Biocompatible and biodegradable porous scaffolds with adjustable pore structure have aroused increasing interest in bone tissue engineering. Here, we report a facile method to fabricate hierarchical macroporous biocompatible (HmPB) scaffolds by combining Pickering high internal phase emulsion (HIPE) templates with three-dimensional (3D) printing. HmPB scaffolds composed of a polymer matrix of poly(l-lactic acid), PLLA, and poly(ε-caprolactone), PCL, are readily fabricated by solvent evaporation of 3D printed Pickering HIPEs which are stabilized by hydrophobically modified silica nanoparticles (h-SiO). The pore structure of HmPB scaffolds is easily tailored to be similar to natural extracellular matrix (ECM) by varying the fabrication conditions of the Pickering emulsion or adjusting the printing parameters. In addition, in vivo drug release studies which employ enrofloxacin (ENR) as a model drug indicate the potential of HmPB scaffolds as a drug carrier. Furthermore, in vivo cell culture assays prove that HmPB scaffolds that possess good biocompatibility as mouse bone mesenchymal stem cells (mBMSCs) can adhere and proliferate well on them. All the results suggest that HmPB scaffolds hold great potential in bone tissue engineering applications.
Finding top-k elephant flows is a critical task in network traffic measurement, with many applications in congestion control, anomaly detection and traffic engineering. As the line rates keep increasing in today's networks, designing accurate and fast algorithms for online identification of elephant flows becomes more and more challenging. The prior algorithms are seriously limited in achieving accuracy under the constraints of heavy traffic and small on-chip memory in use. We observe that the basic strategies adopted by these algorithms either require significant space overhead to measure the sizes of all flows or incur significant inaccuracy when deciding which flows to keep track of. In this paper, we adopt a new strategy, called count-withexponential-decay, to achieve space-accuracy balance by actively removing small flows through decaying, while minimizing the impact on large flows, so as to achieve high precision in finding top-k elephant flows. Moreover, the proposed algorithm called HeavyKeeper incurs small, constant processing overhead per packet and thus supports high line rates. Experimental results show that HeavyKeeper algorithm achieves 99.99% precision with a small memory size, and reduces the error by around 3 orders of magnitude on average compared to the state-of-the-art.
In preclinical studies, neural stem cell (NSC)-based delivery of oncolytic virus has shown great promise in the treatment of malignant glioma. Ensuring the success of this therapy will require critical evaluation of the spatial distribution of virus after NSC transplantation. In this study, the patient derived GBM43 human glioma line was established in the brain of athymic nude mice, followed by administration NSCs loaded with conditionally replicating oncolytic adenovirus (NSC-CRAd-S-pk7). We determined the tumor coverage potential of oncolytic adenovirus by examining NSC distribution using magnetic resonance (MR) imaging and by three-dimensional reconstruction from ex vivo tissue specimens. We demonstrate that unmodified NSCs and NSC-CRAd-S-pk7 exhibit a similar distribution pattern with most prominent localization occurring at the tumor margins. We were further able to visualize the accumulation of these cells at tumor sites via T2-weighted MR imaging as well as the spread of viral particles using immunofluorescence. Our analyses reveal that a single administration of oncolytic virus-loaded NSCs allows for up to 31% coverage of intracranial tumors. Such results provide valuable insights into the therapeutic potential of this novel viral delivery platform.

The automated design of imaging systems involving no or minimal human effort has always been the expectation of scientists, researchers and optical engineers. In addition, it is challenging to choose an appropriate starting point for an optical system design. In this paper, we present a novel design framework based on a point-by-point design process that can automatically obtain high-performance freeform systems. This framework only requires a combination of planes as the input based on the configuration requirements or the prior knowledge of designers. This point-by-point design framework is different from the decades-long tradition of optimizing surface coefficients. Compared with the traditional design method, whereby the selection of the starting point and the optimization process are independent of each other and require extensive amount of human effort, there are no obvious differences between these two processes in our design framework, and the entire design process is mostly automated. This automated design process significantly reduces the amount of human effort required and does not rely on advanced design skills and experience. To demonstrate the feasibility of the proposed design framework, we successfully designed two high-performance systems as examples. This point-by-point design framework opens up new possibilities for automated optical design and can be used to develop automated optical design in the areas of remote sensing, telescopy, microscopy, spectroscopy, virtual reality and augmented reality.
Set queries are fundamental operations in computer systems and applications. This paper addresses the fundamental problem of designing a probabilistic data structure that can quickly process set queries using a small amount of memory. We propose a Shifting Bloom Filter (ShBF) framework for representing and querying sets. We demonstrate the effectiveness of ShBF using three types of popular set queries: membership, association, and multiplicity queries. The key novelty of ShBF is on encoding the auxiliary information of a set element in a location offset. In contrast, prior BF based set data structures allocate additional memory to store auxiliary information. To evaluate ShBF in comparison with prior art, we conducted experiments using real-world network traces. Results show that ShBF significantly advances the state-of-the-art on all three types of set queries.
In this paper, we proposed a general direct design method for three-dimensional freeform surfaces and freeform imaging systems based on a construction-iteration process. In the preliminary surfaces-construction process, the coordinates as well as the surface normals of the data points on the multiple freeform surfaces can be calculated directly considering the rays of multiple fields and different pupil coordinates. Then, an iterative process is employed to significantly improve the image quality or achieve a better mapping relationship of the light rays. Three iteration types which are normal iteration, negative feedback and successive approximation are given. The proposed construction-iteration method is applied in the design of an easy aligned, low F-number off-axis three-mirror system. The primary and tertiary mirrors can be fabricated on a single substrate and form a single element in the final system. The secondary mirror is simply a plane mirror. With this configuration, the alignment difficulty of a freeform system can be greatly reduced. After the preliminary surfaces-construction stage, the freeform surfaces in the optical system can be generated directly from an initial planar system. Then, with the iterative process, the average RMS spot diameter decreased by 75.4% compared with the system before iterations, and the maximum absolute distortion decreased by 94.2%. After further optimization with optical design software, good image quality which is closed to diffraction-limited is achieved.
In this paper, a novel method is proposed to design the freeform off-axis reflective imaging systems. A special algorithm is demonstrated to calculate the data points on the unknown freeform surface using the rays from multiple fields and different pupil coordinates. These points are used to construct the multiple three-dimensional freeform surfaces in an imaging system which works for a certain object size and a certain width of light beam. An unobscured design with freeform surfaces can be obtained directly with this method, and it can be taken as a good starting point for further optimization. The benefit of this design method is demonstrated by designing a freeform off-axis three-mirror imaging system with high performance and system specifications. The final system operates at F/1.49 with a 64mm entrance pupil diameter and an 8° × 9° field-of-view (FOV). The performance of the system is diffraction limited at LWIR (long-wavelength-infrared).
