhưng chiếm tỷ lệ khá cao trong phần dưới mặt đất với hàm lượng đạt 2,23%; 3,26%; 4,54%; 5,75% và 5,23%, tương ứng sau 2-6 năm trồng. Xét chung cả về tiêu chuẩn trọng lượng và hàm lượng hoạt chất, cây SVN sau từ 5 năm trồng bắt đầu có thể thu hoạch sử dụng làm dược liệu.
Fish is a healthy, high protein and low fat food that encourages the health and growth of people, especially children. However, in fact fish is very sensitive to many diseases which affects the productivity and quality of fish. Therefore, identifying the cause of the diseases and finding preventive measures become an urgent task today. In the previous study, we isolated Streptomyces antibioticus strain 1083 that has the ability to antagonize Aeromonas hydrophila - a pathogenic bacterium in fish. Based on the obtained results, we continue to perform this study to determine optimal conditions for the culture of S. antibioticus strain 1083 in order to produce antimicrobial compounds against A. hydrophila. The production of antagonists by the strain 1083 was optimized by controlling the condition of different inoculations such as media, pH, temperature and incubation period. The results indicated that International Streptomyces Project 2 (ISP2) was the best medium for S. antibioticus strain 1083 to produce the highest antimicrobial activity against A. hydrophila with 32 mm in diameter of inhibited zone. The actinomycete strain 1083 could express the maximum antimicrobial activity when they were incubated in shaker incubator (200rpm) at 40oC with pH8 in 8 days. The ability of the actinomycete strain in antagonism against A. hydrophila was evaluated by adding different culture medium volume of S. antibioticus strain 1083. With adding 10% cultured solution volume of S. antibioticus strain 1083 into the culture medium of A. hydrophila, after 1 day of inoculation the number of pathogenic bacteria cells were completely eliminated.
Among 31 bacterial strains isolated from cultivated soil of Vietnamese ginseng in Quang Nam province, the P6 was the highest potential of IAA biosynthesis strain. The analyses of morphological and biochemical characteristics as well as 16S rRNA gene sequencing identified that the P6 strain belongs to Kluyvera cryocrescens species (99.93% similarity). The results of studying on culture conditions of the strain K. cryocrescens showed that King's B medium with nitrogen source as peptone and KNO 3 (0.5% w/v), added 0.1% tryptophan (w/v) at 30 o C gave the highest IAA content (97.7 µg/mL) after 4 days of cultivation. The effect of crude IAA formed in the culture liquid of K.cryocrescens on germination rate of cucumber seeds and growth of cucumber trees indicated that: germination rate of the block treated with IAA liquid is 93.3% higher than that of the control sample of 80%. After 10 days of sowing, the trees of the experimental block had rapid and even growth, the length of their stem and roots, the weight of their stem and leaves as well as their root number all developed better than the control sample.
