Obtención y caracterización de un hidrolizado de colágeno purificado producido mediante el uso de la enzima delvolaseObtaining and characterizing a purified collagen hydrolyzize produced by the use of the enzyme delvolase Tito Eduardo Lleren Daza 1 * ; Wendy Patricia Rodríguez Torres 2 .*Autor de correspondencia Resumen El objetivo de la investigación fue obtener un hidrolizado proteico por vía enzimática, a partir de colágeno purificado de pescado, mediante el uso de la proteasa Delvolase. Además, se buscó caracterizar el hidrolizado de colágeno purificado de pescado, obtenido con los parámetros más eficientes, mediante la determinación de algunas de sus propiedades funcionales. La hidrólisis fue catalizada por una proteasa comercial (Delvolase®), se optimizó las condiciones de hidrólisis con el uso de la metodología de superficie de respuesta (MSR). El modelo fue propuesto con respecto a los efectos de la temperatura (T), tiempo (t) y enzima (E), en rangos de T: 55 -65 °C, t: 2 h -6 h, E: 0,03% -0,15% respectivamente, encontrándose un óptimo grado de hidrólisis (GH) de 12,59% para un tiempo de 2 horas, a una temperatura óptima de 64,97 °C. El hidrolizado de colágeno purificado optimizado, se caracterizó por presentar en base húmeda 12,55% de proteína, 0,32% de grasa y 1% de ceniza. Así mismo, se caracterizó por poseer una buena capacidad para gelificar, formar espuma, emulsificar y absorber aceite. La hidrólisis proteica del colágeno de pescado es una alternativa recomendable para la elaboración de nuevos productos biotecnológicos. Abstract The objective of the research was to obtain a protein hydrolyzate by enzymatic route, from purified fish collagen, with the use of the protease Delvolase. In addition, we sought to characterize the purified fish collagen hydrolyzate, obtained with the most efficient parameters, by determining some of its functional properties. The hydrolysis was catalyzed by a commercial protease (Delvolase®), hydrolytic conditions were optimized with the use of surface response methodology (MSR). The model was proposed in relation to effects of temperature (T), time (t) and enzyme (E), in ranges of T: 55 -65 °C, t: 2h -6h, E: 0,03% -0,15% respectively. The optimal hidrolysis degree (HD) was 12,59% for 2 hours in a slightly alkaline way. The fish collagen hidrolyzate obtained with optimal parameters showed in wet basis 12,55% of protein, 0,32% fat, 1% of ashes. Likewise, it was characterized by having a good ability to gel, foam, emulsify and absorb oil. Protein hydrolysis of fish collagen is an advisable alternative for the development of new biotechnological products.
Extracción y caracterización del aceite crudo obtenido de un hidrolizado enzimático de residuos frescos de anchoveta (Engraulis ringens)Extraction and characterization of crude oil recovered from anchovy (Engraulis ringens) solid wastes enzymatic hydrolysis 1 * Tito Eduardo Llerena Daza y 2 Diana Milagros Aranda Pariasca Resumen Los residuos frescos de la anchoveta (Engraulis ringens) provenientes del procesado para consumo humano directo fue la materia prima para obtener un hidrolizado enzimático que facilite la posterior extracción de aceite crudo. El estudio se realizó en dos etapas. La primera consistió en encontrar las condiciones más favorables para el proceso de hidrólisis enzimática aplicando la metodología de superficie de respuesta (MSR), para ello se seleccionaron cuatro variables independientes: temperatura, tiempo de hidrólisis dosificación de la enzima Protamex TM y proporción agua : materia prima, obteniéndose 30 combinaciones experimentales; y se evaluaron las variables dependientes: cantidad de aceite extraído y color, con ayuda del programa estadístico Statgraphics®. En la segunda etapa se determinó el porcentaje de aceite crudo extraído y sus características físicas y químicas: perfil de ácidos grasos, valor de TBA, acidez libre, materia insaponificable, valor de anisidina, valor peróxido, humedad y materia volátil, densidad, peso específico y color en L*a*b*. En la primera etapa, y de acuerdo a los modelos matemáticos hallados, se determinó que para obtener la mayor cantidad de aceite crudo, los niveles de temperatura, tiempo de hidrólisis, dosificación de la enzima y proporción de agua: materia prima necesario fueron de 54,15 ºC, 44,1 min, 3% del peso de proteína presente en la muestra (PPM) y 0,5: 1 respectivamente. En la segunda etapa el porcentaje de aceite crudo extraído fue 69,7± 1,7% del contenido graso total, en el perfil de ácidos grasos se obtuvo valores promedio de 28,6% de omega 3 (EPA + DHA) y se determinó la acidez libre (5,63% como ácido oleico), valor peróxido (12,7 meq O 2 /kg), materia insaponificable (2,3%), TBA (0,968 mg MA/kg), valor de anisidina (26,5), humedad (0,13%), densidad (0,927 g/ cm 3 ), peso específico (0,937) y en color los valores fueron L* (30,07); a*(11,76) y b*(20,75) respectivamente. Estas comparaciones se realizaron mediante pruebas de penetración de calor comparativas para cada producto con su líquido de gobierno respectivo teniendo como consideración la similitud de los parámetros restantes; fijando como la letalidad objetivo F 0 = 6 min para alimentos de baja acidez. Los métodos de tratamiento utilizados pueden ser determinantes en el contenido de los nutrientes de los productos consumidos. Objetivo: Determinar el efecto del tratamiento térmico sobre la composición proximal, color, perfil de ácidos grasos (EPA y DHA) e inocuidad en conservas de recortes de caballa tratadas con humo líquido, envasados en bolsas flexibles esterilizables para estimar su calidad nutricional. Método: En los recortes crudos, horneados y esterilizados se determinaron por triplicado parámetros de composición proximal, bases volátiles totales (N-BVT), medición de color, perfil de ácidos grasos y análisis microbiológicos en el periodo 2019-2020. Resultados: los niveles de proteína, grasa, ceniza y N-BVT se incrementaron significativamente en las etapas de horneado y esterilizado, el valor L* disminuyó, a* y b* se incrementaron significativamente. Se identificaron 37 ácidos grasos, en los recortes crudos destacaron el DHA y EPA, palmítico y oleico con 0.47, 1.36, 0.49 y 0.28 g/100 g, respectivamente. Todos los ácidos grasos en general se incrementaron por el tratamiento térmico, sobresaliendo el palmítico, linoleico y oleico con 1.28, 1.23 y 1.04 g/100g, respectivamente. Los análisis microbiológicos evidenciaron el cumplimiento con los requisitos de inocuidad. El valor Fo fue de 6.3 min y el proceso programado de 38 min a 120 °C. Las proporciones n-6/n-3 y AGPI/AGS fueron de 0.13, 0.65 y 0.76; 2.45, 1.79 y 1.69, respectivamente en recortes crudos, horneados y esterilizados Conclusiones: El producto desarrollado a partir de recortes de caballa envasado en bolsas flexibles esterilizables, tuvo buenos atributos y características nutricionales beneficiosos para la dieta humana.
