Modulation of ionic channel properties and synaptic functions by neurotransmitters and hormones is called neuro-modulation and may be the basis for many long-lasting changes in animal behavior, for example, changes in the arousal or motivational states. We have previously shown in a teleost, the dwarf gourami, that the terminal nerve (TN) cells are a major component of the gonadotropin-releasing hormone (GnRH) system and are structurally independent from the preoptic/hypophysial-GnRH system, which projects to the pituitary and facilitates gonadotropin release from the gonadotropes. While GnRH fibers originating from TN cells are distributed widely throughout the brain, they do not project to the pituitary. Thus, TN-GnRH system does not function as a hypophysiotropic hormone but probably as a neuro-modulator, capable of affecting widespread regions of the brain. In the present study, we used a whole-brain in vitro preparation to examine the spontaneous electrical activities of TN-GnRH cells and to determine the morphology of individual cells by intracellular injections of either neurobiotin or biocytin. The recorded cells were clearly identified as GnRH-immunoreactive TN cells by using combined intracellular injection and GnRH immunocytochemistry. Most of the TN cells showed endogenous slow (1-7 Hz), regular beating discharges. The intrinsic nature of this activity was demonstrated by the voltage dependency of the beating frequency, rhythm resetting, and persistence of rhythmicity after synaptic isolation. Only a small number of TN cells showed either irregular or bursting discharge patterns. Anatomical observation of intracellularly labeled cells showed that, regardless of discharge patterns, all the TN cells had multiple axonal branches that project to those areas where we had previously demonstrated dense GnRH-immunoreactive fibers. From the present results, we propose a hypothesis that may be relevant to the peptidergic and monoaminergic neuromodulatory systems in general. The modulator neurons have endogenous rhythmic activities that vary according to the animal's hormonal or environmental conditions, and they regulate the excitability of target neurons in a wide variety of brain regions simultaneously via multiple axonal branches.

Filial imprinting in precocial birds is the process of forming a social attachment during a sensitive or critical period, restricted to the first few days after hatching. Imprinting is considered to be part of early learning to aid the survival of juveniles by securing maternal care. Here we show that the thyroid hormone 3,5,3′-triiodothyronine (T3) determines the start of the sensitive period. Imprinting training in chicks causes rapid inflow of T3, converted from circulating plasma thyroxine by Dio2, type 2 iodothyronine deiodinase, in brain vascular endothelial cells. The T3 thus initiates and extends the sensitive period to last more than 1 week via non-genomic mechanisms and primes subsequent learning. Even in non-imprinted chicks whose sensitive period has ended, exogenous T3 enables imprinting. Our findings indicate that T3 determines the start of the sensitive period for imprinting and has a critical role in later learning.

1. To elucidate the neural mechanisms responsible for coordinating undulatory locomotor movements, the intersegmental phase lag was analyzed from ventral roots along the spinal cord during fictive swimming. It was induced by bath application of N-methyl-D-aspartate (NMDA) in in vitro preparations of lamprey spinal cord, while the excitability of different segments were modified. The phase lag between consecutive segments during normal forward swimming is 1% of the cycle duration in a broad range of values. Rostral segments are activated before more caudal ones. 2. Under control conditions, whole preparations (12-24 segment long; n = 22) were perfused with NMDA solutions of the same concentration (100-150 microM). The intersegmental phase lag values varied in a continuous range with a single peak around a median value of forward +0.74% per segment (range: forward +2.23% to backward -0.97%). 3. To examine whether excitability differences along the spinal cord could modify the intersegmental phase lag, different levels of excitatory amino acids (NMDA) were applied to spinal cord preparations positioned in a partitioned chamber. Different portions of the cord could be perfused separately by NMDA solutions of different concentrations (50-150 microM). If rostral segments were perfused with the higher NMDA solution, the lag was inevitably in the forward direction. Conversely, if the caudal portion was perfused with the higher NMDA solution, caudally located ventral roots became activated before the rostral ventral roots in a caudorostral succession, thus reversing the direction of the fictive swimming wave to propagate as during backward swimming. If the middle portion was perfused by the highest NMDA solution, this portion instead became leading, and the activity propagated from this point in both the rostral and the caudal directions. The portion located in the pool with highest NMDA concentration always gave rise to a "leading" segment. 4. When a portion of the preparation was perfused with an NMDA solution of a high concentration (75-150 microM), the cycle duration was close to that recorded when the whole preparation was perfused with the same high NMDA solution. The ensemble cycle duration is, therefore, largely determined by the leading segment. 5. The phase lag changes were not restricted to the region around the barrier separating pools with different NMDA solutions.(ABSTRACT TRUNCATED AT 400 WORDS)

Effects of bilateral chemical lesions of the medial basal ganglia [lobus parolfactorius (LPO)] were examined in 7- to 14-d-old domestic chicks. Chicks were trained in a color discrimination task, in which the subject had to peck one of the two colored beads associated with rewards that differed in quantity (amount of food) and/or temporal proximity (delay of food delivery from peck). In experiment 1, food was given without delay, and chicks successfully learned to choose a colored bead that was associated with a larger reward than the other. In experiment 2, a colored bead (red) was associated with a large reward delivered after a delay (D = 1, 2, or 3 sec), whereas another (yellow) was associated with a small reward delivered immediately. In intact and sham-operated conditions, chicks with a longer D chose the red bead progressively fewer times. Selective lesions to the caudal LPO (but not the rostral LPO) caused impulsive choice, and the ablated chicks chose the yellow bead and gained a small-immediate reward regardless of D. However, when retrained in a null-delay condition (D = 0 sec), the lesioned chick chose the red bead again. Ability to associate novel colors with reward was also unimpaired. These results suggest that the LPO may be responsible for the anticipation of reward proximity and involved in a suppression of impulsiveness by which animals seek immediate gains. The present results also indicate a striking similarity in functional roles between the avian LPO and the nucleus accumbens/ventral striatum in mammals.

Activation of gamma-aminobutyric acid-B (GABAB) receptors during N-methyl-D-aspartate (NMDA)-induced fictive locomotor activity in the lamprey spinal cord reduces the burst frequency and changes the intersegmental coordination. Presynaptic inhibition of both the excitatory and inhibitory synaptic transmission from spinal premotor interneurons occurs through GABAB receptor activation. To further analyze the cellular mechanisms underlying the GABABergic modulation of the locomotor network, the present study investigates somatodendritic effects of GABAB receptor activation on interneurons and motoneurons in the lamprey spinal cord in vitro using single-electrode current- and voltage-clamp techniques. 2. High- (HVA) and low- (LVA) voltage-activated calcium currents were studied with single-electrode voltage clamp when Na+ and K+ currents were blocked--using tetrodotoxin, tetraethylammonium (TEA), and CsCl electrodes--after substituting Ca2+ with Ba2+. Cobalt-sensitive inward barium currents, activated at -50 mV, became larger when the holding potential was set to a more hyperpolarized level, thus suggesting the existence of an LVA calcium current. The presence of cobalt-sensitive inward barium currents activated at -30 and -10 mV suggests the existence of an HVA calcium current. GABAB receptor activation (baclofen) reduced the peak amplitude of both the LVA and HVA Ca2+ component. 3. The late phase of the afterhyperpolarization (AHP), which follows the action potential, was reduced in amplitude by cobalt, thus lending further support to the notion that the Ca2+ influx, and the subsequent activation of Ca(2+)-dependent K+ channels (KCa2+), constitutes the major part of the AHP generation. Application of the GABAB agonist baclofen also reduced the peak amplitude of the AHP in interneurons and motoneurons, and this reduction was counteracted by the GABAB antagonist 2(OH)saclofen. Baclofen reduced the duration of action potentials broadened by TEA, thus suggesting that the Ca2+ inflow was reduced. Intracellular injection of the GTP analogue GTP gamma S also reduced the duration of the action potential and the peak amplitude of the AHP in TEA, thus supporting the notion that a GTP-binding protein (G-protein)-mediated GABAB receptor activation reduced the calcium inflow, leading to less activation of KCa channels and, consequently, to a smaller peak amplitude of the AHP. 4. Baclofen suppressed the subthreshold depolarization induced by a depolarizing current pulse injection without affecting either the spike threshold or the resting membrane conductance.(ABSTRACT TRUNCATED AT 400 WORDS)
