Trypanosomiasis caused by Trypanosoma vivax has increased the reports in Brazil in the last decade. An outbreak is herein first reported in the state of Goiás, from May 2016 to January 2017. The outbreak start occurred in the city of Ipameri (Goiás) after the introduction of 18 auctioned cows from the state of Minas Gerais. Direct parasitological test (blood smears) and polymerase chain reactions targeting the catL genes diagnosed T. vivax infection. Fifty six cows from a herd of 161 were infected; 12 died during the outbreak and 44 animals persistently positive (by blood smears) even after chemical treatment were discarded. After this first case, five other cases were detected in state of Goiás. The spread of this disease can be linked to the commercialization of animals carrying T. vivax, allied to the iatrogenic transmission practice, using a single needle and syringe for all cows, during oxytocin administration before each milking.Keywords: Bovines, lactating cows, oxytocin, PCR, Trypanosomiasis, T. vivax. ResumoTripanossomíase causada por Trypanosoma vivax tem sido reportada em várias regiões do Brasil na última década. Um surto é aqui reportado no Estado de Goiás, de maio de 2016 a janeiro de 2017. O início do referido surto ocorreu na cidade de Ipameri (Goiás) depois da introdução de 18 vacas Girolando, provenientes do Estado de Minas Gerais. Pelo exame parasitológico direto (esfregaço sanguíneo) e pela reação em cadeia pela polimerase, identificou-se a infecção por T. vivax nos animais. Cinquenta e uma vacas de um rebanho de 161 foram infectadas; 12 morreram durante o surto e 44 animais permaneceram positivos (pelo esfregaço sanguíneo) mesmo recebendo tratamento químico (diminazene), e foram descartadas. Após esse primeiro caso, foram detectados outros cinco casos no Estado de Goiás. A disseminação dessa doença pode estar ligada à comercialização de animais portadores de T. vivax, aliada à prática de transmissão iatrogênica, utilizando-se uma única agulha e seringa para, todas as vacas, durante a administração da oxitocina antes de cada ordenha.Palavras-chave: Bovinos, vacas em lactação, oxitocina, PCR, Tripanossomíase, T. vivax.
The present study attempted to verify the prevalence of and risk factors for diarrhea-causing agents in dairy calves from Brazil. Additionally, ages with a higher risk of occurrence for each agent were verified by means of the receiver operating characteristic (ROC) curve. The collections were performed on 39 farms, belonging to 29 municipalities located in eight states of Brazil. It was possible to conclude that the prevalence of Coronavirus, Rotavirus, Cryptosporidium spp., Eimeria spp., and nematodes was 7.20% (95% CI 4.54-9.78), 6.37% (95% CI 3.85-8.89), 51.52% (95% CI 45.26-55.57), 3.46% (95% CI 2.24-4.67), and 3.46% (95% CI 2.24-4.67), respectively. Ages with higher probabilities of occurrence of these diseases in calves were < 10, > 8, > 6, > 37, and > 36 days, respectively. Diarrhea occurred more significantly (P < 0.0001) in animals less than 21 days old and mainly on those receiving milk through automatic feeders (P < 0.001). Cryptosporidium spp. were a risk factor for the occurrence of Rotavirus, and vice versa (P = 0.0039) and presented a positive correlation with Coronavirus (P = 0.0089). Calves that drink water from rivers, streams, and ponds had a higher chance of being infected by Eimeria spp. (P < 0.0001), as well as developing infection by nematodes (P < 0.0001). The results found in this study highlight the importance of studying the agents of diarrhea together, once they act as coinfection where the losses triggered for the owners will involve some of these agents simultaneously.
Voriconazole (VCZ), a triazole with a large spectrum of action is one of the most recommended antifungal agents as the first line therapy against several clinically important systemic fungal infections, including those by Candida albicans. This antifungal has moderate water solubility and exhibits a nonlinear pharmacokinetic (PK) profile. By entrapping VCZ into liposomes, it is possible to circumvent certain downsides of the currently available product such as a reduction in the rate of its metabolization into an inactive form, avoidance of the toxicity of the sulfobutyl ether-beta-cyclodextrin (SBECD), vehicle used to increase its solubility. PKs and biodistribution of VCZ modified by encapsulation into liposomes resulted in improved antifungal activity, due to increased specificity and tissue penetration. In this work, liposomal VCZ resulted in AUC0–24/MIC ratio of 53.51 ± 11.12, whereas VFEND® resulted in a 2.5-fold lower AUC0–24/MIC ratio (21.51 ± 2.88), indicating favorable antimicrobial systemic activity. VCZ accumulation in the liver and kidneys was significantly higher when the liposomal form was used. Protection of the drug from biological degradation and reduced rate of metabolism leads to a 30% reduction of AUC of the inactive metabolite voriconazole-N-oxide (VNO) when the liposomal drug was administered. Liposomal VCZ presents an alternative therapeutic platform, leading to a safe and effective treatment against systemic fungal infections.
Bovine trypanosomosis has been spreading in Brazil. In the present study, we evaluated the spatial distribution, prevalence and risk factors of this disease in the state of Goiás, Brazil, and performed both molecular and phylogenetical analyses of Trypanosoma vivax. A total of 4049 blood samples were collected from cattle for a period of 2 years. The parasitological diagnosis was performed using the Woo method and a questionnaire was administered to the farmers to document risk factors associated with the disease in the herd. Positive samples were DNA sequenced and compared to GenBank codes. The prevalence of T. vivax was 8.84%, occurring on 24 ranches only in dairy cattle and mainly in the central and southern portions of the state. The acquisition of new animals infected with T. vivax and the administration of exogenous oxytocin to cows using the same syringe and needle were the main associated factors (P ≤ 0.05). After an outbreak, milk production decreased by 39.62%. The presence of biting flies (tabanids, Haematobia irritans and Stomoxys calcitrans) was not a risk factor (P > 0.05) for the occurrence of T. vivax. The epidemiological data demonstrate the importance of restricting the practice of auctions as well as eliminating the use of exogenous oxytocin in animals during milking. The samples tested by polymerase chain reaction were positive for T. vivax and were genetically homologous with T. vivax found in different states of Brazil and west Africa based on the 18S rRNA gene.
The present study aimed to determine the susceptibility of 32 R. (B.) microplus populations from Southeast, Midwest and South regions of Brazil, to fluazuron (2.5mg/kg), administered topically (pour-on). Additionally, five populations (Southeast and Midwest regions) of the southern cattle tick were evaluated using in vivo field studies, regarding their susceptibility to a new combination of novaluron (2.0mg/kg)+eprinomectin (0.36mg/kg), administered subcutaneously, compared with two positive controls (fluazuron 2.5mg/kg and eprinomectin 0.5mg/kg), both administered topically (pour-on). Selected bovines were allocated to treatment groups on day 0, and block formation was based on arithmetic means of female ticks (4.5-8.0mm long) counted on three consecutive days (-3, -2 and -1). To evaluate therapeutic and residual efficacies of these formulations, tick counts (females ranging from 4.5 to 8.0mm long) were performed on days 3, 7 and 14 post-treatment, continuing on a weekly basis until the end of each experiment. Results obtained throughout this study, utilizing field efficacy trials, allowed us to conclude that four R. (B.) microplus populations (including two in the Southeast and two in the Midwest regions) could be diagnosed as resistant, or with low susceptibility, to fluazuron (2.5mg/kg). Such fact was detected in farms where owners applied products containing this active component on cattle for at least five years, with treatment intervals of 30-55days during the rainy season. Nonetheless, in vitro studies should be performed in order to reinforce in vivo results obtained on the present study. Regarding efficacy indexes obtained by the association of eprinomectin and the novel molecule novaluron against R. (B.) microplus, none of the trials managed to obtain efficacies superior to 48%. Such results, allied to data obtained by different researchers and previously published in literature, reinforce the perception that maybe these formulations containing novaluron, in the administered dosages and treatment routes, may not be effective tools for controlling R. (B.) microplus. However, future studies must be conducted in order to support such hypothesis. Additionally, all five R. (B.) microplus populations were diagnosed as resistant, or with low susceptibility, to eprinomectin (0.5mg/kg) as well. Even though fluazuron, administered topically (pour on), is still an excellent active principle to be used against R. (B.) microplus, resistance management strategies should be quickly implemented in order to keep selection pressure in Brazil at a minimum level for this compound.
This study aimed to identify the frequency of occurrence of Eimeria species in Holstein (taurine dairy cattle) and Nelore (zebu beef cattle) on 12 farms in the state of Goiás, Brazil. A total of 2,601 animals were evaluated, and the cattle were divided into the following age-group categories: up to three months old, four to eight months old, nine to 16 months old and cows. Fecal samples were collected directly from the rectum of all animals. None of the cattle presented any clinical signs of eimeriosis during the survey. From the results obtained, it could be concluded that higher rates of infection by Eimeria spp. were observed in Holstein cattle between the 1st and 16 th months of age and in Nelore cattle between the 1st and 8th months of age. Twelve and nine Eimeria species were diagnosed in Holstein and Nelore cattle, respectively. However, Eimeria zuernii was more prevalent in animals up to three months old, while Eimeria bovis was more common in the remaining categories (cattle aged four months and over). Future studies on the epidemiology and/or seasonality of infection by Eimeria species in cattle, from birth onwards, are needed in order to better understand the dynamics of E. zuernii and E. bovis occurrences, especially in cattle under three months of age.
Conventional bacteriology techniques and quantitative polymerasechain reaction (qPCR) were applied to the eggshell, albumen, and yolk of washed and unwashed commercial white and brown eggs, to detect Salmonella spp. Pooled samples of eggshells, albumen, and yolk of white and brown eggs were collected at the poultry house and at the egg-storage room. Salmonella spp. was detected by conventional bacteriology in 5.4% (21/387) of analyzed samples and in 16% (68/387) by qPCR. In the 114 unwashed white eggs samples of eggshell, albumen and yolk, the bacterium was identified in 2.6% of the eggs (3/114) by conventional bacteriology and in 13.2% (15/114) by qPCR. In the 90 samples of washed eggs, 6.7% (6/90) were contaminated as detected by conventional bacteriology and 10.0% (9/90) by qPCR. In the 81 samples of unwashed brown eggs, Salmonella spp. was detected in 6.1% of the eggs (5/81) by conventional bacteriology and 27.2% (22/81) by qPCR. In the 102 samples of brown washed eggs, 6.9% (7/102) where positive by conventional bacteriology and 35.3% (16/102) by qPCR. All samples detected as positive by conventional bacteriology were also positive by qPCR. Salmonella Agona represented 18.2% (4/22) of identified serovars, Salmonella enterica subs. enterica O: 4.5 18.2%
RESUMO: O presente trabalho tem como objetivo relatar surtos consecutivos (Agosto de 2015 e 2016) ocasionado por Eimeria zuernii em bezerros de corte, Canchim e Nelore, com aproximadamente 45 dias de vida, em uma propriedade rural localizada no município de São João da Boa Vista, estado de São Paulo. O primeiro surto foi diagnosticado em um lote composto por 78 bezerros, entre 35 e 55 dias de idade. Dos 78 bezerros, 32 eram Nelore e os outros 46 Canchim. Já o segundo, ocorreu em um novo lote de 66 bezerros (sendo 36 Nelore e 47 Canchim), com idade entre 30 e 45 dias. Alguns animais apresentaram diarreia sanguinolenta e desidratação. Colheitas de fezes de todos os animais (vacas e bezerros) foram realizadas para realização de exame coproparasitológico e quantificação de ovos e oocistos por grama (OPG e OoPG) de fezes de parasitos. Com base nos resultados encontrados neste estudo, é possível afirmar que os surtos consecutivos em bezerros de corte entre 30 e 55 dias de idade, na época seca do ano (agosto), foram desencadeados pelo elevado grau de infecção destes animais por E. zuernii. Analisando os dois anos em que os surtos ocorreram, a presença de sinais clínicos (diarreia sanguinolenta) e a mortalidade de bezerros desencadeada pela eimeriose/desidratação, aconteceram em 8,3% 1,4% do rebanho, respectivamente. Estes resultados destacam a importância em se realizar pesquisas de campo, com o objetivo de se melhor entender a dinâmica de parasitismo de E. zuernii e E. bovis em bezerros com idade inferior a três meses, em função dos motivos já discutidos neste estudo. Além disso, é necessário que se investigue as possíveis fontes de infecção, como a água ingerida pelos animais, para que condutas práticas possam ser adotadas/recomendadas aos proprietários, diminuindo desta maneira os prejuízos ocasionados por surtos de Eimeria spp em rebanhos bovinos.
